    Abstract Ferroptosis, a type of cell death involving iron and lipid peroxidation, has been found to be closely associated with the development of many diseases. Mitochondria are vital components of eukaryotic cells, serving important functions in energy production, cellular metabolism, and apoptosis regulation. Presently, the precise relationship between mitochondria and ferroptosis remains unclear. In this study, we aim to systematically elucidate the mechanisms via which mitochondria regulate ferroptosis from multiple perspectives to provide novel insights into mitochondrial functions in ferroptosis. Additionally, we present a comprehensive overview of how mitochondria contribute to ferroptosis in different conditions, including cancer, cardiovascular disease, inflammatory disease, mitochondrial DNA depletion syndrome, and novel coronavirus pneumonia. Gaining a comprehensive understanding of the involvement of mitochondria in ferroptosis could lead to more effective approaches for both basic cell biology studies and medical treatments.

    Abstract With the acceleration of urbanization, the relationship between an area's population and its economy has become the main indicator of regional differences. Therefore, a comprehensive study of population-economy spatial dispersion changes and their influencing factors will help identify the driving force of population change and economic development. Moreover, it will facilitate measurement of the coupling and coordination relationship between industrial layout and the environment. Accordingly, based on the 2018 population and economic data of Anhui Province, China, this study used a land use impact model to spatialize socioeconomic indicators, and considered the population–economic spatial relationship of Anhui Province with its terrain. By doing so, the study hopes to promote the construction of new urbanization in Anhui Province and provide a reference for the development of central provinces in China. The results indicated that: 1) The predicted population and economic densities of the study area fit well with the actual statistical values, which suggests that the spatialization results could reflect the actual situation of population and economic distribution. Based on the simulation data, the spatial relationship between the population and economy can be divided into three types: leading, lagging, and coordinating. The overall distribution characteristics of the spatial relationship between population and economy in the study area were as follows: in the mountainous area of Southern Anhui, the lag type was dominant, and economic agglomeration lagged behind that of population; in northern Anhui, the coordination type was dominant, and population and economic development were in balance; finally, in central Anhui, the lead type was dominant, and economic agglomeration was greater than population agglomeration. 2) The correlation between terrain factors and the population was higher than that with the economy, with changes in the terrain factors strengthening the influence of terrain on population, compared to ...

    Abstract Chuanxiong rhizoma (CX) has been utilized for centuries as a traditional herb to treat blood stasis syndromes. However, the pharmacological mechanisms are still not completely revealed. This research was aimed at exploring the molecular mechanisms of CX treatment for thrombosis. Network pharmacology was used to predict the potential anti-thrombosis mechanism after correlating the targets of active components with targets of thrombosis. Furthermore, we verified the mechanism of using CX to treat thrombosis via molecular docking and in vitro experiments. Network pharmacology results showed that a total of 18 active ingredients and 65 targets of CX treatment for thrombosis were collected, including 8 core compounds and 6 core targets. We revealed for the first time that tissue factor (TF) had a close relationship with most core targets of CX in the treatment of thrombosis. TF is a primary coagulation factor in physiological hemostasis and pathological thrombosis. Furthermore, core components of CX have strong affinity for core targets and TF according to molecular docking analysis. The in vitro experiments indicated that Ligustilide (LIG), the representative component of CX, could inhibit TF procoagulant activity, TF mRNA and protein over-expression in a dose-dependent manner in EA.hy926 cells through the PI3K/Akt/NF-κB signaling pathway. This work demonstrated that hemostasis or blood coagulation was one of the important biological processes in the treatment of thrombosis with CX, and TF also might be a central target of CX when used for treating thrombosis. The inhibition of TF might be a novel mechanism of CX in the treatment of thrombosis.

    Abstract Tracking the immune microenvironment of tumours is essential for understanding the mechanisms behind the effectiveness of cancer immunotherapies. Molecular imaging of tumour-infiltrating leukocytes (TILs) can be used to non-invasively monitor the tumour immune microenvironment, but current imaging agents do not distinguish TILs from leukocytes resident in other tissues. Here we report a library of activatable molecular probes for the imaging, via near-infrared fluorescence, of specific TILs (including M1 macrophages, cytotoxic T lymphocytes and neutrophils) in vivo in real time and also via excreted urine, owing to the probes' renal clearance. The fluorescence of the probes is activated only in the presence of both tumour and leukocyte biomarkers, which allows for the imaging of populations of specific TILs in mouse models of cancers with sensitivities and specificities similar to those achieved via flow-cytometric analyses of biopsied tumour tissues. We also show that the probes enable the non-invasive evaluation of the immunogenicity of different tumours, the dynamic monitoring of responses to immunotherapies and the accurate prediction of tumour growth under various treatments.

    Abstract Real-time imaging of programmed cancer cell death (PCD) is imperative to monitor cancer therapeutic efficacy and tailor therapeutic regimens; however, specific in vivo detection of intratumoral pyroptosis remains challenging. Herein, a dual-locked and tandem activatable probe (DTAP) is reported for near-infrared fluorescence (NIRF) imaging of intratumoral pyroptosis during cancer chemo-immunotherapy in living mice. The probe comprises a hemicyanine dye dual-locked with an enzyme-responsive moiety that can be tandemly cleaved by pyroptosis-related biomarker (Caspase-1) and cancer biomarker (GGT) to turn on its NIRF signal. As pyroptosis plays a vital role in triggering anti-tumor immune responses, the activated signal of DTAP correlates well with the population of tumor-infiltrating cytotoxic T lymphocytes and tumor growth inhibition, consequently permitting the prediction of cancer therapeutic efficacy. This study also provides a non-invasive technique to study the regulatory mechanism of pyroptosis in cancer therapy and to optimize cancer chemo-immunotherapies for precision medicine.

    Abstract In this study, through structural design, thin film doping modification, and electrode preparation, a high-performance triboelectric nanogenerator (TENG) has been achieved, and an electromagnetic generator is incorporated as an energy supplement to construct a self-powered sensing system for gas alarm and environmental monitoring. The high porosity of NiO derived from MOFs, high charge accumulation, and the high charge carrier mobility of N-MWCNTs promote charge capture and transfer, greatly enhancing the output performance of the triboelectric nanogenerator. Under the optimal doping ratio, the open-circuit voltage can reach 860 V, and the maximum short-circuit current up to 47 μA, sufficient to power 600 LEDs lights, achieving a power density of 10.45 W/m². Meanwhile, MXene/Carbon/Ecoflex electrodes were fabricated to enhance the contact efficiency between the thin film and electrodes, further obtaining an open circuit voltage of 1020 V. This work provides a viable solution for the design of high-performance hybrid nanogenerators.

    Abstract Cancer is a major cause of death globally, and traditional treatments often have limited efficacy and adverse effects. Immunotherapy has shown promise in various malignancies but is less effective in tumors with low immunogenicity or immunosuppressive microenvironment, especially sarcomas. Tertiary lymphoid structures (TLSs) have been associated with a favorable response to immunotherapy and improved survival in cancer patients. However, the immunological mechanisms and clinical significance of TLS in malignant tumors are not fully understood. In this review, we elucidate the composition, neogenesis, and immune characteristics of TLS in tumors, as well as the inflammatory response in cancer development. An in-depth discussion of the unique immune characteristics of TLSs in lung cancer, breast cancer, melanoma, and soft tissue sarcomas will be presented. Additionally, the therapeutic implications of TLS, including its role as a marker of therapeutic response and prognosis, and strategies to promote TLS formation and maturation will be explored. Overall, we aim to provide a comprehensive understanding of the role of TLS in the tumor immune microenvironment and suggest potential interventions for cancer treatment.
