Abstract An organism's gut microbiome can alter its fitness, yet we do not know how gut microbiomes change as their hosts evolve in the wild. We took advantage of a five-decade 'chronosequence' of translocated fish populations to examine associated changes in the gut microbiome. Populations of Trinidadian guppies have displayed parallel phenotypic convergence six times when moved from high predation (HP) to low predation (LP) environments. Across four drainages, we found microbiomes of fish translocated 5-6 years prior to sampling were already distinct from the microbiomes of their HP source populations. Changes in environmental conditions were most important in driving this shift, followed by phenotypic shifts in gut morphology. After 30-60 years in LP environments, microbiome composition was still distinct from native LP populations, but microbiome function was not. We found some evidence that nitrogen fixation enhanced gut nutrient absorption, but most functional shifts were not parallel across drainages. Stream-and drainage-specific signatures were present for both composition and function, despite our overall finding of consistent microbiome change across drainages. As we unravel the complexities of host-microbiome evolution in the wild, studies should consider environmental microbial colonization, host phenotypic plasticity in nature, and more realistic environmental conditions excluded from laboratory studies.

Abstract Abstract: For some ranges of its parameters and arguments, the series for Tweedie probability density functions are sometimes exceedingly difficult to sum numerically. Existing numerical implementations utilizing inversion techniques and properties of stable distributions can cope with these problems, but no single one is successful in all cases. In this work we investigate heuristically the nature of the problem, and show that it is not related to the order of summation of the terms. Using a variable involved in the analytical proof of convergence of the series, the critical parameter for numerical non-convergence (“alpha”) is identified, and an heuristic criterion is developed to avoid numerical non-convergence for a reasonably large sub-interval of the latter. With these practical rules, simple summation algorithms provide sufficiently robust results for the calculation of the density function and its definite integrals. These implementations need to utilize high-precision arithmetic, and are programmed in the Python programming language. A thorough comparison with existing R functions allows the identification of cases when the latter fail, and provide further guidance to their use.

Abstract Peak cognitive performance is of paramount importance for the sports and competitive activities that depend on a high level of cognitive performance, such as eSports. The popularity of eSports has increased rapidly in recent years and will probably continue to rise in the forthcoming years. Above all, eSports practitioners require higher levels of cognitive abilities and motor skills for optimal performance and may benefit from adequate body composition and proper nutrition, as happens with more traditional athletes. However, there is a noticeable shortage of scientific knowledge in this area, including in nutritional-related aspects. Therefore, the objective of this non-systematic review is to summarize the nutritional strategies that may enhance health, cognitive performance, decrease reaction time and minimize fatigue. This information may be relevant for eSports competitors and set a base for further investigations, which could ultimately lead to the establishment of nutritional recommendations specific for this competitive population.

Abstract Pseudoexfoliation, one of the most frequent crystalline lens complex disorders, is prevalent in up to 30% of individuals older than 60 years old. This disease can lead to severe conditions, such as subluxation or dislocation of the lens, due to the weakening of the zonules. The goal for the present study was to understand the relevant biomechanical features that can lead to the worsening of an individual’s visual capacity under pseudoexfoliation. To this end, finite element models based on a 62-year-old lens complex were developed, composed by the capsular bag, cortex, nucleus, anterior, equatorial, and posterior zonular fibers. Healthy and pseudoexfoliative conditions were simulated, varying the location of the zonulopathy (inferior/superior) and the degenerated layer. The accommodative capacity of the models with inferior dialysis of the zonular fibers was, on average, 4.7% greater than for the cases with superior dialysis. If the three sets of zonules were disrupted, this discrepancy increased to 14.9%. The present work provides relevant data to be further analyzed in clinical scenarios, as these models (and their future extension to a wider age range) can help in identifying the most influential regions for the reduction of the visual capacity of the lens.

Abstract Abstract The habitat loss is the main threat for many parrot species worldwide. However, the disturbed environments can influence the occurrence of the species positively or negatively, depending on its structure and potential use as an alternative environment. Therefore, this work aims to determine the relationship between land use types and the occurrence of the threatened Red-browed Amazon, identifying the land use types related to species’ occurrence and the direction of these relationships. Seven land use types were significantly associated with the species’ occurrence: Primary/Secondary Advanced/Medium Natural Forest, Secondary Natural Vegetation, Pasture, Outcrop/Bare Soil, Mangroves, Rivers and Urban Areas. We found that some land use types that are structurally different from the original environments (Pasture, Outcrop/Bare Soil and Urban Areas) positively affected the occurrence of the species. The relationship between intensity of occurrence and highly anthropized land use types, suggests a plasticity of the species in habitat use that may be contributing to the maintenance of Red-browed Amazon populations in highly disturbed habitats. Therefore, we proposed that the management of disturbed areas, along with usual conservation strategies (e.g., conservation of forest remnants, restoration of degraded areas), could contribute to Red-browed Amazon conservation.

Abstract Sulfite predominantly accumulates in the brain of patients with isolated sulfite oxidase (ISOD) and molybdenum cofactor (MoCD) deficiencies. Patients present with severe neurological symptoms and basal ganglia alterations, the pathophysiology of which is not fully established. Therapies are ineffective. To elucidate the pathomechanisms of ISOD and MoCD, we investigated the effects of intrastriatal administration of sulfite on myelin structure, neuroinflammation, and oxidative stress in rat striatum. Sulfite administration decreased Fluoromyelin TM and myelin basic protein staining, suggesting myelin abnormalities. Sulfite also increased the staining of NG2, a protein marker of oligodendrocyte progenitor cells. In line with this, sulfite also reduced the viability of MO3.13 cells, which express oligodendroglial markers. Furthermore, sulfite altered the expression of interleukin-1β (IL-1β), interleukin-6 (IL-6), interleukin-10 (IL-10), cyclooxygenase-2 (COX-2), inducible nitric oxide synthase (iNOS) and heme oxygenase-1 (HO-1), indicating neuroinflammation and redox homeostasis disturbances. Iba1 staining, another marker of neuroinflammation, was also increased by sulfite. These data suggest that myelin changes and neuroinflammation induced by sulfite contribute to the pathophysiology of ISOD and MoCD. Notably, post-treatment with bezafibrate (BEZ), a pan-PPAR agonist, mitigated alterations in myelin markers and Iba1 staining, and IL-1β, IL-6, iNOS and HO-1 expression in the striatum. MO3.13 cell viability decrease was further prevented. Moreover, pre-treatment with BEZ also attenuated some effects. These findings show the modulation of PPAR as a potential opportunity for therapeutic intervention in these disorders.

Abstract Soil invertebrates have been widely used in ecotoxicology studies for decades, although their use as in vitro models, albeit promising, has not been pursued as much. The immune cells of earthworms (coelomocytes) and the coelomic fluid can be used, and are a highly relevant in vitro system. Although it has been tested before, to cover the testing of nanomaterials (NMs), several challenges should be considered. NMs characteristics (dispersibility, agglomeration, etc.) can interfere with the common in vitro methodologies, not only during exposure, but also during the measurements. Here, we have assessed the effect of a CuO NMs case study using surface-modified particles, functionalized for safe-by-design strategies with ascorbate, citrate, polyethylenimine, and polyvinylpyrrolidinone, plus the pristine CuO NMs and copper chloride (CuCl 2 ) for comparison. Eisenia fetida ’s coelomocytes were exposed for 24 h via the coelomic fluid. Changes in cell viability were evaluated using flow cytometry. All materials affected the cells in a dose-related manner, where CuCl 2 was the most toxic followed by the citrate-coated CuO NM. There was a strong correlation between NM characteristics, e.g., the hydrodynamic size, and the EC 50 (50% Effect Concentrations) values. This screening further confirms the potential for the usage of the standard earthworm model as an in vitro standard. Further detailed in vitro studies are needed using other NMs aiming toward their implementation and standardization. Additional cell endpoints can also be assessed, making it a high content tool for mechanistic understanding.
