    Abstract Microautoradiography (MAR) is a technique by which assimilated radioactive tracers incorporated into the biomass can be detected by a film emulsion. This allows for the testing of cellular preferences in electron donors and acceptors of individual cells in complex microbial assemblages, as well as the ability to take up substrates under diverse environmental exposures.Combination with staining techniques such as fluorescence in situ hybridization (FISH) can be used to identify the involved cells. Here, the practical aspects of a combined microautoradiography and fluorescence in situ hybridization (MAR-FISH) approach are described.

    Abstract Occupational safety and health (OSH) in construction work continues to be a problematic issue, and OSH coordinators are a pivotal initiative for improving this in the EU. However, no studies on the impact of (OSH) coordinators in construction exists. This study conceptualizes the hierarchy of controls (HOC) as a means for visualizing and evaluating the impact of OSH coordinators’ work. The study engages with a large observational material based on fieldwork notes from 107 days of observations with 12 successful OSH coordinators. The analysis shows that during the 107 observation days, the coordinators implemented 280 OSH measures and were prevented 71 times from implementing measures. Most of the implemented measures were in the administrative (53.6%) and engineering (35%) controls. This may provide part of the explanation of why an increasing focus on OSH coordination has not translated into improved OSH outcomes in construction. The study contributes with insights for OSH coordinators and professionals seeking to improve the visibility and legitimacy of their work. In addition, it may be beneficial to organizations interested in ensuring the effectiveness of their organizational OSH practices. The study also creates foundations for more research-based practices, education, and professionalization of OSH coordinators as a profession.

    Abstract Context: In individuals with hypothyroidism and overweight, levothyroxine substitution therapy is often expected to cause weight loss due to its effect on resting energy expenditure. However, despite levothyroxine-induced enhancement of resting energy expenditure, fat mass loss is rarely seen after levothyroxine substitution therapy. The mechanism behind this conundrum is unknown.
    Aim: The aim of the study was to assess the effect of levothyroxine therapy on hunger sensations and ad libitum food intake in individuals with hypothyroidism.
    Design and setting: Prospective cohort study of 18 newly diagnosed hypothyroid women (thyroid-stimulating hormone (TSH) >10 mU/L). Participants were investigated at diagnosis, after normalization of TSH (<4.0 mU/L), and after 6 months of successful treatment. Eighteen age and body mass index-matched healthy controls were also included.
    Intervention: Hypothyroid individuals were treated with levothyroxine according to European Thyroid Association guidelines.
    Main outcomes: Changes in hunger sensation were assessed using visual analog scales (cm) before and during a standardized mixed meal test, and food intake was measured during a subsequent ad libitum meal (g).
    Results: After 6 months of levothyroxine therapy, mean resting energy expenditure was increased by 144 kcal/day (10%) (P < 0.001). Weight loss was comprised of 0.8 kg fat-free mass while fat mass remained unchanged. Fasting hunger sensation increased from a mean of 4.5 (s.d. 2.2) cm to 5.5 (s.d. 2.2) cm (P = 0.047). The numerical increase in ad libitum meal intake did not reach statistical significance.
    Conclusion: Our data suggest that levothyroxine-induced hunger may be a culprit in the lack of fat mass loss from levothyroxine therapy.

    Abstract Parkinson's disease (PD) is a neurodegenerative disorder, well-known for its motor symptoms; however, it also adversely affects cognitive functions, including language, a highly important human ability. PD pathology is associated, even in the early stage of the disease, with alterations in the functional connectivity within cortico-subcortical circuitry of the basal ganglia as well as within cortical networks. Here, we investigated functional cortical connectivity related to spoken language processing in early-stage PD patients. We employed a patient-friendly passive attention-free paradigm to probe neurophysiological correlates of language processing in PD patients without confounds related to active attention and overt motor responses. MEG data were recorded from a group of newly diagnosed PD patients and age-matched healthy controls who were passively presented with spoken word stimuli (action and abstract verbs, as well as grammatically correct and incorrect inflectional forms) while focussing on watching a silent movie. For each of the examined linguistic aspects, a logistic regression classifier was used to classify participants as either PD patients or healthy controls based on functional connectivity within the temporo-fronto-parietal cortical language networks. Classification was successful for action verbs (accuracy = 0.781, p-value = 0.003) and, with lower accuracy, for abstract verbs (accuracy = 0.688, p-value = 0.041) and incorrectly inflected forms (accuracy = 0.648, p-value = 0.021), but not for correctly inflected forms (accuracy = 0.523, p-value = 0.384). Our findings point to quantifiable differences in functional connectivity within the cortical systems underpinning language processing in newly diagnosed PD patients compared to healthy controls, which arise early, in the absence of clinical evidence of deficits in cognitive or general language functions. The techniques presented here may aid future work on establishing neurolinguistic markers to objectively and noninvasively identify functional changes in the brain's language networks even before clinical symptoms emerge.

    Abstract Objective: Some studies suggest that hypothyroidism is associated with increased oxidative stress. Urinary excretion of 8-oxo-7,8-dihydroguanosine (8-oxoGuo) and 8-oxo-7,8-dihydro-2'-deoxyguanosine (8-oxodG) represents whole-body RNA and DNA oxidation, respectively. These biomarkers have only been explored sparsely in patients with thyroid disorders.
    Methods: In 45 Danish women with newly diagnosed hypothyroidism, we compared 8-oxoGuo and 8-oxodG before or shortly after initiating levothyroxine with the excretion rates at euthyroidism. We also compared the excretion of 8-oxoGuo and 8-oxodG in the patients after restored euthyroidism with 18 healthy control subjects.
    Results: Compared with baseline, none of the biomarkers changed significantly in the patients after becoming euthyroid. The geometric mean of 8-oxoGuo was 1.63 (95% CI: 1.49-1.78) nmol/mmol creatinine at baseline and 1.67 nmol/mmol at euthyroidism (95% CI: 1.53-1.83) (P = 0.39), while that of 8-oxodG was 1.28 nmol/mmol creatinine at baseline (95% CI: 1.14-1.44) and 1.32 nmol/mmol at euthyroidism (95% CI: 1.18-1.48), respectively (P = 0.47). The relative mean differences were 0.97 (95% CI: 0.91-1.04) for 8-oxoGuo and 0.97 (95% CI: 0.88-1.06) for 8-oxodG. At baseline, multiple linear regression revealed a positive association between free thyroxine and both biomarkers (8-oxoGuo, P < 0.001; 8-oxodG, P = 0.04). Furthermore, 8-oxoGuo was positively associated with age (P = 0.04) and negatively associated with thyrotropin (P = 0.02). In the control group, the geometric mean of 8-oxoGuo was 1.23 nmol/mmol creatinine (95% CI: 1.07-1.42), while that of 8-oxodG was 1.04 nmol/mmol creatinine (95% CI: 0.88-1.23). Thus, compared with control subjects, euthyroid patients showed a significantly higher level of both 8-oxoGuo (P < 0.001) and 8-oxodG (P = 0.03).
    Conclusion: In hypothyroid women, no significant effect of levothyroxine treatment on the oxidative stress biomarkers 8-oxoGuo and 8-oxodG could be demonstrated. However, the excretion of these biomarkers was significantly higher than in healthy controls.

    Abstract The present state of progress in laser wakefield acceleration encourages considering it as a practical alternative to conventional particle accelerators. A promising application would be to use a laser plasma accelerator as an injector for a synchrotron light source. The injector could deliver low charge ($\sim 100$~pC) beams to top up the storage ring and reduce the load on the chain of conventional injectors. Yet, the energy spread and jitter of the laser plasma beam pose a significant difficulty for an efficient injection. In this paper we propose a design of a prototype injector to deliver 500 MeV low-intensity electron bunches to the DESY-II electron synchrotron. The design utilizes presently available conventional accelerator technology, such as a chicane and an X-band radio frequency cavity, to reduce the energy spread and jitter of the electron beam down to a sub-per-mille level.

    Abstract Applications of laser-plasma accelerators demand low energy spread beams and high-efficiency operation. Achieving both requires flattening the accelerating fields by controlled beam loading of the plasma wave. Here, we optimize the generation of an electron bunch via localized ionization injection, such that the combination of injected current profile and averaged acceleration dynamics results in optimal beam loading conditions. This enables the reproducible production of 1.2% rms energy spread bunches with 282 MeV and 44 pC at an estimated energy-transfer efficiency of ∼19%. We correlate shot-to-shot variations to reveal the phase space dynamics and train a neural network that predicts the beam quality as a function of the drive laser.
