    Abstract Despite a strong body of evidence supporting benefits of exercise to reduce severity of motor symptoms of Parkinson's Disease (PD) over time, research on the immediate impact of exercise as an adjunctive therapy for nonpharmacologic management of motor symptoms and mobility performance in people with PD is limited. The purpose of this study was to examine immediate effects of a single bout of high-cadence cycling on motor symptoms of PD and performance-based outcomes. Twenty individuals with idiopathic PD participated in a pretest/posttest study investigating immediate impacts of a single high-cadence cycling session on performance-based outcomes across multiple domains of motor function, mobility, and balance. Outcomes were analyzed based on time since last dose of levodopa/carbidopa medication. Immediate improvements were observed in motor symptoms of PD with significant improvements in performance across multiple balance systems, dynamic gait, and upper-extremity coordination following a cycling intervention.

    Abstract Satellite cells (SCs) are muscle stem cells responsible for muscle hypertrophic growth and the regeneration of damaged muscle. Proliferation and differentiation of the pectoralis major (p. major) muscle SCs are responsive to thermal stress in turkeys, which are, in part, regulated by mechanistic target of rapamycin (mTOR) and Frizzled7 (Fzd7)-mediated wingless-type mouse mammary tumor virus integration site family/planar cell polarity (Wnt/PCP) pathways in a growth dependent-manner. It is not known if chicken p. major SCs respond to thermal stress in a manner similar to that of turkey p. major SCs. The objective of the current study was to investigate the effects of thermal stress and mTOR and Wnt/PCP pathways on the proliferation, differentiation, and expression of myogenic transcriptional regulatory factors in SCs isolated from the p. major muscle of a current modern commercial (MC) broiler line as compared to that of a Cornish Rock (BPM8) and Randombred (RBch) chicken line in the 1990s. The MC line SCs had lower proliferation and differentiation rates and decreased expression of myoblast determination factor 1 (MyoD) and myogenin (MyoG) compared to the BPM8 and RBch lines. Heat stress (43°C) increased proliferation and MyoD expression in all the cell lines, while cold stress (33°C) showed a suppressive effect compared to the control temperature (38°C). Satellite cell differentiation was altered with heat and cold stress in a cell line-specific manner. In general, the differentiation of the MC SCs was less responsive to both heat and cold stress compared to the BPM8 and RBch lines. Knockdown of the expression of either mTOR or Fzd7 decreased the proliferation, differentiation, and the expression of MyoD and MyoG in all the cell lines. The MC line during proliferation was more dependent on the expression of mTOR and Fzd7 than during differentiation. Thus, modern commercial meat-type chickens have decreased myogenic activity and temperature sensitivity of SCs in an mTOR- and Fzd7-dependent manner. The decrease in muscle regeneration will make modern commercial broilers more susceptible to the negative effects of myopathies with muscle fiber necrosis requiring satellite cell-mediated repair.

    Abstract There is a paucity of research focusing on adults with scoliosis, yet many of these individuals suffer from pain and disability. Recent literature has demonstrated that for this patient population general physical therapy is no better than other non-operative treatment options.[1-3] This study assessed the perceptions of the effectiveness of Physiotherapeutic Scoliosis Specific Exercises (PSSE) on adult scoliosis. The purpose of this study is to present the results of a retrospective analysis of how adults with scoliosis perceive that physical therapy utilizing PSSE has impacted their quality of life (QoL), function, and pain. A 10 question survey was sent via a secure server (Qualtrics) to all PSSE participating patients ≥ 18 years old from one PSSE specialty clinic from the beginning of the PSSE program, 7 years. Survey responses were anonymous, collected by someone other than the primary investigator, and statistics were calculated with SPSS 24 (IBM Corp., Armonk, NY). Results: Fifty-seven adults (88.9% female) responded to the survey (55% response rate) with 67.2% being over the age of 55 years. The majority (61.9%) felt that PT had moderately or significantly positively impacted their QoL. The most common number of PT visits ranged from 5-10, after which 71.9% of participants were either somewhat or very confident in their ability to perform their PSSE program unsupervised at home. The most common frequency of home exercise program (HEP) performance was 1-2 times per week (46.0%) for a duration of 5-40 minutes. Quantitative analysis of exercise adherence using a Spearman's rho (rs) revealed positive associations between confidence in correctly performing the HEP with the perceived positive impact of the exercises (rs = .45, p < .001), the greater frequency of performing the HEP (rs = .30, p = .024) and greater time performing the HEP (rs = .33, p = .004). This retrospective analysis showed that 61.9% of adults with scoliosis felt that PT utilizing PSSE had moderately or significantly positively impacted their QoL. Adherence to a HEP is critical to the success of the program. Greater confidence in correctly performing the HEP was positively correlated with perceived impact, frequency and time spent exercising. The sample was limited to adults from one clinic that specializes in scoliosis and may not be generalizable to other clinics.

    Abstract Co-speech gestures are integral to human communication and exhibit diverse forms, each serving a distinct communication function. However, existing literature has focused on individual gesture types, leaving a gap in understanding the comparative neural processing of these diverse forms. To address this, our study investigated the neural processing of two types of iconic gestures: those representing attributes or event knowledge of entity concepts, beat gestures enacting rhythmic manual movements without semantic information, and self-adaptors. During functional magnetic resonance imaging, systematic randomization and attentive observation of video stimuli revealed a general neural substrate for co-speech gesture processing primarily in the bilateral middle temporal and inferior parietal cortices, characterizing visuospatial attention, semantic integration of cross-modal information, and multisensory processing of manual and audiovisual inputs. Specific types of gestures and grooming movements elicited distinct neural responses. Greater activity in the right supramarginal and inferior frontal regions was specific to self-adaptors, and is relevant to the spatiomotor and integrative processing of speech and gestures. The semantic and sensorimotor regions were least active for beat gestures. The processing of attribute gestures was most pronounced in the left posterior middle temporal gyrus upon access to knowledge of entity concepts. This fMRI study illuminated the neural underpinnings of gesture-speech integration and highlighted the differential processing pathways for various co-speech gestures.

    Abstract Background: Ehlers-Danlos Syndrome (EDS) is a group of inherited connective tissue disorders which predominantly affects women and has a prevalence as high as 1 in 5,000 individuals. Hypermobile EDS (hEDS) is the most common subtype of EDS and is characterized by multi-joint pain, particularly in large joints such as the shoulder. Physical therapy is often utilized to address the pain, physical impairments, and functional loss in patients with EDS. Kinesiology Tape (KT) is an intervention commonly used by physical therapists for treating shoulder pain and dysfunction. Studies related to the effectiveness of KT in patients with shoulder pain is equivocal and there are a lack of studies specifically studying the effects of KT in an EDS population.
    Purpose: The purpose of this study was to assess the efficacy and short-term effects of two different KT techniques on shoulder pain and function in individuals with hEDS and shoulder pain.
    Methods: Participants were recruited from EDS support groups in the New England area of the United States; were diagnosed with hEDS by their physician; and had shoulder pain. Baseline demographic information was obtained for each participant followed by completion of 4 patient reported outcome (PRO) measures: the Upper Extremity Functional Index, QuickDASH (Disabilities of the Arm, Shoulder, & Hand), Shoulder Pain and Disability Index, and the Western Ontario Shoulder Instability Index. Current pain level, average pain over the past 24 h, and worst pain over the past 24 h were recorded using the numeric pain rating scale (NPRS). Subjects were randomly assigned to receive either an experimental shoulder KT procedure or a control shoulder taping. Immediately after taping, the NPRS was reassessed. Subjects then returned 48 h later to repeat the NPRS and PRO measures.
    Results: There was no significant difference between the experimental and control tape groups for any outcome measure. There was a significant improvement from pre-taping to 48-hours post taping for each of the 4 PRO measures with large effect sizes (
    Conclusion: KT is an inexpensive and relatively safe intervention that is easy to apply and can offer temporary improvements in pain and function for patients with EDS and shoulder pain.

    Abstract Ciliates usually with big cell sizes, complex morphological structures, and diverse life cycles, are good model organisms for studying cell proliferation regulation of eukaryotes. Up to date, the molecular regulation mechanisms for the vegetative cell cycle and encystment of these ciliates are poorly understood. Here, transcriptomes of Apodileptus cf. visscheri, which has an asexual vegetative cell cycle and is apt to encyst when environmental conditions become unfavorable, were sequenced to enrich our related knowledge. In this study, three replicates were sequenced for each of four cell stages, including initial period of growth, morphogenesis, cell division, and resting cyst. The significant transcription differences, involving cell cycle, biosynthesis, and energy metabolism pathways, were revealed between the resting cyst and vegetative cell cycle. Further investigations showed that the cell cycle pathway was enriched during morphogenesis stage and cell division stage. Compared to the initial period of growth stage, the differentially expressed genes involved in cellular components and molecular function were significantly enriched during cell division stage, while cellular components and biological processes were significantly enriched during morphogenesis stage. These provide novel insights into a comprehensive understanding at the molecular level of the survival and adaptive mechanism of unicellular eukaryotes.

    Abstract Purpose: To provide pragmatic guidance for acute rehabilitation management and implementation of early mobility for individuals with critical illness due to COVID-19.
    Methods: Clinical perspective developed through reflective clinical practice and narrative review of best available evidence.
    Results: Current clinical practice guidelines do not provide guidance for implementation of early mobility interventions for individuals with critical illness due to COVID-19 who require enhanced ventilatory support or support of inhaled pulmonary artery vasodilators. Many individuals who may benefit from implementation of early mobility interventions are excluded by strict interpretation of current guidelines.
    Conclusions: Risk vs benefit of implementing early mobility interventions in individuals with critical illness due to COVID-19 can be mitigated through coordinated efforts of interdisciplinary teams to promote shared decision-making through therapeutic alliances with patients and their families. Clinicians must clearly define the goals of care, understand the limitations of monitoring equipment in the intensive care unit, prepare to titrate levels of oxygen based on an individual's physiologic response to mobility interventions, and help individuals maintain external goal-directed focus of attention to optimize outcomes of early mobility interventions.

    Abstract The Postnatal Depression Intervention Program is a screening program looking at postnatal depression in newly delivered mothers at KK Hospital in Singapore. The primary objective of the paper is to evaluate the outcomes based on changes in scores from baseline to end of intervention with reference to depressive symptoms, using the Edinburgh Postnatal Depression Scale and overall functioning based on the Global Assessment of Functioning. These changes were found to be statistically significant. We also described the demographics of the 10-year cohort with majority being married, Chinese, aged between 25 and 34 years old and received at least tertiary education.

    Abstract Background: Children who live with a parent with a substance use disorder (SUD) are more likely to experience adverse health outcomes, including mental health disorders. We assessed whether residing with anyone who used substances was associated with children's anxiety and/or depression, and whether these associations differed by the children's age or sex.
    Methods: We analyzed nationally representative cross-sectional data from the 2019 National Health Interview Survey (n = 6642). The associations between ever residing with someone who used substances and caregiver-reported children's anxiety and depression frequency (never/a few times a year/monthly/weekly/daily) were estimated using multinomial logistic regression models, adjusted for children's age, children's sex, children's race/ethnicity, annual household income, and highest educational attainment by an adult in the household. We assessed whether the associations differed based on the children's age (5-11/12-17 years) or sex.
    Results: Children who had resided with someone who used substances were more likely to be reported by their caregiver as having daily anxiety (risk ratio (RR) = 2.84; 95% confidence interval (CI) = 2.04, 3.95; referent = never anxious) and daily depression (RR = 3.35; 95% CI = 1.98, 5.67; referent = never depressed). Associations with more frequent anxiety were stronger among adolescents than younger children. Associations between residing with someone who used substances and depression frequency differed based on children's age and sex.
    Conclusions: Our results suggest that residing with someone who used substances is associated with children's anxiety and depression. Our findings can help inform screening and treatment efforts for anxiety and depression among children, as well as for the person using substances.
