Abstract Somatic embryogenesis is an essential technology for high productivity in plant culture, and with the advent of nanotechnology, the synergism between these areas could be the answer to developing concepts involving Agriculture 4.0. This perspective permeates both areas, presenting the opportunities and challenges for the consolidation of ideas involving the application of nanoparticles to micropropagation processes (callus induction, preservation, growing, and modification, among others) and also to the production of byproducts (such as biosynthesis of nanoparticles and production of secondary metabolites). Nanotoxicological aspects are also emphasized as well as up-to-date instrumentation involved in these studies.

Abstract The heterogeneity properties shown by cells or unicellular organisms have led to the development of analytical methods at the single-cell level. In this sense, considering the importance of trace elements in these biological systems, the inductively coupled plasma mass spectrometer (ICP-MS) configured for analyzing single cell has presented a high potential to assess the evaluation of elements in cells. Moreover, advances in instrumentation, such as coupling laser ablation to the tandem configuration (ICP-MS/MS), or alternative mass analyzers (ICP-SFMS and ICP-TOFMS), brought significant benefits, including sensitivity improvement, high-resolution imaging, and the cell fingerprint. From this perspective, the single-cell ICP-MS has been widely reported in studies involving many fields, from oncology to environmental research. Hence, it has contributed to finding important results, such as elucidating nanoparticle toxicity at the cellular level and vaccine development. Therefore, in this review, the theory of single-cell ICP-MS analysis is explored, and the applications in this field are pointed out. In addition, the instrumentation advances for single-cell ICP-MS are addressed.

Abstract Upper limb paresis occurs quite commonly after stroke and may result in the affected limb's disuse or learned non-use. As a result, its cortical representation may be suppressed, further inhibiting its spontaneous utilization, resulting in motor function deterioration, increase in spasticity, joint stiffness and pain. The aim of this work was to involve stroke survivors in a qualitative study within a user centred design process to better understand the perspectives of stroke survivors on virtual reality - based serious games (SG) for upper limb rehabilitation during the chronic phase and use those insights to design a VR-based serious games which promotes activation of the affected cortical area.A qualitative research method was performed using a multi professional focus group discussion, including a representative group of stroke survivors. With the insights of this work, the authors initiated the design of a VR-based SG prototype for upper limb rehabilitation with two modes, one version to be played with any arm to pick a virtual hammer and hit targeted objects, and other version, a mirror version based on mirror therapy.

Abstract A wealth of converging research lines has led support to the notion that specialized neural processes output a priori information about the expected effects of gravity to fine-tune motor and perceptual responses to dynamic events. Arguably, these putative internal models of gravity might modulate the efficiency in visual search for objects conforming or not to gravitationally coherent dynamics. In the present work, we explored this possibility with a visual search task involving arrays of two to eight objects moving periodically back and forth. The target could be an accelerating/decelerating ball (as if bouncing on earth's surface-1g) with distractors moving at a constant speed (0g) or the reverse. Moreover, the direction of the gravitational pull, as implied by the 1g motion patterns, could be aligned or misaligned with Earth's gravity. Overall, searches for 1g targets were more efficient than 0g targets except, notably, when stimuli displays were congruent with Earth's gravitational pull, in which case the visual search asymmetry is significantly reduced. Outcomes are interpreted as reflecting the joint and mutually cancelling contribution of low-level detection of acceleration patterns and higher level detection of unexpected violations of gravitational motion. Abstract Objective Patients with multiple long-term conditions visit various healthcare professionals and are exposed to medication information from various sources causing an increased risk of patients perceiving contradictory medication information. The aims of this study are to: (1) characterise conflicting medication information perceived by patients with long-term conditions, (2) better understand the related impact on patients’ medication self-management and healthcare system navigation and (3) explore ways in which such events could be prevented.Design This study was conducted through qualitative semistructured interviews. Data were analysed using thematic analysis.Setting Community pharmacies and medical centres in Geneva, Switzerland.Participants This study included outpatients from April 2019 to February 2020. Patients were included after participating in a quantitative survey of perceived conflicting information about medications for long-term diseases. Methods Semistructured audiotaped interviews of 20 to 60 min following a pre-established interview guide to explore participants’ perceptions of conflicting information. Interviews were transcribed verbatim, and a thematic analysis was conducted with inductive and deductive coding using MAXQDA (2018, Release 18.2.3).Results Twenty-two patients were interviewed, until data saturation, mentioning indication or need for a medication as the main topic of conflicting information between two healthcare professionals. Perceived conflicting information often resulted from insufficient information provided and poor communication leading to confusion, doubts and medication non-adherence. Patients expected more information and more interprofessional communication on their medications. As a result of conflicting information, most participants learnt or were learning to take an active role and become partners of the healthcare providers. Conclusion The need to strengthen and improve communication and interprofessional collaborative practice among healthcare professionals and ...

Abstract Summary: Idiopathic inflammatory myopathies (IIM) are a group of chronic autoimmune diseases mainly affecting proximal muscles. Absence of meaningful prognostic factors in IIM has hindered new therapies development. Glycans are essential molecules that regulate immunological tolerance and consequently the onset of autoreactive immune response. We showed that muscle biopsies from patients with IIM revealed a deficiency in the glycosylation pathway resulting in loss of branched N-glycans. At diagnosis, this glycosignature predicted disease relapse and treatment refractoriness. Peripheral CD4+ T cells from active-disease patients shown a deficiency in branched N-glycans, linked to increased IL-6 production. Glycan supplementation, restoring homeostatic glycosylation profile, led to a decrease in IL-6 levels. This study highlights the biological and clinical importance of glycosylation in IIM immunopathogenesis, providing a potential mechanism for IL-6 production. This pinpoints muscle glycome as promising biomarker for personalized follow-up and a potential target for new therapies in a patients’ subgroup with an ominous evolution.
