    Abstract Background: Psoriasis, a chronic inflammatory skin disease, is increasingly effectively managed with the targeted immunotherapy; however, long-term immunotherapy carries health risks, and loss of response. Therefore, we need to develop the alternative treatment strategies. Mesenchymal stem/stromal cell (M.S.C.) exosomes stand out for their remarkable immunomodulatory properties, gaining widespread recognition. This study investigated whether M.S.C. exosomes can reduce psoriasis-induced hyperplasia by inducing Transforming Growth Factor beta 2 (TGF-beta2) signaling.
    Methodology: Exosomes were isolated from M.S.C.s by ultracentrifugation. Then, scanning electron microscopy was used for the morphology of exosomes. To ascertain the exosome concentration, the Bradford test was used. To ascertain the cellular toxicity of exosomes in Human Umbilical Vein Endothelial Cells ( H.U.V.E.C), an MTT experiment was then conducted. Real-time PCR was used to quantify TGF beta2 expression levels, whereas an ELISA immunosorbent assay was used to determine the protein concentration of TGF beta2.

    Abstract Perceptual decisions rely on accumulating sensory evidence over time. However, the accumulation process is complicated in real life when evidence resulted from separated cues over time. Previous studies demonstrate that participants are able to integrate information from two separated cues to improve their performance invariant to an interval between the cues. However, there is no neural model that can account for accuracy and confidence in decisions when there is a time interval in evidence. We used behavioral and EEG datasets from a visual choice task -Random dot motion- with separated evidence to investigate three candid distributed neural networks. We showed that decisions based on evidence accumulation by separated cues over time are best explained by the interplay of recurrent cortical dynamics of centro-parietal and frontal brain areas while an uncertainty-monitoring module included in the model.

    Abstract Approximately one-third of clinical trials fail to meet their recruitment goals, which can cause costly delays to sponsors and compromise the scientific integrity and generalizability of a trial. Inadequate recruitment and retention of patient groups who have the disease under investigation may produce insufficient medical knowledge about the therapeutic effects of drugs or products for the population at large. It is essential to address these issues to ensure that certain groups are not unduly subjected to disproportionate risks or denied the benefits of research. This commentary will present opportunities for clinical trialists to use emerging technologies and decentralized approaches to improve clinical trial recruitment, mitigate disparities, and improve individual and population-level outcomes within cardiovascular medicine.

    Abstract Nowadays the nano-drug delivery system is an appropriate procedure for solving the problem of controlling drug performance in a traditional method. For this purpose, a new nano-adsorbent of modification multi-walled carbon nanotubes with cyanuric chloride and para-amino acetanilide was synthesized for the effective sorption and desorption of lansoprazole drugs. Physicochemical properties of the multi-walled carbon nanotubes with cyanuric chloride and para-amino acetanilide were then specified with energy dispersive spectroscopy, scanning electron microscope, infrared spectroscopy (IR), thermal gravimetric (TGA), and elemental analysis (CHN). The adsorption process followed by the drug release kinetics and isotherm models has indicated a good and acceptable kinetic adsorption and Langmuir isotherm model in conditions simulated.     Abstract Resistance against glucocorticoids which are used to reduce inflammation and treatment of a number of diseases, including leukemia, is known as the first stage of treatment failure in acute lymphoblastic leukemia. Since these drugs are the essential components of chemotherapy regimens for ALL and play an important role in stop of cell growth and induction of apoptosis, it is important to identify genes and the molecular mechanism that may affect glucocorticoid resistance. In this study, we used the GSE66705 dataset and weighted gene co-expression network analysis (WGCNA) to identify modules that correlated more strongly with prednisolone resistance in type B lymphoblastic leukemia patients. The PPI network was built using the DEGs key modules and the STRING database. Finally, we used the overlapping data to identify hub genes. out of a total of 12 identified modules by WGCNA, the blue module was find to have the most statistically significant correlation with prednisolone resistance and Nine genes including SOD1, CD82, FLT3, GART, HPRT1, ITSN1, TIAM1, MRPS6, MYC were recognized as hub genes Whose expression changes can be associated with prednisolone resistance. Enrichment analysis based on the MsigDB repository showed that the altered expressed genes of the blue module were mainly enriched in IL2_STAT5, KRAS, MTORC1, and IL6-JAK-STAT3 pathways, and their expression changes can be related to cell proliferation and survival. The analysis performed by the WGCNA method introduced new genes. The role of some of these genes was previously reported in the resistance to chemotherapy in other diseases. This can be used as clues to detect treatment-resistant (drug-resistant) cases in the early stages of diseases.
