    Abstract Background: Traditional Common Spatial Pattern (CSP) algorithms for Electroencephalogram (EEG) signal classification are sensitive to noise and can produce low accuracy in small sample datasets.
    New method: To solve the problem, an improved Empirical Mode Decomposition (EMD) Bagging Regularized CSP (RCSP) algorithm is proposed. It filters EEG signals through improved EMD, inhibits high-frequency noise, retains effective information in the characteristic frequency band, and uses Bagging algorithm for data reconstruction. Feature extraction is performed with regularization of spatial patterns and Fisher linear discriminant analysis for feature classification. T-test is used for classification.
    Results: The improved EMD Bagging RCSP algorithm has improved accuracy and robustness compared to CSP and its derivatives. The average classification rate is increased by about 6%, demonstrating the effectiveness and correctness of the proposed algorithm.Comparison with existing methods: The proposed algorithm outperforms CSP and its derivatives by retaining effective information and inhibiting high-frequency noise in small sample EEG datasets.
    Conclusions: The proposed EMD Bagging RCSP algorithm provides a reliable and effective method for EEG signal classification and can be used in various applications, including brain-computer interfaces and clinical EEG diagnosis.

    Abstract Kernel functions play an important role in a wide range of scientific computing and machine learning problems. These functions lead to dense kernel matrices that impose great challenges in computational costs at large scale. In this paper, we develop a set of fast kernel matrix compressing algorithms, which can reduce computation cost of matrix operations in the related applications. The foundation of these algorithms is the polyharmonic spline interpolation, which includes a set of radial basis functions that allow flexible choices of interpolating nodes, and a set of polynomial basis functions that guarantee the solvability and convergence of the interpolation. With these properties, original data points in the interacting kernel function can be randomly sampled with great flexibility, so the proposed method is suitable for complicated data structures, such as high-dimensionality, random distribution, or manifold. To further boost the algorithm accuracy and efficiency, this scheme is equipped with a QR sampling strategy, and combined with a recently developed fast stochastic SVD to form a hybrid method.     Abstract ACGS Best Practice Guidelines for Variant Classification in Rare Disease 2020, a supplementary practical guidelines, is based on the Standards and Guidelines for the Interpretation of Sequence Variations issued by the American Society for Medical Genetics and Genomics (ACMG) and the Association of Molecular Pathology (AMP) in 2015 by the British Medical Genetics Society under the Clinical Genomics Society (ACGS), and has integrated the detailed rules of standards developed by the ClinGen Sequence Variant Interpretation (SVI) Working Group by 2020. The further development of the ACMG/AMP guidelines is currently undertaken by the ClinGen SVI working group in the United States, which focuses on the classification of high penetrance and protein coding variants. ClinGen has established many expert panels on variants for specific diseases which required various evidence thresholds and is currently developing disease/gene specific guidelines. The British Medical Genetics Society has collected and integrated information on the guidelines for sequence variation classification and their extended rules, forming its own "2020 ACGS Best Practice Guidelines for Rare Disease Variation Classification" and is regularly updating it. The author has translated and summarized it for the reference of Chinese Medical Genetics Practitioners.

    Abstract Immune-related adverse events (irAEs) represent an increasingly concerning challenge in the assessment of biopharmaceutical products. In contrast to historically rare allergic reactions associated with small chemical drugs, contemporary biotherapeutics exhibit a significantly higher morbidity of irAEs, because of their complex structure and comprehensive mechanisms of action. While the immunogenicity of protein-based compounds is associated with the induction of anti-drug antibodies, the pathogenesis of irAEs in advanced biologics, such as cell and gene therapy, remains to be further delineated. In the current study, I present an updated profile regarding the untoward immune effects of medications, covering various material categories systematically, with the underlying mechanisms to inspire risk mitigation in biopharmaceutical development and application.
