Abstract Objective The goal of this study is to use summary generation and topic modeling to identify factors contributing to vaccine attitudes for three different vaccine brands, with the aim of generalizing these factors across different regions. Methods A total of 5562 tweets about three vaccine brands (Sinovac, AstraZeneca, and Pfizer) were collected from 14 December 2020 to 30 December 2021. BERTopic clustering is used to group the tweets into topics, and then contrastive learning (CL) is adopted to generate summaries of each topic. The main content of each topic is generalized into three factors that contribute to vaccine attitudes: vaccine-related factors, health system-related factors, and individual social attributes. Results BERTopic clustering outperforms Latent Dirichlet Allocation clustering in our analysis. It can also be found that using CL for summary generation helped to better model the topics, particularly at the center-point of the clustering. Our model identifies three main factors contributing to vaccine attitudes that are consistent across different regions. Conclusions Our study demonstrates the effectiveness of deep learning methods for identifying factors contributing to vaccine attitudes in different regions. By determining these factors, policymakers and medical institutions can develop more effective strategies for addressing concerns related to the vaccination process.

Abstract There is compelling evidence that synergistic drug combinations have become promising strategies for combating complex diseases, and they have evident predominance comparing to traditional one drug - one disease approaches. In this paper, we develop a computational method, namely SyFFM, that takes pharmacological data into consideration and applies field-aware factorization machines to analyze and predict potential synergistic drug combinations. Firstly, features of drug pairs are constructed based on associations between drugs and target, and enzymes, and indication areas. Then, the synergistic scores of drug combinations are obtained by implementing field-aware factorization machines on latent vector space of these features. Finally, synergistic combinations can be predicted by introducing a threshold. We applied SyFFM to predict pairwise synergistic combinations and three-drug synergistic combinations, and the performance is good in terms of cross-validation. Besides, more than 90% combinations of the top ranked predictions are proved by literature and the analysis of parameters in model shows that our method can help to investigate and explain synergistic mechanisms underlying combinatorial therapy. Abstract During the outbreak of novel coronavirus pneumonia, the number of confirmed cases and deaths in Hubei province of China increased sharply, and the situation in Hubei was more severe than that in non-Hubei, so we do a research on psychological health status evaluation of the public in Hubei and non-Hubei areas. In this paper, we adopt textual analysis and contextual analysis using Simplified Chinese Microblog Word Count (SCMBWC), Five-Factors Model (FFM), Semantic Role Labeling (SRL) to interpret and analyze the public perception and psychological personality based on media news. Through the analysis, it was found that there were great differences in public perception to novel coronavirus pneumonia. In Hubei areas, the public perception was mainly reflected in the overall prevention and the treatment of patients, while in non-Hubei areas, the perception was mainly in the orderly promotion of enterprises to return to work. Through contextual analysis, the novel coronavirus pneumonia had a great psychological impact on the public in different regions. The media covered a large number of social process words and cognitive process words, public showed a personality that was inclined to be “open” and “neurotic” in different areas. Furthermore, we find out some reasons like all kinds of rumors, wildlife trade, all kinds of illegal and criminal acts disturbing social order cause this psychology personality through emotional entity mining based on semantic role labeling. This is conducive to the government's better policies and management in line with local conditions.

Abstract Inspired by the wide application of amides in plant pathogens, a series of novel 1-substituted-5-trifluoromethyl‑1H‑pyrazole-4-carboxamide derivatives were designed and synthesized. Bioassay results indicated that some target compounds exhibited excellent and broad-spectrum in vitro and certain in vivo antifungal activities. Among them, the in vitro EC50 values of Y13 against G. zeae, B. dothidea, F. prolifeatum and F. oxysporum were 13.1, 14.4, 13.3 and 21.4 mg/L, respectively. The in vivo protective activity of Y13 against G. zeae at 100 mg/L was 50.65%. SAR analysis revealed that the phenyl on the 1-position of the pyrazole ring was important for this activity. An antifungal mechanism study of Y13 against G. zeae demonstrated that this compound may disrupt the cell membrane of mycelium, thus inhibiting the growth of fungi. These mechanistic study results were inconsistent with those for traditional amides and may provide a novel view for deep study of this series of pyrazole carboxamide derivatives.

Abstract To clarify the residue formation of prothioconazole and its main metabolite in wheat plants, the uptake, translocation, and metabolism of prothioconazole in wheat roots and leaves were investigated by nutrient solution culture and the spraying method. The results showed that prothioconazole and its metabolites showed a trend of increasing and then decreasing in wheat plants under two treatment methods, and the concentration of prothioconazole and its metabolites was higher in the high-concentration group (5000 μg/mL) than in the low-concentration group (1000 μg/mL). The transferability from stem to leaf was stronger than that from root to stem. In the nutrient solution culture, prothioconazole and its metabolites were mainly enriched in wheat roots. The concentration of prothioconazole in wheat roots increases with the increase in prothioconazole concentration and was significantly higher than the prothioconazole concentration on stems and leaves. In wheat leaves in the spraying method, prothioconazole and its metabolites were conducted from leaves to stems and roots up to the nutrient solution. Prothioconazole-desthio was detectable in wheat nutrient solution, while prothioconazole was not detected. Analysis of actual samples of 9 wheat grains and 28 flours showed that the residues of prothioconazole and its metabolites met the maximum residue limit (0.1 mg/mL) set in China and by the Codex Alimentarius Commission. The results will provide a theoretical basis for the scientific use of prothioconazole and food security assurance.

Abstract Transarterial chemoembolization (TACE) is a first-line treatment for intermediate to advanced-stage liver cancer, with drug-eluting microspheres commonly used as embolic agents. However, currently available drug-eluting microspheres suffer from low drug-loading capacity and limited drug options. In this work, we developed polydopamine-modified polyvinyl alcohol dual-drug-loaded microspheres encapsulating celecoxib and cisplatin (referred to as PCDMS). Physicochemical characterization revealed that the surface of the microspheres displayed increased roughness after polydopamine modification, and celecoxib and cisplatin were successfully loaded onto the microsphere surface. In vitro cell experiments demonstrated that the PCDMS significantly inhibited the proliferation and migration of highly metastatic human liver cancer cells (MHCC-97H) and human liver cancer cells (SMMC-7721). Furthermore, the dual-loaded microspheres exhibited remarkable tumor growth inhibition and reshaped the tumor microenvironment in both subcutaneous H22 liver cancer model in Balb/c mice and intrahepatic VX2 tumor model in New Zealand rabbits, demonstrating a synergistic antitumor effect where 1 + 1>2. This work provides a potential therapeutic approach for the treatment of refractory liver cancer and holds significant translational potential.

Abstract Chinese herbal medicine has recently gained worldwide attention. The curative mechanism of Chinese herbal medicine is compared with that of western medicine at the molecular level. The treatment mechanism of most Chinese herbal medicines is still not clear. How do we integrate Chinese herbal medicine compounds with modern medicine? Chinese herbal medicine drug-like prediction method is particularly important. A growing number of Chinese herbal source compounds are now widely used as drug-like compound candidates. An important way for pharmaceutical companies to develop drugs is to discover potentially active compounds from related herbs in Chinese herbs. The methods for predicting the drug-like properties of Chinese herbal compounds include the virtual screening method, pharmacophore model method and machine learning method. In this paper, we focus on the prediction methods for the medicinal properties of Chinese herbal medicines. We analyze the advantages and disadvantages of the above three methods, and then introduce the specific steps of the virtual screening method. Finally, we present the prospect of the joint application of various methods.

Abstract A type of sandwich-type and mesoporous CoS-based coaxial nanocables with conductive CNT backbone core, well-confined CoS nanoparticle interlayer and conformal carbon coating shell (denoted as CNT@CoS@C) are developed through a bottom-up method and investigated as potential anode materials for sodium/lithium ion storage. The rationally constructed architecture successively achieves the integration of one-dimensional conducting networks, ultrafine active nanoparticles, well-developed mesoporosity and sophisticated surface modification via a layer-by-layer assembly strategy, thus upholding good structural/interfacial robustness and enhanced charge-transfer reaction kinetics. As a result, the CNT@CoS@C coaxial nanocables exhibit a high reversible capacity of 494 mAh g−1, stable cycling with more than 318 mAh g−1 at 500 mA g−1 over 500 cycles (corresponding to 74% capacity retention with 0.05% decay rate per cycle) and impressive rate capability (278 mAh g−1 at 5000 mA g−1) for sodium ion batteries (SIBs) and excellent electrochemical performance for lithium ion batteries (LIBs) (1010 mAh g−1 during 200 cycles with no capacity loss and 467 mAh g−1 at 5000 mA g−1). In addition, the electrochemical experimental results and simulated calculations suggest that the CoS-based active species possesses better electrochemical properties in term of reaction reversibility and structural stability than its Co3O4-based counterpart with similar morphological features.
