    Abstract Background: Cholelithiasis is one of the most common disorders of hepatobiliary system. Gut bacteria may be involved in the process of gallstone formation and are, therefore considered as potential targets for cholelithiasis prediction.
    Objective: To reveal the correlation between cholelithiasis and gut bacteria.
    Methods: Stool samples were collected from 100 cholelithiasis and 250 healthy individuals from Huzhou Central Hospital; The 16S rRNA of gut bacteria in the stool samples was sequenced using the third-generation Pacbio sequencing platform; Mothur v.1.21.1 was used to analyze the diversity of gut bacteria; Wilcoxon rank-sum test and linear discriminant analysis of effect sizes (LEfSe) were used to analyze differences in gut bacteria between patients suffering from cholelithiasis and healthy individuals; Chord diagram and Plot-related heat maps were used to analyze the correlation between cholelithiasis and gut bacteria; six machine algorithms were used to construct models to predict cholelithiasis.
    Results: There were differences in the abundance of gut bacteria between cholelithiasis and healthy individuals, but there were no differences in their community diversity. Increased abundance of Costridia, Escherichia flexneri, and Klebsiella pneumonae were found in cholelithiasis, while Bacteroidia, Phocaeicola, and Phocaeicola vulgatus were more abundant in healthy individuals. The top four bacteria that were most closely associated with cholelithiasis were Escherichia flexneri, Escherichia dysenteriae, Streptococcus salivarius, and Phocaeicola vulgatus. The cholelithiasis model based on CatBoost algorithm had the best prediction effect (sensitivity: 90.48%, specificity: 88.32%, and AUC: 0.962).
    Conclusion: The identification of characteristic gut bacteria may provide new predictive targets for gallstone screening. As being screened by the predictive model, people at high risk of cholelithiasis can determine the need for further testing, thus enabling early warning of cholelithiasis.

    Abstract Humans have adopted many different methods to explore matter imaging, among which high content imaging (HCI) could conduct automated imaging analysis of cells while maintaining its structural and functional integrity. Meanwhile, as one of the most important research tools for diagnosing human diseases, HCI is widely used in the frontier of medical research, and its future application has attracted researchers' great interests. Here, the meaning of HCI was briefly explained, the history of optical imaging and the birth of HCI were described, and the experimental methods of HCI were described. Furthermore, the directions of the application of HCI were highlighted in five aspects: protein localization changes, gene identification, chemical and genetic analysis, microbiology, and drug discovery. Most importantly, some challenges and future directions of HCI were discussed, and the application and optimization of HCI were expected to be further explored.

    Abstract Triazole pesticides are organic nitrogen-containing heterocyclic compounds, which contain 1,2,3-triazole ring. In order to develop potential glucosamine-6-phosphate synthase (GlmS) inhibitor fungicides, forty compounds of triazole derivatives were synthesized in an efficient way, thirty nine of them were new compounds. The structures of all the compounds were confirmed by high resolution mass spectrometer (HRMS), 1H-NMR and 13C-NMR. The fungicidal activities results based on means of mycelium growth rate method indicated that some of the compounds exhibited good fungicidal activities against P. CapasiciLeonian, Sclerotinia sclerotiorum (Lib.) de Bary, Pyricularia oryzae Cav. and Fusarium oxysporum Schl. F.sp. vasinfectum (Atk.) Snyd. & Hans. at the concentration of 50 µg/mL, especially the inhibitory rates of compounds 1-d and 1-f were over 80%. At the same time, the preliminary studies based on the Elson-Morgan method indicated that the compounds exhibited some inhibitory activity toward glucosamine-6-phosphate synthase (GlmS). These compounds will be further studied as potential antifungal lead compounds. The structure-activity relationships (SAR) were discussed in terms of the effects of the substituents on both the benzene and the sugar ring.

    Abstract The Yangtze River is the longest river in China, and the river basin spans one fifth of the area of the whole country. Based on statistical data, the excretion of manure-borne steroid hormones, including steroid estrogens (SEs) and steroid androgens (SAs), in 10 provinces of China within the region has been estimated. The potential environmental and ecological risk of manure-borne steroid estrogens to the surface water in this region was also assessed. The manure-borne SE and SA excretions in the 10 provinces and municipalities vary in the order: Sichuan>Hunan>Hubei>Yunnan>Jiangsu>Anhui>Jiangxi>Chongqing>Qinghai>Shanghai. The highest increase of manure-borne SEs (1434.3kg) and SAs (408.5kg) was found in Hunan and Hubei provinces, respectively, and the total excretion in 2013 was 65% more than 15years earlier in these two provinces. However, the emissions in Anhui and Shanghai decreased in this 15year period of time. Swine urine, chicken feces, cattle urine, and laying hen feces were considered the dominant sources of manure-borne E1, βE2, αE2, and SAs, respectively. Although Jiangsu province did not have the largest excretion of manure-borne SEs, it had the highest level of predicted 17β-estradiol equivalency (EEQs) value of 16.65ng/L in surface water because of the limited surface water resources. According to the lowest observable effect level of 10ng/L for 17β-estradiol, the manure-borne SEs in Jiangsu province might potentially pose ecological risk to its wild aquatic organisms.
