    Abstract The intricate mechanical interplay between cell membranes and enclosed filaments or filament networks plays an essential role in governing cellular morphological transformations and various cellular activities. A profound understanding of fundamental mechanical mechanisms underlying these interactions can be gained by exploring mechanical behaviors of a filament–vesicle system. Here we theoretically investigate the axial stretching of a lipid vesicle containing an enclosed filament loop. The vesicle either freely adjusts its volume or maintains a fixed volume. Depending on the relative stiffness and size of the filament loop to the vesicle as well as the vesicle scenario, the vesicle prior to stretching exhibits a spherical, prolate or oblate shape, and during stretching it undergoes morphological transformations with pronounced filament distortion and reorientation. The filament–vesicle system could exhibit a spring-softening or -stiffening response during stretching. Moreover, effects of filament inhomogeneity on the stretching process are studied. Our results could serve as a quantitative benchmark for further investigations into cellular complexity.

    Abstract Objective This study aimed to estimate the prevalence of hyperuricaemia (HUA) and investigate its risk factors in the general adult population of Ningxia Hui Autonomous Region (NHAR), China.Design Cross-sectional study.Setting Survey of cardiovascular disorders and their related risk factors in NHAR, China.Participants 10 803 permanent residents aged 18 and older.Main outcome measures HUA was defined as serum uric acid levels >420 µmol/L for men and >360 µmol/L for women.Results The overall prevalence of HUA in NHAR adults was 19.81% (95% CI 19.06 to 20.57), with prevalence values of 24.91% (95% CI 23.70 to 26.14) in men and 15.58% (95% CI 14.66 to 16.53, p<0.001) in women. The prevalence of HUA was higher in urban residents than in rural residents (23.26% vs 17.02%, p<0.001). HUA prevalence was relatively high in individuals younger than 30 years for both men and women, then decreased with age, and began to increase at the age of 40 for women and 60 for men. Higher level of education, being overweight or obese, alcohol consumption, hypertension, diabetes, higher triglycerides, higher total cholesterol and poorer renal function were associated with an increased risk of HUA.Conclusions HUA prevalence is high among adults in NHAR. Young adults under 30 years and women over 50 years were identified as populations at high risk for HUA. Further attention ought to be placed to promoting healthy diets and implementing early interventions to manage dyslipidaemia, obesity and blood glucose level, as well as advocating for moderation of alcohol consumption.

    Abstract Background RRM2 plays an important role in different malignant tumors, but there are few studies in breast cancer. Public databases were used to analyze the expression of RRM2 in breast cancer and its prognostic value. Materials and methods A total of 2,509 breast cancer samples were downloaded from the METABRIC database. The relationship between RRM2 expression and clinical pathology was evaluated. Using the BCIP database and real-time-PCR, and western blotting, RRM2 mRNA and protein expression of RRM2 in breast cancer tissues and cell lines were evaluated. Univariate and multivariate analysis defined independent prognostic factors that affected the overall survival of patients with breast cancer. The Kaplan-Meier method was used to study the relationship between the high expression of RRM2 and overall survival and distant metastasis-free survival (DMFS) of breast cancer patients. Finally, We performed Gene Set Enrichment Analysis (GSEA) and obtained the relevant pathways associated with high expression of RRM2 potentially influencing breast cancer progression. Results RRM2 expression was significantly correlated with age, tumor size, grade, menopausal status, molecular typing, ER, PR, and Her-2 of patients with breast cancer(P<0.05). Univariate and multivariate regression analysis showed that RRM2, the number of positive lymph nodes, ER, Her-2, tumor size, and tumor stage can be used as independent prognostic factors for overall survival of patients with breast cancer. Kaplan-Meier analysis showed that in patients with Luminal A and Normal like breast cancers and Stage1 and stage2 breast cancers, patients with high expression of RRM2 had worse overall survival and DMFS. The analysis of the GSEA pathway showed that RRM2 is mainly enriched in the ERBB signaling pathway and other pathways. Conclusion The high expression of RRM2 has a worse prognosis in patients with breast cancer with specific features. It can be used as a biomarker for the prognosis of breast cancer.

    Abstract Diabetes causes lipid peroxide to accumulate within cardiomyocytes. Furthermore, lipid peroxide buildup is a risk factor for ferroptosis. This study is aimed at examining whether curcumin can ameliorate ferroptosis in the treatment of diabetic cardiomyopathy. Hematoxylin and eosin and Masson sections were used to examine the morphology, arrangement, and degree of fibrosis of the myocardium of diabetic rabbit models. The expression levels of nuclear Nrf2, Gpx4, Cox1, and Acsl4 in diabetic animal and cell models were quantitatively analyzed using immunofluorescence and western blotting. Nrf2-overexpression lentivirus vectors were transfected into cardiomyocytes, and the protective effects of curcumin and Nrf2 on cardiomyocytes under high glucose stimulation were assessed using terminal deoxynucleotidyl transferase dUTP nick-end labelling and reactive oxygen species probes. Diabetes was found to disorder myocardial cell arrangement and significantly increase the degree of myocardial fibrosis and collagen expression in myocardial cells. Curcumin treatment can increase nuclear transfer of Nrf2 and the expression of Gpx4 and HO-1, reduce glucose induced myocardial cell damage, and reverse myocardial cell damage caused by the ferroptosis inducer erastin. This study confirmed that curcumin can promote the nuclear translocation of Nrf2, increase the expression of oxidative scavenging factors, such as HO-1, reduce excessive Gpx4 loss, and inhibit glucose-induced ferroptosis in cardiomyocytes. This highlights a potentially new therapeutic route for investigation for the treatment diabetic cardiomyopathy.

    Abstract Enjoying local food could be one of the motives for tourism, and local food and restaurant recommendation information would be important for tourists to decide their destination. Recommendation agents are the sorting and searching function to find the best local food and restaurant among the complexity of information, and they could also be helpful for the tourist to decide their destination. Online tourism websites (e.g., Ctrip.com ) started to provide restaurant recommendations containing food-related information and recommendation agents to attract tourists. However, few studies have investigated their impact on the destination visit intention of potential Chinese tourists. This study aims to empirically validate how restaurant recommendation information, including food-related information and recommendation agents, could impact online tourists’ reactions, such as satisfaction, continuous website usage, and destination visits. We developed our hypothesis based on the information system (IS) success model. We gathered 202 data points from potential tourists using quasi-experimental methods, and these data were analyzed by the PLS algorithm. The results indicate that restaurant recommendation information and recommendation agents significantly increase the perceived information quality and perceived system quality. Increased perceived information quality and system quality could significantly increase potential tourists’ satisfaction, website continuous usage intention, and destination visit intention. The results of this study could contribute to making tourism websites more attractive by using local food and restaurant information and recommendation agents.

    Abstract Northeast China is an important ecological barrier and commodity grain base in China. The coupling coordination relationship between geology-geomorphology and ecology has become a critical background condition for ecosystem protection and sustainable development. Taking Northeast China as a case (accounting for about 13% of China's land area), 9 divisions are divided according to the characteristics of regional ecology and geology-geomorphology, and 17 indicators are selected to build an evaluation index system. Methods of analytic hierarchy process, entropy weight and game theory are used to determine the index weights. Based on the coupling coordination degree (CCD) model, the spatial coupling coordination characteristics of geology-geomorphology and ecology are studied. The variation characteristics of the Normalized Difference Vegetation Index (NDVI) are evaluated by Sen+Mann-Kendall (Sen+MK) method. Our results are as follows. (1) The coupling between geology-geomorphology and ecology is strong, but the spatial differentiation of CCD is obvious. Nine divisions are evaluated as two high-level, three medium-level and three low-level coordination types and one mild imbalance type. (2) The plain divisions Ⅰ and Ⅳ where the typical black soil belt is located are high coordination types. Restricted by geology-geomorphological conditions or ecological conditions, mountain divisions Ⅲ and Ⅶ and plain division Ⅴ are moderate coordination types, mountain divisions Ⅱ and Ⅷ and plateau division Ⅸ are low coordination types, and mountain division Ⅵ is mild imbalance type. (3) The variation trend of NDVI shows a significant increase in divisions Ⅲ, Ⅴ, Ⅰ, Ⅱ and Ⅶ. it shows a significant decrease in part of divisions Ⅳ, Ⅵ, Ⅷ and Ⅸ, and ecological management and construction should be strengthened in these divisions. The research shows that the CCD model method is feasible for evaluating the relationship between geology-geomorphology and ecology and can provide eco-geological background information for Northeast China.

    Abstract Pain and injury of the triangular fibrocartilage complex (TFCC) due to overuse or trauma are commonly diagnosed through static MRI scanning, while TFCC is always involved in radial and ulnar deviation of the wrist. To the best of our knowledge, a dynamic MRI diagnostic method and auxiliary tool have not been applied or fully developed in the literature. As such, this study presents the design and evaluation of a dynamic magnetic resonance imaging (MRI) auxiliary tool for TFCC injury diagnosis. First, 3D scanning and Python are used to measure and fit the radial and ulnar deviation trajectories of healthy participants and patients. 3D printing is then used to manufacture the auxiliary tool for dynamic MRI, and dynamic MRI diagnosis is then conducted to explore the clinical effect. The radial and ulnar deviation trajectory is presented as an asymmetric curve without an obvious circular centre, and the results indicate that the designed auxiliary device meets the requirements of the ulnar and radial movements of the human wrist. According to the MRI contrast test results, the image quality score of patients wearing the auxiliary device is higher than for those without. Such devices could assist clinicians in the diagnosis of TFCC damage, and our method could not only serve as the reference standard for clinical noninvasive diagnosis but also help in understanding the disease and improving the accuracy of TFCC diagnosis.

    Abstract A new species, Achalinus hunanensis sp. nov., is described from middle and western Hunan Province based on the results of molecular systematics and morphological characters. It diverges from known congeners by a significant genetic divergence (p-distance 3.2%–16.9% based on CO1 mitochondrial gene), and it can be distinguished from all known congeners by the following morphological characters: (1) all dorsal scales strongly keeled, 23 rows throughout the body, the outmost one strongly keeled and enlarged; (2) tail relatively short, TaL/TL 0.221 ~ 0.225; (3) maxillary teeth 23; (4) the suture between internasals 2 × as long as that between prefrontals; (5) loreal one, subrectangular, LorH/LorL 0.62 ~ 0.70; (6) supralabials 6, the 4th and 5th touch the eye; (7) the two anterior temporals in contact with eye; (8) ventrals 163–165, subcaudals 69–72, not paired. This raises the number of known species of Achalinus to 24.
