    Abstract Intergenerational relationships have become increasingly crucial for maintaining well-being in aging families. Under a changing sociocultural background, families in Hong Kong increasingly exhibit diverse intergenerational relationships and functioning. Focusing on families with mutiple children, this study investigated how the characteristics of parents and their adult children jointly affect different domains of intergenerational relationship quality. A two-stage stratified random sampling design was adopted. Face-to-face questionnaire interviews were conducted between November 2016 and March 2017 with 1,001 Hong Kong residents aged ≥ 50 years. Data of 612 parents and 1,745 adult children were included for analysis. Hierarchical linear modeling was performed to examine child- and parent-level correlates of intergenerational relationship quality. Parents who were female, were married, had higher self-perceived economic status, owned a house, and had fewer depressive symptoms, exhibited higher intergenerational relationship quality. Parents' age was positively related to affectual closeness, whereas their educational level was negatively related to both affectual closeness and conflict. More favorable intergenerational relationships were reported by aging parents whose adult children were younger, female, and married. Children with higher educational levels exhibited higher levels of both affectual closeness and conflict with their parents. Moreover, affectual closeness was found to be transmitted between generations. The findings can help improve awareness of the factors affecting the different domains of intergenerational relationships, thus informing the development of targeted services and interventions to promote family relationships and well-being.

    Abstract The plant growth-promoting rhizobacterium Tsukamurella tyrosinosolvens P9 can improve peanut growth. In this study, a co-culture system of strain P9 and peanut was established to analyze the transcriptome of peanut roots interacting with P9 for 24 and 72 h. During the early stage of co-culturing, genes related to mitogen-activated protein kinase (MAPK) and Ca2+ signal transduction, ethylene synthesis, and cell wall pectin degradation were induced, and the up-regulation of phenylpropanoid derivative, flavonoid, and isoflavone synthesis enhanced the defense response of peanut. The enhanced expression of genes associated with photosynthesis and carbon fixation, circadian rhythm regulation, indoleacetic acid (IAA) synthesis, and cytokinin decomposition promoted root growth and development. At the late stage of co-culturing, ethylene synthesis was reduced, whereas Ca2+ signal transduction, isoquinoline alkaloid synthesis, and ascorbate and aldarate metabolism were up-regulated, thereby maintaining root ROS homeostasis. Sugar decomposition and oxidative phosphorylation and nitrogen and fatty acid metabolism were induced, and peanut growth was significantly promoted. Finally, the gene expression of seedlings inoculated with strain P9 exhibited temporal differences. The results of our study, which explored transcriptional alterations of peanut root during interacting with P9, provide a basis for elucidating the growth-promoting mechanism of this bacterial strain in peanut.

    Abstract Background: Uveal melanoma (UVM) is an aggressive malignant tumor originating from melanocytes in the eye. Here, we screened the possible genes involved in the development and prognosis of UVM, and identified that FOXM1 and MET were associated with the prognosis of UVM patients. Forkhead box protein M1 (FOXM1) is a transcription factor that regulates the expression of cell cycle-related genes that are necessary for DNA duplication. However, the regulatory mechanism of FOXM1 in UVM was still not clear. Here, we investigated the regulation of FOXM1 in the malignant phenotype of UVM cells and its effect on the prognosis of UVM patients.
    Methods: UVM gene expression profiles were obtained using GSE22138 data from the gene expression omnibus (GEO). Weighted gene co-expression network analysis (WGCNA) was used to construct a key module gene for metastasis, which was strongly correlated with UVM prognosis. The latent biological pathways were identified through gene ontology analysis. Protein-protein interaction (PPI) networks and hub shared gene authentication were performed. GEPIA and UALCAN databases were used for the analysis of relationship between candidate genes (FOXM1 or MET) and the prognosis of UVM patients. The abundance of FOXM1 was examined by quantitative real time polymerase chain reaction (qRT-PCR) and western blot. Colony formation and cell counting kit-8 (CCK-8) assays for cell proliferation, wound healing assay for migration, and transwell invasion analysis for invasion were performed.
    Results: GEO database showed the differentially expressed genes between UVM samples with or without metastasis, and a key module gene for metastasis was constructed by WGCNA. The PPI network revealed that seven candidate genes (VEGFA, KRAS, MET, SRC, EZR, FOXM1, and CCNB1) were closely associated with UVM metastasis. GEPIA and UALCAN analyzes suggested that FOXM1 and MET are related to the prognosis of patients with UVM. These experimental results suggested that FOXM1 was highly expressed in UVM cells. FOXM1 deficiency represses the proliferative, migratory, and invasive abilities of UVM cells.
    Conclusions: FOXM1 silencing may hinder UVM cell progression, providing a novel theoretical basis and new insights for UVM treatment.

    Abstract Objective: This study aims to establish a screening method for canine distemper virus (CDV) microneutralizing activity suitable for microvolume samples.
    Methods: This method is based on the Indirect immunofluorescence assay (IFA) established on Vero-slam cells. First, by comparing the sensitivities of CDV neutralizing monoclonal antibody (1C42H11) and NP protein monoclonal antibody (CDV-NP) in IFA experiments, CDV-NP was selected as the primary antibody. Then, by detecting the infection rates of multi-concentrations of CDV neutralized with water, the minimum CDV concentration with an infection rate greater than 90% was defined as the minimum stable infection concentration, which was used as the neutralizing solution for this method. Finally, the CDV-positive neutralizing serum (neutralizing titer 1:708) was diluted into multiple dilution groups as test samples, and then neutralized in equal volumes with the neutralizing solution to detect the neutralizing activity detection rates of each dilution group and the lowest detection limit of this method.

    Conclusion: This study established an economical, stable, and easy-to-operate CDV microneutralizing activity high-throughput screening method, laying a methodological foundation for the development of native CDV neutralizing antibodies based on single B cells.

    Abstract With the rapid development of intelligent and connected vehicles, the experimental road test for the advanced driving assistance system (ADAS) is dramatically increasing around the world. Considering its high cost and hazardous situations, simulation test based on a dummy model is becoming a promising way for ADAS road test practice to reduce the experiment expanses. This study proposed a methodology for the evaluation of the performance of human and dummies with distinct designed materials based on the data extracted from the Doppler effect of millimeter-wave radar. Echo data of 8 different angles from 0 to 360 degrees, with the an interval of 45 degrees, at the same distance between the test object and the signal source is collected. Meanwhile, the echo energy is collected for correlation modeling and analysis among groups. By evaluating the performance of humans and dummies via statistical analysis, a close correlation was found which results verified the substitutability of the dummy for the ADAS experiment test. The correlation coefficient between human and dummies ranges from 0.75 to 0.93. The support vector machine (SVM) model was developed and fitted to predict the echo energy in diverse environments. The mean average error (MAE) is 5.42-11.42 in the training and testing datasets while root mean square error (RMSE) is 0.43-0.90. The methods developed in the study can simulate the real ADAS road test environment and support future experimental research.

    Abstract The aging of microplastics (MPs) is extremely influenced by photochemically-produced reactive intermediates (PPRIs), which are mediated by natural photosensitive substances. Algal extracellular polymeric substances (EPS) can produce PPRIs when exposed to sunlight. Nonetheless, the specific role of EPS in the aging process of MPs remains unclear. This work systematically explored the aging process of polystyrene (PS) MPs in the EPS secreted by Chlorella vulgaris under simulated sunlight irradiation. The results revealed that the existence of EPS accelerated the degradation of PS MPs into particles with sizes less than 1 µm, while also facilitating the formation of hydroxy groups on the surface.     Abstract As a burgeoning category of heterogeneous catalysts, atomic catalysts have been extensively researched in the field of electrocatalysis. To satisfy different electrocatalytic reactions, single-atom catalysts (SACs), diatomic catalysts (DACs) and triatomic catalysts (TACs) have been successfully designed and synthesized, in which microenvironment structure regulation is the core to achieve high-efficiency catalytic activity and selectivity. In this review, the effect of the geometric and electronic structure of metal active centers on catalytic performance is systematically introduced, including substrates, central metal atoms, and the coordination environment. Then theoretical understanding of atomic catalysts for electrocatalysis is innovatively discussed, including synergistic effects, defect coupled spin state change and crystal field distortion spin state change. In addition, we propose the challenges to optimize atomic catalysts for electrocatalysis applications, including controlled synthesis, increasing the density of active sites, enhancing intrinsic activity, and improving the stability.     Abstract Gastric carcinoma, being one of the most prevalent types of solid tumors, has emerged as the third leading cause of death worldwide. The symptoms of gastric cancer (GC) are typically complex, which makes early detection challenging. Immune checkpoint inhibition has become the new standard targeted therapy for advanced or metastatic GC. It is currently being explored in various combinations, both with and without chemotherapy, across multiple therapies in clinical trials. Immunotherapy can stimulate immune responses in GC patients, leading to the destruction of cancer cells. Compared with traditional therapies, immunotherapy has shown strong effectiveness with tolerable toxicity levels. Hence, this innovative approach to the treatment of advanced GC has gained popularity. In this review, we have outlined the recent advancements in immunotherapy for advanced GC, including immune checkpoint inhibitors, cancer vaccines, vascular endothelial growth factor-A inhibitors, and chimeric antigen receptor T-cell therapy. Our current emphasis is on examining the immunotherapies presently employed in clinical settings, addressing the existing challenges associated with these therapeutic approaches, and exploring promising strategies to overcome their limitations.

    Abstract This study examined ageing parents' care expectations across multiple care domains (financial and material, emotional, personal and informational) towards filial and formal sources and identified intergenerational correlates of care expectation patterns using a proposed care expectation model. Data of 780 eligible ageing parents were drawn from a representative household survey of ageing adults (≥50 years) conducted in 2016-2017. Latent class analysis was used to examine the typological structure underlying ageing parents' care expectations. Four patterns of care expectations were discovered: mixed-maximal, filial-modest, formal-modest and neither-minimal. Multinomial logistic regression analysis was conducted to validate the newly proposed care expectation model. In addition to certain predisposing factors (participants' age, sex and education), parental enabling resources (economic status), health characteristics (physical, mental and functional health status), children-related enabling characteristics (number of sons and marital status of children), and intergenerational enabling circumstances (intergenerational relationships and caregiving to their own parents) were introduced into the model and found to be associated with ageing parents' care expectations. The findings can inform policy and programmes that effectively respond to ageing adults' diverse care expectations in Hong Kong and have implications for other Asian societies facing rapid population ageing and increasing care demands.
