    Abstract Abstract Pfs48/45 is a leading antigen candidate for a transmission blocking (TB) vaccine. However, efforts to produce affordable, safe and correctly folded full-length Pfs48/45 using different protein expression systems have not produced an antigen with satisfactory TB activity. Pfs48/45 has 16 cysteines involved in disulfide bond formation, and the correct formation is critical for proper folding and induction of TB antibodies. Moreover, Pfs48⁄45 is not a glycoprotein in the native hosts, but contains potential glycosylation sites, which are aberrantly glycosylated during expression in eukaryotic systems. Here, we demonstrate for the first time that full length, Endo H in vivo enzymatic deglycosylated Pfs48/45 antigen is produced at a high level in plants and is structurally stable at elevated temperatures. Sera from mice immunized with this antigen showed strong inhibition in SMFA. Thus, Endo H in vivo enzymatic deglycosylated Pfs48/45 is a promising candidate for the development of an affordable TB vaccine, which may have the potential to save millions.

    Abstract Modelling is carried out for eleven major weeds in cucumber to develop estimated models for weed emergence time. Weed species were grouped according to their emergence patterns. Amaranthus retroflexus, Chenopodium album, Heliotropium europaeum, Polygonum aviculare and Solanum nigrum were early emerging, Convolvulus arvensis, Cyperus rotundus, Cynodon dactylon, Portulaca oleracea and Sorghum halepense were season long emerging Tribulus terrestris was the late emerging weed species. Different non-linear growth curves (Chapman-Richard, Weibull, logistic, Gompertz and cubic spline) fitted to the data of cumulative percent emergence for the different species and years. Cubic spline seemed the best model for many species.

    Abstract The effect of varying potassium (0-control, 5, 10, 18 and 28 mM as potassium nitrate) and calcium (0-control, 5, 10, 18 and 28 mM as calcium nitrate) supply on growth of NaCl stressed (0-control, 100 and 150 mM) mature embryos of cucumber ( Cucumis sativus L.) in vitro condition. NaCl caused decrease root (rooting percent, root number, root length and root fresh weight) and shoot (shoot length, shoot FW, leaf FW, whole plant FW) growth. Root/shoot length index and root/plant FW index was generally lower in the 100 and 150 mM NaCl concentrations than control (absence of NaCl). There was an increase in callus induction of NaCl treated cucumber embryos. Callus induction was decreased by K + and Ca 2+ applications at both NaCl concentrations In general, root and shoot growth of cucumber embryos was improved by supplemental potassium (10 mM potassium nitrate) and calcium (20 mM calcium nitrate).

    Abstract Red hot pepper production for spices is very important for Kahramanmaras City located in Eastern Mediterranean Region of Turkey as around the world. However, there are major production problems. One of the biggest problems is absence of improved variety yet inside of cultivated peppers and low quality of seed material in region. In experiment, 115 red hot pepper accessions for spice production originally collected from region were evaluated based on quantitative and qualitative characters. When accessions were evaluated based on each one character, successful of accessions varied. Therefore accessions were evaluated with weighed grade method and 17, 122, 43 and 140 (elongate), 52, 135 and 51 (triangular), 118 (campanulate) and 123, 26 and 65 (blocky) accessions were selected as use in breeding research of red hot spice pepper in Kahramanmaras.

    Abstract The effects of NaCl concentration (0, 50, 100 and 150 mM NaCl) on the growth and development of pepper varieties, Demre, �etinel 150 and Ilica 256, were investigated for germination and seedling growth. The parameters measured were germination (speed and germination rate), radicle length, hypocotyl length, radicle fresh weight, hypocotyl fresh weight and the ratio of hypocotyls to radicle fresh weight in the germination stage. High NaCl had a negative effect on plant growth and development. In the seedling stage, the negative effects were relative growth rate for root and shoot length; Relative growth rate for fresh and dry weights of roots, shoots and whole plant; fresh and dry weight of shoot to root indices, relative growth rate for leaf number and area. Leaf content of K + and Na + and K + /Na + were negatively affected by increased NaCl concentrations. It was found that the Na + concentration increased whereas the K + and K + /Na + values decreased in leaves. Ilica 256 showed better NaCl tolerance in the germination stage and Demre was the most tolerant at the seedling stage than other varieties. This research demonstrated different NaCl tolerances for the varieties tested based on growth and development ability at germination and seedling development

    Abstract The three transcription factors, PDX1, NGN3 and MAFA, are very important in pancreatic development. Overexpression of these three factors can reprogram both pancreatic exocrine cells and SOX9-positive cells of the liver into cells resembling pancreatic beta cells. In this study we investigate whether other cell types can be reprogrammed. Eight cell types are compared and the results are consistent with the idea that reprogramming occurs to a greater degree for developmentally related cells (pancreas, liver) than for other types, such as fibroblasts. Using a line of mouse hepatocyte-derived cells we screened 13 compounds for the ability to increase the yield of reprogrammed cells. Three are active and when used in combination they can increase the yield of insulin-immunopositive cells by a factor of six. These results should contribute to the eventual ability to develop a new cure for diabetes based on the ability to reprogram other cells in the body to a beta cell phenotype.
