    Abstract Hemophilia A (HA) is an X-linked recessive disorder and the second most common coagulation disorder with an incidence of 1 in 5,000 live born males. Worldwide, there are 178,500 affected individuals, 60% with the severe form of the disease. Intron 22 and 1 inversions (Inv22 and Inv1) are the most frequent molecular alterations found in severe HA patients with a frequency of 45-50% and 0.5-5%, respectively. We have implemented a systematic cost-effective strategy for the identification of the molecular alteration in HA patients using Inverse shifting-PCR for Inv22 and Inv1, followed by the analysis of the F8 gene coding region by means of high resolution melting (HRM) PCR and Sanger sequencing in Inv22 and Inv1 negative patients. A total of 33 male HA patients and 6 women were analyzed. Inversion 22 was detected in 14/33 male patients (42.4%), 3/33 (9.1%) had Inv1, 3/33 (9.1%) had large structural variants, and 11/33 (33.3%) single nucleotide/ small frameshift variants. No genetic variant was found in 2/33 patients (6%). With this systematic approach we detected pathogenic variants in 31 out of 33 male affected individuals (94%) tested for the first time.in a cohort of patients from Colombia.

    Abstract Chemical defoliation of seed corn production fields accelerates seed maturation and desiccation and expedites seed harvest. Early seed harvest is important to minimize the risk of frost damage while in the field. This newly adopted seed production practice also allows seed companies to plan harvest and manage dryer space more efficiently. However, premature defoliation may interfere with the migration of oil bodies within embryo cells during desiccation and affect seed germination and vigor. The objective of this study was to investigate the effect of chemical defoliation on the migration patterns of oil bodies within embryo cells during desiccation. Chemically defoliated and non-defoliated plants from five commercial hybrid seed corn fields were sampled in 2014 and 2015. Whole ears with husks were harvested before and after defoliant application at 600 g H₂O kg⁻¹ fresh weight (fw), and weekly thereafter until seed reached approximately 300–350 g H₂O kg⁻¹ fw. Ten embryos extracted from center-row seeds were fixed to stop metabolic processes, then sliced, processed, and photographed using scanning transmission electron microscopy. The oil bodies within embryo cells followed normal migration patterns according to seed moisture content, regardless of defoliation treatment. Seed germination and vigor were verified and were not significantly affected by defoliation. Chemical defoliation is a viable production practice to accelerate seed corn desiccation and to manage harvest and seed dryer availability more efficiently without negatively affecting seed germination and vigor.

    Abstract Introduction: Between 60 and 80% of patients with B-cell acute lymphoblastic leukemia show genetic abnormalities which influence the prognosis of the disease and the biology of the tumor.
    Objective: To analyze different genetic abnormalities in acute B lymphoblastic leukemia in children, its relationship with the immunophenotype and the proliferative rate compared with normal B cell precursors.
    Materials and methods: We assessed immunophenotype, DNA content and proliferative rate in 44 samples by flow cytometry, and translocations t(9;22), t(12;21), t(4;11), and t(1;19) by RT-PCR. Using a hierarchical cluster analysis, we identified some immunophenotypic patterns associated to genetic abnormalities when compared with normal B cell precursors.
    Results: DNA quantification showed that 21% of the cases had high hyperdiploidy and 47.7% has low hyperdiploidy. The presence of hyperdiploidy was associated with increased tumor proliferation and aberrant immunophenotypes, including abnormal expression of CD10, TdT, CD38, and CD45 and an increased size of the lymphoblasts. The presence of t(9;22) and t(12;21) discriminates normal cells from tumor cells with aberrant immunophenotype in the expression of CD19, CD22, CD13, CD33, CD38, CD34, and CD45.
    Conclusions: The aberrant immunophenotype profile detected in neoplastic cells along with abnormalities in the proliferative rate were significantly associated with DNA hyperdiploidy and clearly distinguished lymphoblasts with t(9;22) and t(12;21) from normal B cell precursors. The identification of these parameters is useful as a tool for classification and monitoring of these patients.

    Abstract The aim of this study was to evaluate the diversity of Enterobacteriaceae present in soil and tomato fruits from three greenhouses with fertirigation system. These crop systems are an important alternative for production in protected agriculture; however, there is little information about the microbiological quality of the fruit and its relationship with chemical soil characteristics. Soil evaluations consisted of analyzing organic matter content and pH. In the microbiological analysis were isolated and identified enterobacterias organisms from composite samples of soil and fruits at different stages of maturity (0, 50 and 100%). Culture media used was selective, differential and confirmatory testing with VITEK system. Enteropathogenic E. coli (EPEC) and enterotoxigenic E. coli (ETEC) were characterized genotypically, amplifying the lngA and bfpA genes by the technique of polymerase chain reaction (PCR). Diversity index (Simpson (D), Shannon-Wiener (H') and Chao estimator (SChao1) were calculated with the identified species. The species Enterobacter cloacae, Citrobacter freundii and C. brakii had a higher frequency of isolation, EPEC and ETEC were identified in soil samples and in fruits with 100% maturity. In soil, H' indices were positively correlated with the highest organic matter percentages. In fruit, although H 'and D showed less diverse bacterial communities, the isolation of ETEC and Shigella boydii on the fruit surface compromise their safety because they are usually consumed raw. DOI: 10.17268/sci.agropecu.2012.02.07
