    Abstract This study aimed to quantify verbascoside (VEB), perform molecular docking studies of VEB with the α-glucosidase (GL) of Bacillus stearothermophilus, and evaluate the inhibition of the enzyme by L. dulcis preparations. The substrate concentration and presence of reduced glutathione were evaluated for their effect on the in vitro inhibition of the GL enzyme. Assays were also performed in the presence and absence of simulated gastric fluid. The antidiabetic fractions 2 and 3 were the most inhibited GL, but their activity were significantly decreased in the presence of gastric fluid. Chromatographic analyses confirmed the predominant presence of VEB in the samples. The samples had VEB concentrations between 49.9 and 243.5 mg/g. Simulation of the molecular docking of VEB were consistent with its GL-inhibitory activity. It can conclude that the crude ethanol extract and fractions show inhibitory activity against the GL enzyme.

    Abstract This study aimed to define progesterone 5β-reductases (P5βR, EC 1.3.99.6, enone 1,4-reductases) as function-associated molecular markers at the plant family level. Therefore cDNAs were isolated from 25 Brassicaceae species, including two species, Erysimum crepidifolium and Draba aizoides, known to produce cardiac glycosides. The sequences were used in a molecular phylogeny study. The cladogram created is congruent to the existing molecular analyses. Recombinant His-tagged forms of the P5βR cDNAs from Aethionema grandiflorum, Draba aizoides, Nasturtium officinale, Raphanus sativus and Sisymbrium officinale were expressed in E. coli. Enone 1,4-reductase activity was demonstrated in vitro using progesterone and 2-cyclohexen-1-one as substrates. Evidence is provided that functional P5βRs are ubiquitous in the Brassicaceae. The recombinant P5βR enzymes showed different substrate preferences towards progesterone and 2-cyclohexen-1-one. Sequence comparison of the catalytic pocket of the P5βR enzymes and homology modelling using Digitalis lanata P5βR (PDB ID: 2V6G) as template highlighted the importance of the hydrophobicity of the binding pocket for substrate discrimination. It is concluded that P5βR genes or P5βR proteins can be used as valuable function-associated molecular markers to infer taxonomic relationship and evolutionary diversification from a metabolic/catalytic perspective.

    Abstract The VIP collaboration is performing high sensitivity tests of the Pauli Exclusion Principle for electrons in the extremely low cosmic background environment of the underground Gran Sasso National Laboratory INFN (Italy). In particular, the VIP-2 Open Systems experiment was conceived to put strong constraints on those Pauli Exclusion Principle violation models which respect the so-called Messiah-Greenberg superselection rule. The experimental technique consists of introducing a direct current in a copper conductor, and searching for the X-rays emission coming from a forbidden atomic transition from the L shell to the K shell of copper when the K shell is already occupied by two electrons. The analysis of the first three months of collected data (in 2018) is presented. The obtained result represents the best bound on the Pauli Exclusion Principle violation probability which fulfills the Messiah-Greenberg rule.

    Abstract Mikania glomerata Spreng. and Mikania laevigata Sch.Bip. ex Baker are medicinal plants popularly named 'guaco', whose leaves are used to treat respiratory diseases, with coumarin (1) and kaurane-type diterpenes regarded as the bioactive constituents. The goal of the study was to undertake seasonal studies on the contents of chemical markers in leaves under different shading conditions. Species were cultivated under different levels of solar radiation and full sunlight. The leaves were collected in the middle of each season year.The contents of 1, o-coumaric (2), benzoylgrandifloric (3), cinnamoylgrandifloric (4) and kaurenoic (5) acids were quantified in dried leaves of both species by RP-HPLC [1].Significant differences were found in the contents of cinnamic acid derivatives (1 and 2) and kaurane-type diterpenes (3, 4 and 5) for the evaluated harvesting periods and cultivation environments. o-Coumaric acid was solely detected in M. laevigata in concentrations bellow the limit of quantification (<0,045%), in plants under 80% shading, collected in the autumn. Both 1 and 2 were not detected in the analyzed samples of M. glomerata.The average concentration of coumarin reached its maximum (0.94%) in the summer, in plants growing under 80% shading.In general, both species presented higher amounts of the kaurane-type diterpenes in plants cultivated under sunlight, except for 3 in M. glomerata. Altogether, the obtained results point out that the highest content of coumarin is reached in M. laevigata cultivated under 80% shading, preferentially harvested in the summer, but with reduced levels of kaurane-type diterpenes.On the other hand, M. glomerata shall be cultivated under full sunlight for maximum contents of the kaurane-type diterpenes.

    Abstract Mikania laevigata Sch. Bip. ex Baker and Mikania glomerata Spreng., known in Brazil as guaco, are medicinal species widely used to treat respiratory affections. Stability analyses of vegetal drugs are crucial to assure the quality of derived products. The present study aimed at undertaking qualitative and quantitative analysis of cheminal markers [coumarin (CO), o-coumaric (OC), kaurenoic (KA), benzoylgrandifloric (BA) and cinnamoylgrandifloric (CA) acids] in dried leaves of M. laevigata and M. glomerata submitted to long-term storage. The plant materials were stored in a dark room with controlled temperature and humidity, and had their fingerprints analyzed three-monthly up to 18 months. Changes in chemical markers were evaluated by UV spectral purity of the peaks and by quantitative analysis of their contents (% w/w in dried leaves), employing an HPLC method previously reported by us [1]. The concentrations of the chemical markers did not vary significantly within the evaluated storage period (p>0.05) for both species. In contrast, changes in BA, CA and KA peaks were detected for three-months stored samples of both species and CO peak, found only in M. laevigata, was detected after six months of storage, suggesting compound degradation. The CO contents in M. laevigata samples ranged from 0.10±0.03% to 0.12±0.03% and therefore fulfill the Brazilian pharmacopoeical requirement established for the species (minimum of 0.1% w/w), except for the 12-month sample (0.09±0.03%) [2].Therefore, the quality control of Mikania species should be based both on the quantification of the selected compounds and fingerprint analysis.
