    Abstract Cistanche tubulosa (Schenk) R. Wight is a valuable herbal medicine in China. The study aimed to explore the potential mechanisms of C. tubulosa on antioxidant activity using spectrum-effect relationship and network pharmacology and the possibilities of utilizing herbal dregs. In this work, different extracts of C. tubulosa, including herbal materials, water extracts, and herbal residues, were evaluated using high-performance liquid chromatography (HPLC) technology. In addition, the antioxidant activities were estimated in vitro, including 2, 2-diphenyl-1-picrylhydrazyl; superoxide anion; and hydroxyl radical scavenging assays. The spectrum-effect relationships between the HPLC fingerprints and the biological capabilities were analyzed via partial least squares regression, bivariate correlation analysis, and redundancy analysis. Furthermore, network pharmacology was used to predict potential mechanisms of C. tubulosa in the treatment of antioxidant-related diseases. According to the results, eleven common peaks were shared by different extracts. Geniposidic acid, echinacoside, verbascoside, tubuloside A, and isoacteoside were quantified and compared among different forms of C. tubulosa. The spectrum-effect relationship study indicated that peak A6 might be the most decisive component among the three forms. Based on network pharmacology, there were 159 target genes shared by active components and antioxidant-related diseases. Targets related to antioxidant activity and relevant pathways were discussed. Our results provide a theoretical basis for recycling the herbal residues and the potential mechanisms of C. tubulosa in the treatment of antioxidant-related diseases.

    Abstract Objective: To explore the effect of Sirt1 on the function of endothelial progenitor cells (EPCs) in rats with chronic obstructive pulmonary disease (COPD). Methods: A rat COPD model was established via smoking and endotoxin administration for three months. The peripheral circulating EPCs were isolated by gradient centrifugation, and their functions, cell cycle distribution, apoptosis, and Sirt1 expression were examined. The function changes of EPCs in the presence or absence of Sirt1 agonist and inhibitor were estimated; meanwhile, the expressions of Sirt1, FOXO3a, NF-κB, and p53 were also evaluated. Results: The proliferation, adhesion, and migration of EPCs decreased while the apoptosis rate was increased in the COPD rats. The expression of Sirt1 protein in EPCs of the COPD group was significantly lower than that in the control group (P<0.01). The overexpression of the Sirt1 gene using a gene transfection technique or Sirt1 agonists (SRT1720) improved the proliferation, migration, and adhesion, and decreased the apoptosis of EPC. However, Sirt1 inhibitor (EX527) decreased EPC functions in the COPD group. The effect of Sirt1 expression on EPC function may be related to reduction of FOXO3a and increase of NF-κB and p53 activity. Conclusions: Increased expression of Sirt1 can improve the proliferation and migration of EPCs and reduce their apoptosis in COPD rats. This change may be related to FOXO3a, NF-κB, and p53 signaling pathways.

    Abstract In this study, a water‐air two‐phase flow model was employed to investigate the formation, extension, and dissipation of groundwater ridging induced by recharge events in a hypothetical hillslope‐riparian zone, considering interactions between the liquid and gas phases in soil voids. The simulation results show that, after a rain begins, the groundwater table near the stream is elevated instantaneously and significantly, thereby generating a pressure gradient driving water toward both the stream (the discharge of groundwater to the stream) and upslope (the extension of groundwater ridging into upslope). Meanwhile, the airflow upslope triggered by the advancing wetting front moves downward gradually. Therefore, the extension of groundwater ridging into upslope and the downward airflow interact within a certain region. After the rain stops, groundwater ridging near the stream declines quickly while the airflow in the lower part of upslope is still moving into the hillslope. Thus, the airflow upslope mitigates the dissipation of groundwater ridging. Additionally, the development of groundwater ridging under different conditions, including rain intensity, intrinsic permeability, capillary fringe height, and initial groundwater table, was analyzed. Changes in intrinsic permeability affect the magnitude of groundwater ridging near the stream, as well as the downward speed of airflow, thereby generating highly complex responses. The capillary fringe is not a controlling factor but an influence factor on the formation of groundwater ridging, which is mainly related to the antecedent moisture. It was demonstrated that groundwater ridging also occurs where an unsaturated zone occurs above the capillary fringe with a subsurface lateral flow.

    Abstract Tao-He-Cheng-Qi decoction (THCQ) is an effective traditional Chinese medicine used to treat intracerebral hemorrhage (ICH). This study was performed to investigate the possible neuroprotective effect of THCQ decoction on secondary brain damage in rats with intracerebral hemorrhage and to elucidate the potential mechanism based on a metabolomics approach. Sprague-Dawley (SD) rats were randomly divided into five groups: the sham group, collagenase-induced ICH model group, THCQ low-dose (THCQ-L)-treated group, THCQ moderate-dose (THCQ-M)-treated group and THCQ high-dose (THCQ-H)-treated group. Following 3 days of treatment, behavioral changes and histopathological lesions in the brain were estimated. Untargeted metabolomics analysis with multivariate statistics was performed by using ultrahigh-performance liquid chromatography–mass spectrometry (UPLC-Q-Exactive Orbitrap MS). THCQ treatment at two dosages (5.64 and 11.27 g/kg·d) remarkably improved behavior (p < 0.05), brain water content (BMC) and hemorheology (p < 0.05) and improved brain nerve tissue pathology and inflammatory infiltration in ICH rats. Moreover, a metabolomic analysis demonstrated that the serum metabolic profiles of ICH patients were significantly different between the sham group and the ICH-induced model group. Twenty-seven biomarkers were identified that potentially predict the clinical benefits of THCQ decoction. Of these, 4 biomarkers were found to be THCQ-H group-specific, while others were shared between two clusters. These metabolites are mainly involved in amino acid metabolism and glutamate-mediated cell excitotoxicity, lipid metabolism-mediated oxidative stress, and mitochondrial dysfunction caused by energy metabolism disorders. In addition, a correlation analysis showed that the behavioral scores, brain water content and hemorheology were correlated with levels of serum metabolites derived from amino acid and lipid metabolism.     Abstract The methylerythritol phosphate pathway is responsible for the biosynthesis of terpenoids, the largest class of secondary metabolites. Although the structures and functions of the proteins involved in this pathway have been well studied in Bacillus subtilis, only a few studies have reported the transcriptional profile of the genes involved. Therefore, we analyzed methylerythritol phosphate pathway genes in the genome of B. subtilis 916, which has been developed as a biological control agent against some rice diseases in China. Our results showed that methylerythritol phosphate pathway genes were distributed throughout the genome of this strain. These genes were transcribed during both the exponential and stationary phases. We further confirmed the transcription units of dxs, dxr, ispD, ispF, ipK, ispG, ispH, idi, and ispA in B. subtilis 916 through reverse transcription-PCR analyses; the results showed that these nine genes were located in seven different operons. The transcript start sites of the seven different operons were determined by 5’-rapid amplification of cDNA ends-PCR. Thus, our study provides a molecular basis at the transcriptional level for investigating homoterpene synthesis in the methylerythritol phosphate pathway of B. subtilis 916.

    Abstract An investigation of groundwater table fluctuations induced by rainfall should consider interactions between the liquid and gas phases in soils. In this study, a water‐air two‐phase flow model was initially verified by simulating an infiltration experiment. It was then employed to model the interactions between liquid and gas phases regarding actions of airflow on the groundwater table and the fluctuations of the phreatic level and water level in the well induced by rainfall. The effects of airflo7w caused by rainfall on phreatic level fluctuations were also studied quantitatively by comparing the results obtained using the proposed model with those obtained from a water single‐phase flow model. The simulation results show that in addition to actual recharge, compressed airflow in unsaturated zones causes the phreatic level to increase, but the rise in the phreatic level is lower than that in the pore‐air pressure head in unsaturated zones due to the mitigation of capillary fringe. The existence of airflow enhances the phreatic level rise during and after rainfall. In addition, the water level in the well, pushed by the phreatic level fluctuations, varies similarly to the phreatic level, but it experiences somewhat delayed and slightly attenuated. The Lisse effect precisely reflects the phreatic level fluctuations before actual recharge. Furthermore, the fluctuations in the phreatic level and water level in the well and the contributions of airflow to phreatic level fluctuations are affected by many factors: rain intensity, initial moisture, overlying aquitard, groundwater table depths, and screen depths of the well.

    Abstract Bacillus thuringiensis subspecies morrisoni strain HD12, whose genome harbors multiple insecticidal protein-encoding genes, includes eight cry genes, as indicated by genome sequencing. This strain produces crystals that are toxic to lepidopteran species. These crystal inclusions were purified by sucrose gradients and sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE), followed by liquid chromatography-mass spectrometry, and found to contain five proteins (Cry1Da, Cry1Ae, Cry1Bb, Cry1Fb, and Cry1Ja). The transcriptional activities of the cry1Da, cry1Ae, cry1Bb, cry1Fb, and cry1Ja promoters indicated that transcription of cry1Da is controlled by SigE; however, the other four cry genes were found to be controlled by both SigE and SigK. The activities of the cry1Ja and cry1Fb promoters were the strongest among the five genes studied. These promoters were therefore used to direct the expression of the Cry1Ac- and Cry2Ab-encoding genes concurrently in a single strain. Our findings indicate that promoters with the same transcriptional profile may be used to direct the expression of different cry genes in one strain. Our results are expected to be valuable for the construction of strains with efficient expression of multiple cry genes in order to overcome current limitations associated with the application of B. thuringiensis-based insecticides.

    Abstract We aimed to determine if electrical signals generated by soil microorganisms in microbial fuel cells (MFCs) can accurately respond to Cd2+ concentrations and to evaluate the linear detection range of the method. MFCs were packed with soil spiked with 10 to 400mgkg−1 Cd2+ and operated for 37h at constant temperature (30°C). The generated coulomb (C) and start-up time (ST) of the MFCs were calculated as electrical signals. The dehydrogenase activity (DHA) of the Cd2+-spiked soil was determined as a reference method. After MFC operation, the diversity of anodic bacteria was studied by using PCR-DGGE and the sequencing of 16S rRNA genes. The results showed that with increasing Cd2+ concentration, C decreased while ST increased significantly. In the control, C was 30.86±3.84C and ST was 14.39±1.44h; these values decreased to 8.29±5.08C and 30.39±2.87h, respectively, for the treatment with 400mgkg−1 Cd2+. Both C (r2=0.943, P<0.01) and ST (r2=0.937, P<0.01) were linearly related to Cd2+ at concentrations ranging from 10 to 100mgkg−1 and were in agreement with DHA. Cadmium stress significantly decreased the Shannon diversity and changed the community structure of anodic bacteria. Phylogenetic analysis of DGGE bands showed that the anodic bacteria were mainly affiliated with Bacillus, Comamonas, Clostridium and Enterobacter. We propose that electrical signals can be used to evaluate the toxicity of Cd2+ to soil microorganisms at concentrations below 100mgkg−1.

    Abstract Abstract Thioredoxins (Trxs) play an important role in defending against oxidative stress and keeping disulfide bonding correct to maintain protein function. Edwardsiella piscicida, a severe fish pathogen, has been shown to encode several thioredoxins including TrxA, TrxC, and TrxH, but their biological roles remain unknown. In this study, we characterized TrxH of E. piscicida (named TrxHEp) and examined its expression and function. TrxHEp is composed of 125 residues and possesses typical thioredoxin H motifs. Expression of trxH Ep was upregulated under conditions of oxidative stress, iron starvation, low pH, and during infection of host cells. trxH Ep expression was also regulated by ferric uptake regulator (Fur), an important global regulatory of E. piscicida. Compared to the wild type TX01, a markerless trxH Ep in-frame mutant strain TX01∆trxH exhibited markedly compromised tolerance of the pathogen to hydrogen peroxide, acid stress, and iron deficiency. Deletion of trxH Ep significantly retarded bacterial biofilm growth and decreased resistance against serum killing. Pathogenicity analysis shows that the inactivation of trxH Ep significantly impaired the ability of E. piscicida to invade host cells, reproduce in macrophages, and infect host tissues. Introduction of a trans-expressed trxH gene restored the lost virulence of TX01∆trxH. There is likely to be a complex relationship of functional complementation or expression regulation between TrxH and another two thioredoxins, TrxA and TrxC, of E. piscicida. This is the first functional report of TrxH in fish pathogens, and the findings suggest that TrxHEp is essential for coping with adverse circumstances and contributes to host infection of E. piscicida.

    Abstract Background: The accurate measurement of the femoral anteversion (FA) angle is always a topic of much debate in the orthopedic surgery and radiology research. We aimed to explore a new FA measurement method to acquire accurate results without radiation damage using piglet model. Methods: A total of thirty piglets were assigned to two groups based on the age. Bilateral femora were imaged with 3.0-T magnetic resonance (MR) and 64-slice computed tomography (CT) examinations on all piglets. FA was measured on MR-three-dimensional (3D) postprocessing software with a four-step method: initial validation of the femoral condylar axis, validation of the condylar plane, validation of the femoral neck axis, and line-plane angle measurement of FA. After MR and CT examinations, all piglets were sacrificed and their degree of FA was measured using their excised, dried femora. MR, CT, and dried-femur measurement results were analyzed statistically; MR and CT measurements were compared for accuracy against each other and against the gold standard dried femur measurement. Results: In both groups, the mean FA value measured by MR was lower than that measured by CT. A statistically significant difference was observed between CT- and dried-femur measurements but not between MR- and dried-femur measurements. A higher correlation (0.783 vs. 0.408) and a higher consistency (0.863 vs. 0.578) with dried-femur measurement results were seen for MR measurements than CT measurements in the 1-week age group. However, in the 8-week age group, similar correlations (0.707 vs. 0.669) and consistencies (0.864 vs. 0.821) were observed. Conclusions: Noninvasive MR-3D-Cube reconstruction was able to accurately measure FA in piglets. Particularly in the 1-week age group with a larger proportion of cartilaginous structures, the correlation and consistency between MR- and dried-femur measurement results were higher than those between CT- and dried-femur measurements, suggesting that MR may be a new useful examination tool for FA-related diseases in children.
