    Abstract In this study the spectrophotometric behaviour of gelatin-based hydrogels, in the presence and absence of dyes, was studied. The aim was to formulate equivalent-tissue phantoms to be used as 3D-dosimeter, suitable for Optical Computed Tomography (OCT). The hydrogels show good transparency and good stability of baseline optical density and, in the presence of dye, the response of optical density as a function of concentration was higher than in aqueous solution. The hydrogels were formulated in order to reduce the diffusion of the image of the irradiated field over time and to have stable fantoms as a function of time. To this purpose, the diffusion coefficients of two dyes, Bromophenol Blue (BPB) and Bromocresol Green (BCG), were determined as a function of the hydrogel chemical composition. As comparison, in some experiments Xylenol Orange (XO) was used. In particular, the presence of sucrose, as thickener, can reduce to almost half BPB mobility. In conclusion, it was shown that optical properties and controlled dye diffusion in gelatin-based hydrogels could allow using them as 3D-dosimeter for optical detection.

    Abstract The INTRABEAM Carl Zeiss Surgical system (Oberkochen, Germany) is a miniature accelerator producing low energy photons (50 keV maximum). The published dosimetric characterization of the INTRABEAM was based on detectors (radiochromic films or ionization chambers) not allowing measuring the absorbed dose in the first millimeters of the irradiated medium, where the dose is actually prescribed. This study aims at determining with Magnetic Resonance Imaging (MRI) the sensitivity of a paramagnetic gel in order to measure the dose deposit produced with the INTRABEAM from 0 to 20 mm. Although spherical applicators are mostly used with the INTRABEAM system for breast applications, this study focuses on surface applicators that are of interest for cutaneous carcinomas. The irradiations at 12 different dose levels (between 2 Gy and 50 Gy at the gel surface) were performed with the INTRABEAM and a 4 cm surface applicator. The gel used in this study is a new « sensitive » material. In order to compare gel sensitivity at low energy with high energy, gels were irradiated by an 18 MV photon beam produced by a Varian Clinac 2100 CD. T2 weighted multi echo MRI sequences were performed with 16 echo times. The T2 signal versus echo times was fitted with a mono-exponential function with 95% confidence interval. The calibration curve determined at low energy is a linear function (r2 = 0.9893) with a sensitivity of 0.0381 s-1.Gy-1, a similar linear function was obtained at high energy (0.0372 s-1.Gy-1 with r2 = 0.9662). The calibration curve at low energy was used to draw isodose maps from the MR images. The PDD (Percent Depth Dose) determined in the gel is within 5%-1mm of the ionization chamber PDD except for one point. The dosimetric sensitivity of this new paramagnetic ferrous gel was determined with MRI measurements. It allowed measuring the dose distribution specifically in the first millimeters for an irradiation with the INTRABEAM miniature accelerator equipped with a surface applicator.

    Abstract Five populations of tetraploid Acacia heterophylla, endemic from La Réunion island, were compared together and with their Australian diploid relative A. melanoxylon for cytogenetic and DNA characteristics. A. melanoxylon (2n = 26) had 1·59 pg nuclear DNA; A. heterophylla (2n = 4x = 52) had double this value (3·19 pg), and there was no difference between populations within species. Both species had 39 % GC. Interchromosome connections were evident at metaphase and mitotic irregularities at anaphase were twice as frequent in A. heterophylla as in A. melanoxylon, again with no difference between populations within species. These results argue for a recent autotetraploid origin of A. heterophylla from A. melanoxylon. Yet, fluorochrome banding showed that in some A. heterophylla populations, GC-rich bands had slightly changed from the supposed ancestral pattern, probably by means of translocations involving parts of nuclear organizer areas. No clear relation was found between banding patterns and ecological factors.

    Abstract Background: Unlike other strains, spontaneously type 1 non-obese diabetic (NOD) experience transient hyperinsulinemia after weaning. The same applies for NOD/SCID mice, which lack functional lymphocytes, and unlike NOD mice, do not develop insulitis and diabetes like NOD mice.
    Aims: Given that beta-cell stimulation is a natural feature of gestation, we hypothesized that glucose homeostasis is disturbed in gestate pre-diabetic NOD and non-diabetic NOD/SCID mice, which may accelerate the onset of diabetes and increase diabetes prevalence.
    Methods: During gestation and postpartum, mice were analyzed under basal feed conditions followed by glucose injection (1 g/kg, i.p.) after overnight fast, using glucose tolerance test (GTT). Glycemia, corticosteronemia, blood and pancreatic insulin, glucagon levels, islet size, and islet morphology were evaluated. Glycemia and mortality were assessed after successive gestations in NOD mice mated for the first time at 2 different ages.
    Results: 1. Basal glucagonemia rose markedly in first-gestation fed NOD mice. 2. beta-cell hyperactivity was present earlier in first-gestation non-diabetic fasted NOD and NOD/SCID mice than in age-matched C57BL/6 mice, assessed by increased insulin/glucose ratio after GTT. 3. Overnight fasting increased corticosteronemia rapidly and sharply in pre-diabetic gestate NOD and NOD/SCID mice. 4. Islet size increased in non-diabetic gestate NOD mice compared with C57BL/6 mice. 5. Successive gestations accelerated diabetes onset, and contributed to increased mortality in NOD mice.
    Conclusions: First-gestation pre-diabetic NOD and non-diabetic NOD/SCID mice exhibited beta-cell hyperactivity and deregulation of glucagon and/or corticosterone secretion. This amplified normally occurring insulin resistance, further exhausted maternal beta-cells, and accelerated diabetes in NOD mice.
