    Abstract The determination of mineral concentrations in rice grain samples is crucial for analyzing their nutritional content. Most mineral content analysis techniques depend on inductively coupled plasma (ICP) spectrometry and are often complicated, expensive, time-consuming, and laborious. Recently, the handheld X-ray fluorescence (XRF) spectrometer has been randomly used in earth sciences; however, it is hardly practiced in quantifying mineral content in rice samples. In this research, the reliability of XRF results was compared with that of the ICP-OES to determine zinc (Zn) concentration in rice (Oryza sativa L.). Approximately 200 dehusked rice samples and four known high-Zn samples were analyzed using both XRF and ICP-OES techniques. The concentrations of Zn were recorded using the XRF technique and then correlated with the ICP-OES results. The results indicated a high positive relationship between two methods, with R² = 0.83, p = 0.000, and the Pearson correlation value of 0.91 at the level of 0.05. This work demonstrates the potential of XRF as a reliable and low-cost as well as an alternative technique to ICP-OES methods for determining Zn content in rice as it allows the analysis of a greater number of samples in a short period at a considerably low price.

    Abstract By using PacBio HiFi technology, we produced over 700 Gb of long-read sequencing (LRS) raw data; and by using Illumina paired-end whole-genome shotgun (WGS) sequencing technology, we generated more than 70 Gb of short-read sequencing (SRS) data. With LRS data, we assembled one genome and then generate a set of annotation data for an early-matured Geng/japonica glutinous rice mega variety genome, Longgeng 57 (LG57), which carries multiple elite traits including good grain quality and wide adaptability. Together with the SRS data from three parents of LG57, pedigree genome variations were called for three representative types of genes. These data sets can be used for deep variation mining, aid in the discovery of new insights into genome structure, function, and evolution, and help to provide essential support to biological research in general.

    Abstract Treated wastewater can promote to minimizing the water stress in agricultural field due to the scarcity of irrigation water. The aim of the experiment was to determine the toxic metallic contamination for wastewater irrigation in the investigated area. Analytical techniques were used to determine quality parameter and ionic constituents. Samples were considerably alkaline (pH = 7.55 to 10.05). As for recommended TDS, samples were categorizing as freshwater (60%) and brackish water (40%). Electrical Conductivity (EC) and Sodium Absorption Ratio (SAR) express the salinity and alkalinity indicates the classes C4S1, C3S1 & C2S1. Soluble sodium percentage (SSP) categorized in different grade in samples. Regarding Hardness (HT), samples were rank as very hard, hard, and moderately hard. Upon analysis, all HCO₃ was extremely hazardous, noxious for watering to plants and 56% samples of Cl, 92% of Cu and all sample of Cd were exceeded the limit thus risky for irrigation usage but zinc and iron have no effect on irrigation. Finally, the results revealed that all samples except few under test were unsuitable for irrigation due to having toxic metals such as Cu, Mn, Cd, HCO₃, and Cl. From the above discussion, we concluded that most of the parameters like Cu, Mn, Cd, HCO₃, and Cl were exceeded the maximum acceptable level and wastewater must be treated to remove all toxic elements before irrigation into the field. Moreover, this wastewater is a great value to reducing the water stress in farm level due to limited irrigation water.

    Abstract Dominant early heading (DEH) in rice (Oryza sativa L.) is of interest in both breeding and genetics. The genetic mechanisms underlying DEH have remained largely unclear. We have developed a near-isogenic DEH line without yield drag named DEH_229 by sister-line backcross (BC) breeding with MH63, a restorer, as the genetic background. We conducted a pilot genetic investigation under both short-day (SD) and long-day (LD) conditions. The DEH line harbored only 1.06% variation in the genome sequence relative to MH63. The variants were distributed throughout the genome. Using QTL mapping by sequencing (QTL-seq) on an F₂ population derived from a cross of MH63 × DEH_229, 57 loci were detected under the SD condition. Joint mapping employing a genome-wide association study with accessions from the 3000 rice genome sequencing project (3K-RG), reduced the number of QTL by 43.9%. Using Rice Functional Genomics & Breeding (RFGB) database, the number of SNP cluster regions within the QTL regions reduced by 27.3%. Further comparison of the genome variation between DEH_229 and MH63 in addition to gene annotation information revealed a new DEH allele of DTH3 with multiple variable sites as a possible major factor underlying the early-heading phenotype of DEH_229. An InDel marker, ZMEH_1, was designed based on the variation between DEH_229 and MH63 within this region. It accounted for 86.0% of heading date variation under both SD and LD conditions in 109 randomly chosen progeny derived from extreme lines of the MH63 × DEH_229 population. This study reveals the genetic complexity of DEH in the near-isogenic line and may provide useful material and marker information for plant molecular breeding.
