    Abstract The ongoing coronavirus disease (COVID-19) caused by severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) is highly contagious and fatal, posing a direct threat to human health and the global economy. Most strategies to prevent, control, and eradicate COVID-19 are established based on the specific characteristics of the pathogen. The quest for interruption and eradication of COVID-19 has moved research forward in understanding fundamental aspects of the virus genome, proteome, replication mechanisms, and virus-host interactions, which pave the way for the development of effective antiviral drugs and vaccines. This chapter provides an overview of recent progress in human coronavirus taxonomy, molecular features of the SARS-CoV-2 genome and proteome, and virus life cycle.

    Abstract Defects in graphene are both a boon and a bane for applications - they can induce uncontrollable effects but can also provide novel ways to manipulate the properties of pristine graphene. Nanoporous Graphene, which contains nanoscopic holes, has found impactful applications in sustainability domains, e.g. gas separation, water filtration membranes and battery technologies. For this report, we investigate pore formation in graphene with no defect, one and two mono-vacancies, and two di-vacancies using bespoke Dimensionally Restricted Molecular Dynamics (DR-MD) designed for the purpose. We show DR-MD to be superior to free-standing or substrate suspended configurations for simulating stable defected structures. Applying DR-MD, stable pore configurations are identified, and their formation mechanisms elucidated. We also investigated formation mechanisms due to two Stone-Wales 55-77 defects, and the formation energies of their linearly extended structures, along the zigzag and armchair directions, and when they are placed in different relative orientations. This study offers a way to identify stable porous defect structures in graphene and insights into atomistic pore formation mechanisms for an environmentally important material.

    Abstract Ankylosing spondylitis (AS) is an autoimmune inflammatory disease associated with joint inflammation and destruction. Current treatment modalities alleviate symptoms; however, they cannot cure the disease and are associated with significant side effects. Thus, we aimed to confirm the inhibitory effect of isofraxidin, a herbal extract, on pathological osteogenesis in ankylosing spondylitis to better treat patients affected by the disease. Mouse preosteoblast MC3T3-E1 subclone 14 cells were used in vitro to establish control and isofraxidin intervention groups. Cell viability was then determined using the MTT assay; the expression of osteogenic factors, including Runx2, OSX, collagen I, and ALP was measured using qRT-PCR and western blotting. Final osteogenic mineralization was performed by alizarin red staining. The results showed that isofraxidin could inhibit osteoblast viability; however, this effect was nullified at concentrations of 0-20 µM after adding 1% serum. Gene and protein expression of the osteogenic factors RUNX2, OSX, Collagen I, and ALP was inhibited, and a similar trend was exhibited at 7, 14, and 21 days after isofraxidin treatment. This trend was further verified by alizarin red staining of the final osteogenic mineralized nodules on days 7, 14, 21, and 35. Isofraxidin inhibits MC3T3-E1 subclone 14 proliferation and differentiation and may be considered a potential drug therapy for treating pathological osteogenesis in ankylosing spondylitis.

    Abstract Super-resolution imaging based on single-molecule localization has been developed for more than a decade. These techniques can break through diffraction limit of fluorescent microscopy and initially improve the resolution by an order of magnitude to ~20 nm, by introducing photoactivatable/photoswitching probes and centroid fitting method. As the demand of biological research, the localization precision of single-molecules was further improved by several state-of-the-art methods in the past several years. This review focuses on the latest developed techniques which have greatly improved the performance of single-molecule localization microscopy, from measurement principle to hardware design. These methods are essential for the study of nanostructures and biomacromolecule dynamics inside of cells.
