    Abstract Las metodologías de enseñanza en las ciencias han sido una permanente preocupación en las investigaciones en enseñanza de la Física, y a la fecha se siguen proponiendo alternativas que puedan mejorar la calidad de los aprendizajes. La presente investigación es una propuesta de enseñanza de los Fundamentos de la Teoría Cuántica, para estudiante del profesorado en Física, que permita mejorar rendimiento académico, promover aprendizaje de calidad, y desarrollar competencias genéricas. La propuesta se fundamenta epistemológicamente en el constructivismo, además, con aportes de Popper, Khun, y Lakatos. Se sustenta en las teorías del aprendizaje significativo de Ausubel, la teoría sociocultural de Vygostky, los Campos conceptuales de Vergnaud, los aportes de Novak y Gowin. La investigación se resuelve mediante una metodología de investigación fundamentalmente cuantitativa con algunos elementos de tipo cualitativo, además, es de tipo cuasi-experimental, con grupo control y experimental. Su estudio es descriptivo, comparativo e interpretativo. Sus principales resultados indican un aumento importante en los rendimientos académicos, del 55% al 67%, los aprendizajes obtenidos son significativos. En lo que es aprendizaje, sus resultados nos indican que mayoritariamente consiguen aprendizaje significativo en los fundamentos que dan origen a mecánica cuántica. Las competencias: cognitivas, metodológicas y sociales superan el 70% de logro. El grado de satisfacción de la propuesta es sobre el 85% por parte de los estudiantes.

    Abstract Water electrolysis to obtain hydrogen in combination with intermittent renewable energy resources is an emerging sustainable alternative to fossil fuels. Among the available electrolyzer technologies, anion exchange membrane water electrolysis (AEMWE) has been paid much attention because of its advantageous behavior compared to other more traditional approaches such as solid oxide electrolyzer cells, and alkaline or proton exchange membrane water electrolyzers. Recently, very promising results have been obtained in the AEMWE technology. This review paper is focused on recent advances in membrane electrode assembly components, paying particular attention to the preparation methods for catalyst coated on gas diffusion layers, which has not been previously reported in the literature for this type of electrolyzers. The most successful methodologies utilized for the preparation of catalysts, including co-precipitation, electrodeposition, sol–gel, hydrothermal, chemical vapor deposition, atomic layer deposition, ion beam sputtering, and magnetron sputtering deposition techniques, have been detailed. Besides a description of these procedures, in this review, we also present a critical appraisal of the efficiency of the water electrolysis carried out with cells fitted with electrodes prepared with these procedures. Based on this analysis, a critical comparison of cell performance is carried out, and future prospects and expected developments of the AEMWE are discussed.

    Abstract Pearson syndrome is a rare multisystem disease caused by single large-scale mitochondrial DNA deletions (SLSMDs). The syndrome presents early in infancy and is mainly characterised by refractory sideroblastic anaemia. Prognosis is poor and treatment is supportive, thus the development of new models for the study of Pearson syndrome and new therapy strategies is essential. In this work, we report three different cell models carrying an SLMSD: fibroblasts, transmitochondrial cybrids and induced pluripotent stem cells (iPSCs). All studied models exhibited an aberrant mitochondrial ultrastructure and defective oxidative phosphorylation system function, showing a decrease in different parameters, such as mitochondrial ATP, respiratory complex IV activity and quantity or oxygen consumption. Despite this, iPSCs harbouring ‘common deletion’ were able to differentiate into three germ layers. Additionally, cybrid clones only showed mitochondrial dysfunction when heteroplasmy level reached 70%. Some differences observed among models may depend on their metabolic profile; therefore, we consider that these three models are useful for the in vitro study of Pearson syndrome, as well as for testing new specific therapies.     Abstract Abstract Background The vision loss in Leber hereditary optic neuropathy patients is due to mitochondrial DNA mutations. No treatment has shown a clear-cut benefit on a clinically meaningful end-point. However, clinical evidences suggest two therapeutic approaches: the reduction of the mutation load in heteroplasmic patients or the elevation of mitochondrial DNA amount in homoplasmic patients. Results Here we show that ketogenic treatment, in cybrid cell lines, reduces the percentage of the m.13094 T > C heteroplasmic mutation and also increases the mitochondrial DNA levels of the m.11778G > A mitochondrial genotype. Conclusions These results suggest that ketogenic diet could be a therapeutic strategy for Leber hereditary optic neuropathy.

    Abstract Illicit and legal drugs of abuse, including alcohol, continue to be in the focus of many governmental national and international studies due to the important consequences of their consumption at both individual and social level. Estimation of drug use at the community level from the concentrations of the drugs themselves or their major metabolites measured in wastewater has become an increasingly accepted and extended tool, complementary to the methods traditionally used for this purpose. The present work describes the application of this approach, generally known as wastewater epidemiology, to investigate the latest drug consumption patterns and trends in the European city of Barcelona. To this end, a total of 19 selected drugs of abuse and metabolites were monitored at the inlet of one of the main wastewater treatment plants of Barcelona every day during one week in March between 2011 and 2015. Analysis of the selected drugs and metabolites in the wastewater samples was performed by means of two methodologies based on liquid chromatography-tandem mass spectrometry (LC-MS/MS), and the concentrations obtained were translated into consumption data. In agreement with official records, alcohol, followed by cannabis, cocaine, amphetamine-like compounds, and methadone were the most consumed drugs. Alcohol, cannabis, and cocaine consumption were on average 18mL(14g)/day/inhabitant (>15), 38g/day/1000 inhabitants aging 15–64, and 2.4g/day/1000 inhabitants aging 15–64, respectively. As for drug use trends, consumption increased over the 5years monitored for all drugs, but for heroin and diazepam. Weekly profiles characterized by higher consumption over the weekend as compared to weekdays were observed only for alcohol, cocaine, and MDMA. Extrapolation of the data obtained for the area under study to the national Spanish territory yields consumption figures of 142t of illicit drugs per year and >2500 million euro turnover per year in the black market.

    Abstract Mitochondrial oxidative phosphorylation disorders are extremely heterogeneous conditions. Their clinical and genetic variability makes the identification of reliable and specific biomarkers very challenging. Until now, only a few studies have focused on the effect of a defective oxidative phosphorylation functioning on the cell’s secretome, although it could be a promising approach for the identification and pre-selection of potential circulating biomarkers for mitochondrial diseases. Here, we review the insights obtained from secretome studies with regard to oxidative phosphorylation dysfunction, and the biomarkers that appear, so far, to be promising to identify mitochondrial diseases. We propose two new biomarkers to be taken into account in future diagnostic trials.

    Abstract Abstract Immune effector responses against Plasmodium falciparum include antibody-mediated activation of innate immune cells, which can induce Fc effector functions, including antibody-dependent cellular cytotoxicity, and the secretion of cytokines and chemokines. These effector functions are regulated by the composition of immunoglobulin G (IgG) Fc N-linked glycans. However, a role for antibody-mediated natural killer (NK) cells activation or Fc N-linked glycans in pregnant women with malaria has not yet been established. Herein, we studied the capacity of IgG antibodies from pregnant women, with placental malaria or non-placental malaria, to induce NK cell activation in response to placental malaria-associated antigens DBL2 and DBL3. Antibody-mediated NK cell activation was observed in pregnant women with malaria, but no differences were associated with susceptibility to placental malaria. Elevated anti-inflammatory glycosylation patterns of IgG antibodies were observed in pregnant women with or without malaria infection, which were not seen in healthy non-pregnant controls. This suggests that pregnancy-associated anti-inflammatory Fc N-linked glycans may dampen the antibody-mediated activation of NK cells in pregnant women with malaria infection. Overall, although anti-inflammatory glycans and antibody-dependent NK cell activation were detected in pregnant women with malaria, a definitive role for these antibody features in protecting against placental malaria remains to be proven.

    Abstract Summary: Immunoglobulin G (IgG) antibodies that activate Fc-mediated immune functions have been correlated with vaccine efficacy, but it is difficult to unravel the relative roles of multiple IgG and Fc receptor (FcR) features that have the capacity to influence IgG-FcR complex formation but vary on a personalized basis. Here, we develop an ordinary differential-equation model to determine how personalized variability in IgG subclass concentrations and binding affinities influence IgG-FcγRIIIa complex formation and validate it with samples from the HIV RV144 vaccine trial. The model identifies individuals who are sensitive, insensitive, or negatively affected by increases in HIV-specific IgG1, which is validated with the addition of HIV-specific IgG1 monoclonal antibodies to vaccine samples. IgG1 affinity to FcγRIIIa is also prioritized as the most influential parameter for dictating activation broadly across a population. Overall, this work presents a quantitative tool for evaluating personalized differences underlying FcR activation, which is relevant to ongoing efforts to improve vaccine efficacy.

    Abstract Adipogenesis is accompanied by differentiation of adipose tissue-derived stem cells to adipocytes. As part of this differentiation, biogenesis of the oxidative phosphorylation system occurs. Many chemical compounds used in medicine, agriculture or other human activities affect oxidative phosphorylation function. Therefore, these xenobiotics could alter adipogenesis. We have analyzed the effects on adipocyte differentiation of some xenobiotics that act on the oxidative phosphorylation system. The tested concentrations have been previously reported in human blood. Our results show that pharmaceutical drugs that decrease mitochondrial DNA replication, such as nucleoside reverse transcriptase inhibitors, or inhibitors of mitochondrial protein synthesis, such as ribosomal antibiotics, diminish adipocyte differentiation and leptin secretion. By contrast, the environmental chemical pollutant tributyltin chloride, which inhibits the ATP synthase of the oxidative phosphorylation system, can promote adipocyte differentiation and leptin secretion, leading to obesity and metabolic syndrome as postulated by the obesogen hypothesis.

Para aumentar a densidade energética sem causar transtornos ao animal, atenção tem sido direcionada à suplementação lipídica e alimentos com altas concentrações de lipídeos. Várias fontes de lipídeos podem ser utilizadas como o sebo animal, as sementes de oleaginosas e a gordura protegida. O objetivo desta revisão foi estudar os efeitos da utilização de dietas com suple- mentação de diferentes fontes de gordura na lactação de vacas em produção.
