    Abstract Randomized controlled trials are essential for evaluating clinical interventions; however, selective reporting and publication bias in medical journals have undermined the integrity of the clinical evidence system. ClinicalTrials.gov serves as a valuable and complementary repository, yet synthesizing information from it remains challenging. This study introduces a curated dataset that extends beyond the traditional PICO framework. It links efficacy with safety results at the experimental arm group level within each trial, and connects them across all trials through a knowledge graph. This novel representation effectively bridges the gap between generally described searchable information and specifically detailed yet underutilized reported results, and promotes a dual-faceted understanding of interventional effects. Adhering to the "calculate once, use many times" principle, the structured dataset will enhance the reuse and interpretation of ClinicalTrials.gov results data. It aims to facilitate more systematic evidence synthesis and health technology assessment, by incorporating both positive and negative results, distinguishing biomarkers, patient-reported outcomes, and clinical endpoints, while also balancing both efficacy and safety outcomes for a given medical intervention.

    Abstract Atherosclerotic cardiovascular disease (ASCVD) consistently ranks as the primary mortality factor among diabetic people. A thorough comprehension of the pathophysiological routes and processes activated by atherosclerosis (AS) caused by diabetes mellitus (DM), together with the recognition of new contributing factors, could lead to the discovery of crucial biomarkers and the development of innovative drugs against atherosclerosis. Selenoprotein S (SELENOS) has been implicated in the pathology and progression of numerous conditions, including diabetes, dyslipidemia, obesity, and insulin resistance (IR)-all recognized contributors to endothelial dysfunction (ED), a precursor event to diabetes-induced AS. Hepatic-specific deletion of SELENOS accelerated the onset and progression of obesity, impaired glucose tolerance and insulin sensitivity, and increased hepatic triglycerides (TG) and diacylglycerol (DAG) accumulation; SELENOS expression in subcutaneous and omental adipose tissue was elevated in obese human subjects, and act as a positive regulator for adipogenesis in 3T3-L1 preadipocytes; knockdown of SELENOS in Min6 β-cells induced β-cell apoptosis and reduced cell proliferation. SELENOS also participates in the early stages of AS, notably by enhancing endothelial function, curbing the expression of adhesion molecules, and lessening leukocyte recruitment-actions that collectively reduce the formation of foam cells. Furthermore, SELENOS forestalls the apoptosis of vascular smooth muscle cells (VSMCs) and macrophages, mitigates vascular calcification, and alleviates inflammation in macrophages and CD4

    Abstract The generation of occlusive thrombi in stenotic arteries involves the rapid deposition of millions of circulating platelets under high shear flow. The process is mediated by the formation of molecular bonds of several distinct types between platelets; the bonds capture the moving platelets and stabilize the growing thrombi under flow. We investigated the mechanisms behind occlusive thrombosis in arteries with a two-phase continuum model. The model explicitly tracks the formation and rupture of the two types of interplatelet bonds, the rates of which are coupled with the local flow conditions. The motion of platelets in the thrombi results from competition between the viscoelastic forces generated by the interplatelet bonds and the fluid drag. Our simulation results indicate that stable occlusive thrombi form only under specific combinations for the ranges of model parameters such as rates of bond formation and rupture, platelet activation time, and number of bonds required for platelet attachment.

    Abstract Aim: It is essential for health researchers to have a systematic understanding of third-party variables that influence both the exposure and outcome under investigation, as shown by a directed acyclic graph (DAG). The traditional construction of DAGs through literature review and expert knowledge often needs to be more systematic and consistent, leading to potential biases. We try to introduce an automatic approach to building network linking variables of interest.
    Methods: Large-scale text mining from medical literature was utilized to construct a conceptual network based on the Semantic MEDLINE Database (SemMedDB). SemMedDB is a PubMed-scale repository of the "concept-relation-concept" triple format. Relations between concepts are categorized as Excitatory, Inhibitory, or General.
    Results: To facilitate the use of large-scale triple sets in SemMedDB, we have developed a computable biomedical knowledge (CBK) system (https://cbk.bjmu.edu.cn/), a website that enables direct retrieval of related publications and their corresponding triples without the necessity of writing SQL statements. Three case studies were elaborated to demonstrate the applications of the CBK system.
    Conclusions: The CBK system is openly available and user-friendly for rapidly capturing a set of influencing factors for a phenotype and building candidate DAGs between exposure-outcome variables. It could be a valuable tool to reduce the exploration time in considering relationships between variables, and constructing a DAG. A reliable and standardized DAG could significantly improve the design and interpretation of observational health research.

    Abstract In this research, an empirical investigation is made to analyze the operation mode, effect, and feedback of digital health communication, which is based on China’s new media, and the result shows that: firstly, although the common new media’s means of communication are diverse and the profit mode is varied, traditional means such as articles and videos still prevail. Yet, new media has a heavy dependence on the profit mode based on network advertising and online shopping, while the operation mode profiting by outputting high-quality content requires a further improvement; secondly, on the whole, no obvious difference is found in the effect of communication between people of different ages and genders, but the main difference is observed between people having health problems and those not. If an in-depth analysis is respectively conducted for different kinds of new media, some specific differences can be found in the communication effect; thirdly, although many audiences may concern about the risks brought by new media’s credibility and commercial factors, commercial operation in a standardized way is acceptable to a large number of audiences. Therefore, effort shall be made to propel the development of new media-based health communication from such perspectives as optimizing the operation mode, deepening the user research and regulating the commercial operation, etc.

    Abstract Health is a representative domain data-driven research since health research data are growingly generated at a massive scale. There is an intuitive logic that the degree to which disease burden and the number of data resources align. In order to figure out disease-specific data sharing and reuse level, we took the number of data records and their citations in the scientific literature in the Data Citation Index platform as approximate indicators. The results indicated that only a small percentage (7.5%) of health data records had received documented citations by scientific publications. We find the level of data sharing and reuse varies across diseases. Our study suggested that the more socioeconomic burden and the more research funding, the more likely scientific data for diseases will be produced and made available. But such a correlation could not be observed for the activity of data reuse. Secondary reuse of scientific data is a complex behavior.

    Abstract To meet the demand for rapid bacterial detection in clinical practice, this study proposed a joint determination model based on spectral database matching combined with a deep learning model for the determination of positive-negative bacterial infection in directly smeared urine samples. Based on a dataset of 8124 urine samples, a standard hyperspectral database of common bacteria and impurities was established. This database, combined with an automated single-target extraction, was used to perform spectral matching for single bacterial targets in directly smeared data. To address the multi-scale features and the need for the rapid analysis of directly smeared data, a multi-scale buffered convolutional neural network, MBNet, was introduced, which included three convolutional combination units and four buffer units to extract the spectral features of directly smeared data from different dimensions. The focus was on studying the differences in spectral features between positive and negative bacterial infection, as well as the temporal correlation between positive-negative determination and short-term cultivation. The experimental results demonstrate that the joint determination model achieved an accuracy of 97.29%, a Positive Predictive Value (PPV) of 97.17%, and a Negative Predictive Value (NPV) of 97.60% in the directly smeared urine dataset. This result outperformed the single MBNet model, indicating the effectiveness of the multi-scale buffered architecture for global and large-scale features of directly smeared data, as well as the high sensitivity of spectral database matching for single bacterial targets. The rapid determination solution of the whole process, which combines directly smeared sample preparation, joint determination model, and software analysis integration, can provide a preliminary report of bacterial infection within 10 min, and it is expected to become a powerful supplement to the existing technologies of rapid bacterial detection.

    Abstract Methanol is an important chemical product with wide application. Due to the high pollutant emission of coal-to-methanol route, feeding mixed raw material of biomass and coal is seen as a promising measure of improvement. More than just focusing on co-gasification unit, this study performs a full process simulation from co-gasification to methanol and cogeneration, aiming at revealing the impact of biomass on process operation and optimizing the key operating parameters. The simulation results imply that coal and biomass co-gasification can improve the utilization of raw materials with more effective gas produced. As an example, the process based on the co-gasification of bituminous coal and straw is specially studied and telling the best operating situation of the whole process. The gasification agent and the critical effects of crude syngas split ratio (SR) and unreacted gas recycle ratio (CR) on the net methanol yield are fully discussed to gain insight into the process. The simulation results show that when the ratio of biomass to coal is 1:1, gasification agent is water steam, SR = 0.36, CR = 0.8, the net methanol production can reach a maximum of 7022.32 kg/h, 241.51% higher than that of merely using coal, producing 37.68% less CO₂ than merely using coal.
