    Abstract The character of metabolism in the precalving period is one of the factors determining the reproductive potential of dairy cattle. The purpose of the present investigation was a comparative analysis of the activity of a number of metabolic enzymes participating in protein and carbohydrate metabolism in Black Pied heifer blood at different stages of pregnancy in connection with further reproduction indices. Regardless of the length of the subsequent service period, the activity of aspartate aminotransferase (AST) in blood in the late stage of pregnancy was 1.7 times lower than in the middle stage. The activity of alanine aminotransferase (ALT) in blood also decreased in the late stage; however, this decrease was expressed more strongly in animals with a service period length greater than 150 days (by 1.8 times) than in animals with a service period length less than 150 days (by 1.3 times). In this case, the AST/ALT ratio decreased with progressing pregnancy in heifers with increased reproductive capacity (p < 0.05) and didn’t change in heifers with reduced reproductive capacity. In the blood of animals with a longer service period, the AST/ALT ratio was 1.3–1.7 times higher (p < 0.05) than in blood of animals with a shorter period. No significant variability was revealed in the activity of alkaline phosphatase and lactate dehydrogenase depending on progressing stages of pregnancy and on the reproductive potential of heifers.

    Abstract Cyclic AMP is one of the key regulators of mammalian meiosis. In the present work, realization pathways of the previously revealed modulating influence of prolactin (PRL) on the cAMP-dependent mechanisms of meiosis regulation in bovine oocytes were studied. A comparative investigation of individual and combined effects of PRL (50 ng/ml) and an activator of adenylate cyclase forskolin (FK. 20 microM) on the meiotic reinitiation and nuclear maturation completion in cumulus-enclosed and cumulus-free oocytes was performed. It has been shown that the pattern of PRL influence on the meiotic resumption in oocytes devoid of cumulus cells depends on the presence of FK in the culture medium. Furthermore, realization of this influence is not associated with the activation of cytoplasmic isoforms of protein kinase C. It has been also found that PRL inhibits the retarding action of FK on the completion of oocyte nuclear maturation in both the presence and absence of cumulus cells. The findings suggest that PRL may modulate the functioning of the cAMP-dependent mechanism of meiosis regulation by the direct action on bovine oocytes, with realization of the action being independent of the metabolic coupling of oocytes with cumulus cells.

    Abstract Maintenance of the phenotype of spermatogenic stem cells in vitro depends on the microenvironment and a number of factors. Culturing boar type A1–A4 spermatogonia on the STO feeder layer and addition to the growth medium of a conditioned medium containing DIA/LIF led over the course of 3–4 weeks to the formation of colonies which had morphology similar to embryonic stem cells and were distinguished by a high alkaline phosphatase activity.
