    Abstract Carried in the nasal passages by up to 30% of humans, Staphylococcus aureus is recognized to be a successful opportunistic pathogen. It is a frequent cause of infections of the upper respiratory tract, including sinusitis, and of the skin, typically abscesses, as well as of food poisoning and medical device contamination. The antimicrobial resistance of such, often chronic, health conditions is underpinned by the unique structure of bacterial biofilm, which is the focus of increasing research to try to overcome this serious public health challenge. Due to the protective barrier of an exopolysaccharide matrix, bacteria that are embedded within biofilm are highly resistant both to an infected individual’s immune response and to any treating antibiotics. An in-depth appraisal of the stepwise progression of biofilm formation by S. aureus , used as a model infection for all cases of bacterial antibiotic resistance, has enhanced understanding of this complicated microscopic structure and served to highlight possible intervention targets for both patient cure and community infection control. While antibiotic therapy offers a practical means of treatment and prevention, the most favorable results are achieved in combination with other methods. This review provides an overview of S. aureus biofilm development, outlines the current range of anti-biofilm agents that are used against each stage and summarizes their relative merits.

    Abstract Clinical infection with malaria, caused by parasites of the genus Plasmodium, is considered a serious medical condition with the potential to become a life-threatening emergency. This is especially relevant to low-income countries in tropical and subtropical regions of the world where high rates of malaria-related morbidity and mortality are recorded. As a means to combat this major global public health threat, rapid and effective diagnosis remains the frontline action to initiate a timely and appropriate medical intervention. From all the approaches to parasite detection, rapid diagnostic tests, so-called RDTs, are the easiest to use and most cost-effective. However, some of the limitations inherent in this methodology could hinder effective patient treatment. A primary drawback is that the vast majority of commercially available RDTs detect only one of the five species of human malaria, P. falciparum. While this is the main cause of infection in many areas, it excludes the possibility of infection with another parasite (P. vivax, P. ovale, P. malariae, and P. knowlesi) or of mixed infections containing different species. Hence, a diagnosis of non-P. falciparum malaria is missed. In turn, in resource-constrained settings where optimal microscopy is not available, a misdiagnosis of bacterial infection based on signs and symptoms alone often results in an inappropriate prescription of antibiotics. Here, we discuss how effective diagnosis of malaria and indiscriminate use of antibiotics in sub-Saharan Africa, a hot spot for P. falciparum transmission, may both be addressed by the development of innovative multiplexing RDTs that detect two or more species of Plasmodium.

    Abstract Abstract Background Dengue virus, an Aedes mosquito-borne flavivirus, is associated with close to 400 million reported infections per annum worldwide. Reduction of dengue virus transmission depends entirely on limiting Aedes breeding or preventing adult female contact with humans. Currently, the World Health Organization promotes the strategic approach of integrated vector management in order to optimise resources for mosquito control. Main text Neglected tropical disease researchers focus on geographical zones where the incidence of clinical cases, and prevalence of vectors, are high. In combatting those infectious diseases such as dengue that affect mainly low-income populations in developing regions, a mosquito-centric approach is frequently adopted. This prioritises environmental factors that facilitate or impede the lifecycle progression of the vector. Climatic variables (such as rainfall and wind speed) that impact the vector’s lifecycle either causally or by happenstance also affect the human host’s ‘lifecycle’, but in very different ways. The socioeconomic impacts of the same variables that influence vector control impact host vulnerability but at different points in the human lifecycle to those of the vector. Here, we argue that the vulnerability of the vector and that of the host interact in complex and unpredictable ways that are characteristic of (complex and intransigent) ‘wicked problems’. Moreover, they are treated by public health programs in ways that may ignore this complexity. This opinion draws on recent evidence showing that the best climate predictors of the scale of dengue outbreaks in Bangladesh cannot be explained through a simple vector-to-host causal model. Conclusions In mapping causal pathways for vector-borne diseases this article makes a case to elevate the lifecycle of the human host to a level closer in equivalence to that of the vector.     Abstract A characteristic of bacterial zoonoses, diseases caused by bacteria that can be transmitted to humans from animals, is a propensity to re-emerge. Several studies demonstrate their ongoing transmission in Nigeria, the most populous country in Africa. However, as local epidemiological data on bacterial zoonoses are inadequate the extent and impact of these infectious diseases is under-reported. Consequently, they are not a targeted priority of national public health policies. This limited recognition is despite indications of their possible roles in the widespread prevalence of non-malarial undifferentiated fever in Nigeria. While a number of animal reservoirs and arthropod vectors have been identified in the transmission routes of these diseases, an escalation of cases of undiagnosed febrile illness highlights the urgent need for a comprehensive assessment of other potential reservoirs, vectors and transmission cycles that may increase the local risk of infection with bacterial zoonoses. Animal health interventions have been proposed as a cost-effective strategy. Here, we present a broad overview of bacterial zoonotic infections of humans in Nigeria in the context of evolving epidemiological patterns. Further, we propose that facilitating the operation of a community-based One Health program is essential to providing the comprehensive epidemiological information that is required to improve prioritization of bacterial zoonoses. This would provide a driver for much needed investment in relevant public health interventions in Africa's most populous country.

    Abstract Schistosomiasis is one of the leading neglected tropical diseases in sub-Saharan Africa. Recorded case numbers of this chronic and debilitating helminth disease indicate Nigeria to be the most endemic country within this region. National control efforts have focused intensively on restricting human contact with freshwater sources of intermediate host snails. However, limited attention has been paid to the role of livestock as reservoir hosts and the prevalence of transmission of schistosomes to humans via farmed animals. The West African nations of Mali, Senegal, and the neighbouring Niger, Benin, and Cameroon have all reported the hybridization of the closely related species of Schistosoma haematobium , which infects humans, and S. bovis , which infects cattle. As these countries share the Niger and Benue rivers, with their tributaries, there is a distinct possibility of aquatic snails infected with hybrid schistosomes migrating to become established in the Nigerian river system. Here, we report on the current state of research in Nigeria that aims to elucidate key aspects of zoonotic schistosomiasis epidemiology. Factors promoting the hybridization of Schistosoma species are highlighted, and how available control measures can be optimized to address the emergence of schistosome hybrids is discussed.

    Abstract A central characterization of retinal immunobiology is the prevention of proinflammatory activity by macrophages. The retinal pigment epithelial cells (RPEs) are a major source of soluble anti-inflammatory factors. This includes a soluble factor that induces macrophage apoptosis when the activity of the immunomodulating neuropeptide alpha-melanocyte-stimulating hormone (α-MSH) is neutralized. In this manuscript, isolated extracellular soluble membranes (ESMs) from primary RPE were assayed to see if they could be the soluble mediator of apoptosis. Our results demonstrated that RPE ESMs mediated the induction of macrophage apoptosis that was suppressed by α-MSH. In contrast, the RPE line ARPE-19, cultured under conditions that induce similar anti-inflammatory activity to primary RPEs, did not activate apoptosis in the macrophages. Moreover, only the ESMs from primary RPE cultures, and not those from the ARPE-19 cell cultures, expressed mFasL. The results demonstrate that RPE ESMs are a soluble mediator of apoptosis and that this may be a mechanism by which the RPEs select for the survival of α-MSH-induced suppressor cells.
