    Abstract Human leukocyte antigen (HLA) proteins are a group of glycoproteins that are expressed at the cell surface, where they present peptides to T cells through physical interactions with T-cell receptors (TCRs). Hence, characterizing the set of peptides presented by HLA proteins, referred to hereafter as the immunopeptidome, is fundamental for neoantigen identification, immunotherapy, and vaccine development. As a result, different methods have been used over the years to identify peptides presented by HLA proteins, including competition assays, peptide microarrays, and yeast display systems. Nonetheless, over the last decade, mass spectrometry-based immunopeptidomics (MS-immunopeptidomics) has emerged as the gold-standard method for identifying peptides presented by HLA proteins. MS-immunopeptidomics enables the direct identification of the immunopeptidome in different tissues and cell types in different physiological and pathological states, for example, solid tumors or virally infected cells. Despite its advantages, it is still an experimentally and computationally challenging technique with different aspects that need to be considered before planning an MS-immunopeptidomics experiment, while conducting the experiment and with analyzing and interpreting the results. Hence, we aim in this chapter to provide an overview of this method and discuss different practical considerations at different stages starting from sample collection until data analysis. These points should aid different groups aiming at utilizing MS-immunopeptidomics, as well as, identifying future research directions to improve the method.

    Abstract AI systems in medicine and healthcare are being extensively explored in prevention, diagnosis, novel drug designs and after-care. The application of AI technology in healthcare systems promises impressive outcomes such as equalising healthcare, reducing mortality rate and human error, reducing medical costs, as well as reducing reliance on social services. In the light of the WHO "Decade of Healthy Ageing", AI applications are designed as digital innovations to support the quality of life for older persons. However, the emergence of evidence of different types of algorithmic bias in AI applications, ageism in the use of digital devices and platforms, as well as age bias in digital data suggests that the use of AI might have discriminatory effects on older population or even cause harm. This paper addresses the issue of age biases and age discrimination in AI applications in medicine and healthcare systems and try to identify main challenges in this area. It will reflect on the potential of AI applications to amplify the already existing health inequalities by discussing two levels where potential negative impact of AI on age inequalities might be observed. Firstly, we will address the technical level of age bias in algorithms and digital datasets (especially health data). Secondly, we will discuss the potential disparate outcomes of automatic decision-making systems (ADMs) used in healthcare on the older population. These examples will demonstrate, although only partially, how AI systems may create new structures of age inequalities and novel dimensions of exclusion in healthcare and medicine.

    Abstract This paper provides a systematic review of studies assessing the impact of climate change on crop yields in southern Africa. Moreover, it synthesises the current knowledge of the impact of elevated ambient CO₂ levels (eCO₂) and temperatures on physiological processes, and the application of this knowledge in mechanistic crop models. While eCO₂ evidently has a strong impact on photosynthesis and crop water use, it is uncertain how this will work out for the climatic and crop management conditions prevailing in southern Africa. The impact of heat stress on crop reproductive processes and the process of transpiration cooling mitigating heat stress are poorly represented in models, while both process are relevant given the climatic conditions prevailing in southern Africa. Twenty studies assessing the impact of climate change on future yields of crops, mostly maize, have been retrieved. The results suggest that potato, Bambara groundnut and sugarcane yields may improve. No consistent trends for maize and sorghum could be identified. While yield predictions are obviously context-specific, large uncertainties related to climate predictions and crop models imply results should be treated with caution. Suggestions are made for field experimentation and the improved application of crop models for climate change research in the region.
