    Abstract In shock there is a significant mismatch between oxygen supply and consumption. In recent years the classification of forms of shock has been established based on pathophysiological and clinical aspects. The term distributive shock includes septic, anaphylactic and neurogenic shock. All these forms share a distinct vasoplegia with a relative volume deficiency. The adequate treatment of patients with distributive shock includes a rapid diagnosis and a consistent emergency treatment consisting of volume and catecholamine administration as well as additional specific emergency procedures when necessary.

    Abstract Not only in Europe and USA, but also in many other countries rare disorders-so-called orphan diseases-have attracted more and more attention. The formation of specialized centers for rare disorders has enabled the diagnosis of diseases that have been widely unknown before. In addition, pharmaceutical companies have recognized orphan diseases as a profitable source of revenue. The development and marketing of new drugs for rare diseases-so-called orphan diseases-means a great challenge for all who participate in the health care system: Because the number of patients who are available for a clinical study is mostly very small, it is often very difficult or even impossible to show statistically firm evidence of efficacy. The standard placebo-controlled, double-blind clinical trial is often inappropriate for the approval procedure of an orphan drug; thus other study designs or other parameters (e.g. biomarkers) have to be used to prove clinical efficacy of the study drug. Only relatively small amounts of drugs can be sold to the generally few patients affected by an orphan disease and clinical trials require an high amount of financial investment; therefore orphan drugs have in general extremely high prices. How long these high expenses can be borne by the health care system in view of the great number of rare diseases remains questionable.

    Abstract Quantitative mass spectrometry enables to monitor the abundance of thousands of proteins across biological conditions. Currently, most data analysis approaches rely on the assumption that the majority of the observed proteins remain unchanged across compared samples. Thus, gross morphological differences between cell states, deriving from, e.g., differences in size or number of organelles, are often not taken into account. Here, we analyzed multiple published datasets and frequently observed that proteins associated with a particular cellular compartment collectively increase or decrease in their abundance between conditions tested. We show that such effects, arising from underlying morphological differences, can skew the outcome of differential expression analysis. We propose a method to detect and normalize morphological effects underlying proteomics data. We demonstrate the applicability of our method to different datasets and biological questions including the analysis of sub‐cellular proteomes in the context of Caenorhabditis elegans aging. Our method provides a complementary perspective to classical differential expression analysis and enables to uncouple overall abundance changes from stoichiometric variations within defined group of proteins.

    Abstract On the basis of revisions of some of the systematic errors, we reanalyzed the electron-antineutrino angular correlation (a coefficient) in free neutron decay inferred from the recoil energy spectrum of the protons which are detected in 4π by the aSPECT spectrometer. With a=-0.104 02(82) the new value differs only marginally from the one published in 2020. The experiment also has sensitivity to b, the Fierz interference term. From a correlated (b,a) fit to the proton recoil spectrum, we derive a limit of b=-0.0098(193) which translates into a somewhat improved 90% confidence interval region of -0.041≤b≤0.022 on this hypothetical term. Tighter constraints on b can be set from a combined [shown as superscript (c)] analysis of the PERKEO III (β asymmetry) and aSPECT measurement which suggests a finite value of b with b^{(c)}=-0.0181±0.0065 deviating by 2.82σ from the standard model.

    Abstract Static posturography is a simple non-invasive technique commonly used in contemporary labs and clinics to quantify the central nervous system adaptive mechanisms involved in the control of posture and balance. Its diagnostic value, however, is quite limited due to the lack of posturographic standards for the stable posture. To solve this problem, in this research, we aimed to establish reference values for the stable human posture using our novel parameters of static posturography including the sway anteroposterior directional index (DIAP), the mediolateral directional index (DIML), the stability vector amplitude (SVamp), and the stability vector azimuth (SVaz). Towards this end, in a population of young (mean age 22 yrs), healthy able-bodied volunteers (50 males and 50 females), trajectories of postural sway, based upon the center-of-pressure (COP), were assessed. The experiment consisted of ten 60 s trials that were carried out 5 times while subjects were standing quietly on the force plate with eyes open (EO test) and 5 times with eyes closed (EC test). Results showed that in young healthy subjects, regardless of gender, the basic variables of COP remained at the following levels: SVamp = 9.2 ± 1.6 mm/s, SVaz = 0.9 ± 0.1 rad, and directional indices DIAP = 0.7 ± 0.05, DIML = 0.56 ± 0.06. Some of the measures were sensitive to visual input (EC trials), and showed a weak to moderate correlation with anthropometric features. These measures can be recommended as reference values that characterize the most stable erect posture.
