Abstract Proposing the concept of a brain-gut-skin axis has led some researchers to recognize the relationship among brain activity, gut microbiota, and the skin. Hair follicles are skin accessory organs, a previously unnoticed target tissue for classical neurohormones, neurotrophins, and neuropeptides. Some studies have shown a relationship between the central nervous system and hair follicles that an imbalance in the gut bacteria can affect hair follicle density. This review summarizes existing evidence from literature and explores clues supporting a connection linking the brain, gut microbiota, and hair follicles. It amalgamates previously proposed partial concepts into a new, unified concept-the "brain-gut microbiota-hair follicle" axis, -which suggests that modulation of the microbiome via probiotics can have positive effects on hair follicles. This review also explores how preclinical research on hair follicles can propel novel and clinically untapped applications.

Abstract Digital technology promotes the innovation and development of design. With the development of science and technology, design has changed the traditional design mode. We must stimulate the innovative potential of design and let digital technology play a greater driving force. The current era of "internet of all things" is also a new challenge to the development of human civilization. Game design as an important part of modern design, its design innovation is worth thinking about: How to use the rapidly changing Science and Technology; how to organically connect the real world with the virtual world; how to skillfully integrate human care into the game design; how to use the game to pay attention to special groups; how to make the function of the game get multi-dimensional presentation and other aspects are worth discussing.

Abstract Sports injuries, particularly bone injuries, have become increasingly prevalent in recent years. As athletes push themselves harder and become more competitive, the risk of injury rises. Additionally, the aging population has contributed to the increased prevalence of bone injuries, including those related to the knee joint. As a result, significant research efforts have focused on developing new techniques for knee joint repair. In this study, a novel hyaluronic acid-Alginate filler composition containing hydroxyapatite-titanium nanoparticles (HA/Ti-NPs) was prepared through freeze-drying. The crystalline properties of the porous scaffolds were analyzed using X-ray diffraction (XRD), while scanning electron microscope (SEM) images were used to visualize the scaffold structure. The morphological investigation demonstrated that adding 15 wt% Ti-NP to the scaffold reduces percentages of porosity to about 76.5 ± 5, while increasing the elastic modulus. Furthermore, the presence of nanoparticles decreased the rate of degradation, increased crystallinity, reduced pore size, and promoted calcium release, creating a favorable environment for bone tissue growth. The scaffold's degradation pH is close to neutral, which is ideal for bone tissue treatment. Adding 15 wt% Ti-NP reduces the contact angle from 120° to 93°. The composite scaffolds promote the proliferation and growth of osteoblast cells, and after 48 h, degraded material improved cell viability, growth, and proliferation. The three-component hyaluronic acid-Alginate/HA scaffolds with 15 wt% Ti-NP exhibited suitable morphological, mechanical, physical, and chemical properties for bone applications, as well as high potential for joint bonding. These porous scaffolds have the potential to be used as bone fillers of sport injuries and further research is needed to confirm their efficacy.

Abstract To address the challenges encountered by Chinese primary school students, particularly left-behind and migrant children, who exhibit a preference for animations, video games, and short videos over reading books and struggle with Chinese-English bilingual skills, this study introduces an educational robot AI-assisted method for simultaneous bilingual reading. To assess the effectiveness of this method, a 6-month Chinese-English bilingual extracurricular reading comparative experiment was conducted involving 85 grade 5 students from two classes in a primary school in Hangzhou, China. The AI-assisted class freely read 100 bilingual/English electronic picture books and 200 Chinese electronic classic serial picture books by employing the AI-assisted human-computer interactive electronic reading mode of the "Educational Robot+Audio Electronic Picture Book+Character-play Based Reading." In contrast, the paper book group read the same content presented in the traditional paper book format, following the "regular independent reading" mode. Post-experimental analyses were conducted employing

Abstract Background: Exercise is an important factor for the treatment and rehabilitation of depression. Traditional Chinese exercise is considered as an effective treatment for depression. In recent years, many studies have shown that Chinese exercise therapy may be safe in the treatment of depression. To evaluate the therapeutic effect of traditional Chinese exercise on depression using network meta-analysis.
Methods: PubMed, Cochrane Library, Web of Science, China National Knowledge Infrastructure, VIP shop Database, China Biomedical Literature Database, and Wan Fang Data Knowledge Service Platform were included in randomized controlled studies on traditional Chinese exercises for the treatment of depression from the establishment of the library to July 2023. The literature was extracted and assessed for risk of bias using the Cochrane Risk of Bias Assessment and statistically analyzed using Review Manager 5.4 and Stata 14.2.
Results: A total of 25 studies involving 3 exercises with a total of 1605 cases were included. The results of the network meta-analysis showed that each exercise significantly reduced the 24-item Hamilton Depression Scale (HAMD-24), Hamilton Anxiety Scale, Beck Depression Inventory, and Self-rating Depression Scale scores. However, conventional treatments have the advantage of improving the sleep quality of patients with depression.
Conclusions: The 3 exercises included in this study were effective for depression. Baduanjin and Tai Chi were better at improving the outcomes related to depression and anxiety. The effect of sleep quality intervention was slightly worse. Larger, high-quality randomized controlled trials are needed to compare interventions across exercises in a more granular manner.

Abstract Variable screening methods have been shown to be effective in dimension reduction under the ultra-high dimensional setting. Most existing screening methods are designed to rank the predictors according to their individual contributions to the response. As a result, variables that are marginally independent but jointly dependent with the response could be missed. In this work, we propose a new framework for variable screening, random subspace ensemble (RaSE), which works by evaluating the quality of random subspaces that may cover multiple predictors. This new screening framework can be naturally combined with any subspace evaluation criterion, which leads to an array of screening methods. The framework is capable to identify signals with no marginal effect or with high-order interaction effects. It is shown to enjoy the sure screening property and rank consistency. We also develop an iterative version of RaSE screening with theoretical support. Extensive simulation studies and real-data analysis show the effectiveness of the new screening framework.

Abstract The Rumei Hydropower Station is a typical cascade hydropower development project in a plateau area. The dam site is located in an area with complex topography, lithology, and geological structure. Geological disasters are developed in the area, mainly debris flow. Thus, taking the dam site and the surrounding areas as key evaluation objects, the engineering geological characteristics, geological environment characteristics, and the susceptibility and risk of geological disasters that may be caused are predicted and evaluated. The main methods used in this assessment are the binary logistic regression model and expert evaluation. The results show that the susceptibility to geological disasters is small and medium. The results of this study could provide a scientific basis for the rationality of the general layout and site selection of the project construction in the plateau water elevator level development reservoir area.

Abstract Objective: To investigate the effect of low-frequency rTMS combined with fasudil on neurological function in patients with acute cerebral infarction and its mechanism. Method:120 patients with acute cerebral infarction admitted from January 2016 to January 2019 were randomly divided into four groups: control group (n = 30), fasudil group (n = 30), rTMS group (n = 30), fasudil + rTMS group (n = 30). The changes of NIHSS score, ADL score, serum GAP- 43, serum RhoA and serum ROCK-II were observed and recorded before and after treatment. Results: The ADL scores and serum GAP-43 levels of the four groups after treatment were higher than those before treatment (P < 0.05); the NIHSS scores, serum RhoA and serum ROCK-II levels of the four groups after treatment were lower than those before treatment (P < 0.05); After treatment, ADL score and serum GAP-43 level in fasudil+rTMS group were higher than those in other three groups (P < 0.05); After treatment, NIHSS score, serum RhoA and serum ROCK-II levels in fasudil+rTMS group were lower than those in other three groups (P < 0.05). After treatment, NIHSS score was positively correlated with RhoA and ROCK-II content, negatively correlated with GAP-43 content and ADL score. Conclusions: Low-frequency rTMS combined with fasudil in the treatment of acute cerebral infarction can significantly improve neurological deficits, improve daily living ability, promote axonal remodeling and regeneration. It may be one of its mechanisms by increasing the expression of GAP-43 in serum and reducing the expression levels of RhoA and ROCK-II in serum. It has high clinical value.

Developing New Lys-C Peptide Mapping Methods with Minimized Assay-Induced Artifacts to Fully Characterize Antibodies

    2023  Volume 1327

Abstract Peptide mapping is an important tool used to confirm that the correct sequence has been expressed for a protein and to evaluate protein post-translational modifications (PTMs) that may arise during the production, processing, or storage of protein drugs. Our new orally administered drug (Ab-1), a single-domain antibody, is highly stable and resistant to proteolysis. Analysis via the commonly used tryptic mapping method did not generate sufficient sequence coverage. Alternative methods were needed to study the Ab-1 drug substance (75 mg/mL) and drug product (3 mg/mL). To meet these analytical needs, we developed two new peptide mapping methods using lysyl endopeptidase (Lys-C) digestion. These newly developed protein digestion protocols do not require desalting/buffer-exchange steps, thereby reducing sample preparation time and improving method robustness. Additionally, the protein digestion is performed under neutral pH with methionine acting as a scavenger to minimize artifacts, such as deamidation and oxidation, which are induced during sample preparation. Further, the method for low-concentration samples performs comparably to the method for high-concentration samples. Both methods provide 100% sequence coverage for Ab-1, and, therefore, enable comprehensive characterization for its product quality attribute (PQA) assessment. Both methods can be used to study other antibody formats.
