    Abstract Abstract Background Ocular blood flow provides a new perspective for studying the effects of diabetes and ischemic heart disease on systemic blood flow, pathological mechanisms, and prognosis. Previous studies have analyzed the hemodynamic changes of the ophthalmic artery (OA) in patients with diabetes and ischemic heart disease, but the results remain controversial due to limited observation methods. We aimed to explore the morphological and hemodynamic features in the OA in patients with type 2 diabetes (T2D) with and without acute coronary syndrome (ACS). Methods In total, 134 participants, including 30 control participants, 34 with ACS only, 34 with T2D only, and 36 with both ACS and T2D, undergoing computed tomography angiography were enrolled. Three-dimensional OA models were reconstructed, and morphological parameters of the OA were measured. In addition, numerical simulations using computational fluid dynamics were used to acquire hemodynamic parameters of the OA. Results In this study, 134 OA models were reconstructed. Morphological measurements revealed a smaller initial OA diameter in the T2D group than in the other two ACS groups. A hemodynamic simulation showed a significantly lower OA blood velocity in patients with ACS and T2D than that in controls (P < 0.001). The mass flow ratios in all disease groups were lower than those in the control group (P < 0.001, P = 0.020, and P < 0.001, respectively). The ACS and T2D groups had higher OA pressure levels than those of the control group (P = 0.013). The OA blood velocity and mass flow ratio were correlated with several clinical parameters. Conclusions This study revealed morphological and hemodynamic differences in the OA between patients with T2D with and without ACS. Furthermore, the hemodynamic characteristics of the OA correlated with clinical prognostic biomarkers, suggesting the potential predictive ability of the OA.

    Abstract Abstract Background Cerebral small vessel disease (CSVD) is a systemic disease, affecting not only the brain, but also eyes and other organs. The total CSVD score is a tool for comprehensive evaluation of brain lesions in patients with CSVD. The ophthalmic artery (OA) is a direct response to ocular blood flow. However, little is known about the correlation between CSVD and characteristics of OA. We investigated the OA morphologies and hemodynamics in patients with CSVD and the correlation between these changes and the total CSVD score. Methods This cross-sectional observational study included 34 eyes from 22 patients with CSVD and 10 eyes from 5 healthy controls. The total CSVD score was rated according to the CSVD signs on magnetic resonance imaging. OA morphological characteristics were measured on the basis of 3D OA model reconstruction. OA hemodynamic information was calculated using computational fluid dynamics simulations. Results The total CSVD score negatively correlated with the OA diameter, blood flow velocity, and mass flow ratio (all P < 0.05). After adjusting for potential confounding factors, the total CSVD score was still independently correlated with the OA blood velocity (β = − 0.202, P = 0.005). The total CSVD score was not correlated with OA angle (P > 0.05). The presence of cerebral microbleeds and enlarged perivascular spaces was correlated with the OA diameter (both P < 0.01), while the lacunar infarcts and white matter hyperintensities were correlated with the OA blood velocity (both P < 0.001). Conclusions The decrease of the blood velocity in the OA was associated with the increase in the total CSVD score. The changes of the OA diameter and velocity were associated with the presence of various CSVD signs. The findings suggest that more studies are needed in the future to evaluate CSVD by observing the morphologies and hemodynamics of OA.

    Abstract Graphene oxide–silver nanoparticle (GO-AgNPs) nanocomposites have drawn much attention for their potential in biomedical uses. However, the potential toxicity of GO-AgNPs in animals and humans remains unknown, particularly in the developing fetus. Here, we reported the GO-AgNP-mediated cytotoxicity and epigenetic alteration status in caprine fetal fibroblast cells (CFFCs). In brief, the proliferation and apoptosis rate of GO-AgNP-treated CFFCs (4 and 8 µg/mL of GO-AgNPs) were measured using the cell-counting kit (CCK-8) assay and the annexin V/propidium iodide (PI) assay, respectively. In addition, the oxidative stress induced by GO-AgNPs and detailed mechanisms were studied by evaluating the generation of reactive oxygen species (ROS), superoxide dismutase (SOD), lactate dehydrogenase (LDH), malondialdehyde (MDA), and caspase-3 and abnormal methylation. The expression of pro- and anti-apoptotic genes and DNA methyltransferases was measured using reverse transcription followed by RT-qPCR. Our data indicated that GO-AgNPs cause cytotoxicity in a dose-dependent manner. GO-AgNPs induced significant cytotoxicity by the loss of cell viability, production of ROS, increasing leakage of LDH and level of MDA, increasing expression of pro-apoptotic genes, and decreasing expression of anti-apoptotic genes. GO-AgNPs incited DNA hypomethylation and the decreased expression of DNMT3A . Taken together, this study showed that GO-AgNPs increase the generation of ROS and cause apoptosis and DNA hypomethylation in CFFCs. Therefore, the potential applications of GO-AgNPs in biomedicine should be re-evaluated.

    Abstract An integrated sampling technique, dubbed trigger sampling, was developed to capture characteristic industrial emissions or plumes. In the field experiment, a hydrogen sulfide (H2S) analyzer was used as the triggering instrument at the boundary of a refinery plant due to frequent complaints of foul smell from local residents. Ten episodic samples were captured when the H2S level surpassed the prescribed trigger level of 8.5 ppbv over a three-day period. Three non-episodic (blank) samples and 23 road-side samples were also collected for comparison. All the 36 flask samples were analyzed by gas chromatography–mass spectrometry/flame ionization detection (GC–MS/FID) for 108 volatile organic compounds (VOCs). The total VOC abundance of the event samples was exceedingly higher than the non-episodic samples by over 80 times in the extreme case. Alkanes were found to be the dominant constituents in the event samples, amounting to over 90% of the total VOC concentrations vs. only 30–40% for the blank and metropolitan samples. In addition, light alkanes in the event samples were highly correlated with the trigger species H2S (R2 = 0.82), implying their common origin. The matrix of chemical composition vs. sample types permitted easy visualization of the dominance of light alkanes for the event samples compared to other types of samples. Principle component analysis (PCA) identified two major contributors to cover 93% of the total variance arising from the 36 samples, further quantifying the distinction of the triggered episodic samples from the contrast samples. The proposed trigger sampling is a coupling of fast-and-slow measurement techniques. In this example, the fast-response H2S analyzer served to “guide” sampling to capture industrial plumes which were then characterized by a relatively slow method of GC–MS/FID for detailed chemical composition representative of the prominent sources.

    Abstract Peptidoglycan recognition proteins (PGRPs), which are evolutionarily conserved from invertebrates to vertebrates, function as pattern-recognition and effector molecules in innate immunity. In this study, a PGRP (HaPGRP-A) from the cotton bollworm, Helicoverpa armigera was identified and characterized. Sequence analysis indicated that HaPGRP-A is not an amidase-type PGRP. Increased levels of HaPGRP-A mRNA were observed in the fat body and hemocytes of H. armigera larvae following the injection of microbes or Sephadex beads. Analysis using purified recombinant HaPGRP-A showed that it (i) could bind and agglutinate Gram-negative Escherichia coli and Gram-positive Staphylococcus aureus, (ii) enhanced prophenoloxidase activation in the presence of microbes, (iii) promoted the formation of melanotic nodules in vivo, and (iv) enhanced the melanization of Sephadex beads in vivo. RNA interference assays were performed to further confirm the function of HaPGRP-A. When the expression of HaPGRP-A in H. armigera larvae was inhibited by dsHaPGRP-A injection, the phenoloxidase activity in larval hemolymph was significantly decreased and RNAi-treated insects infected with bacteria showed higher bacterial growth in hemolymph compared with infected control larvae. These results indicated that HaPGRP-A acts as a pattern recognition receptor and binds to the invading organism to trigger the prophenoloxidase activation pathway of H. armigera, and the activated phenoloxidase may participate in the melanization process of nodulation and encapsulation responses.

    Abstract This study is to develop a GC×GC method with electron capture detection (ECD) to analyze atmospheric halocarbons in the concentration range of parts per trillion by volume (pptv). To enrich atmospheric halocarbons a home-built thermal desorption (TD) device was coupled to the GC×GC-ECD. The technique of flow modulation was adopted using a Deans switch for GC×GC. Several column combinations of first and second dimensions were tested and the column set of DB–5×TG-1301 was found to show the best orthogonality for halocarbons. A series of modulation parameters were tested for their optimal settings. The modulation period (PM) was found to have minimal wrap-around when set at 3s. The modulation ratio (MR) was determined to be 7.82 to ensure reproducible results and maximum sensitivity. The modulation duty cycle (DC) was calculated to be approximately 0.17. Nine halocarbons were separated successfully and seven were calibrated with the use of a certified standard gas mixture. The correlation coefficients (R2) were greater than 0.9972. The reproducibility was better than 1.90% as expressed in relative standard deviation (RSD; N=30) and the detection limits were in the range of pptv for the target halocarbons. A field test by continuous analyzing ambient air with hourly resolution was performed to show the stability of the method as suggested by the homogeneity of certain halocarbons, while also reflecting concentration variation for others when emissions did arise.

    Abstract The encapsulation reaction in invertebrates is analogous to granuloma formation in vertebrates, and this reaction is severely compromised when ecdysone signaling is blocked. However, the molecular mechanism underlying the encapsulation reaction and its regulation by ecdysone remains obscure. In our previous study, we found that the C-type lectin HaCTL3, from the cotton bollworm Helicoverpa armigera, is involved in anti-bacterial immune response, acting as a pattern recognition receptor (PRR). In the current study, we demonstrate that HaCTL3 is involved in defense against parasites and directly binds to the surface of nematodes. Our in vitro and in vivo studies indicate that HaCTL3 enhances hemocytic encapsulation and melanization, whereas H. armigera β-integrin (Haβ-integrin), located on the surface of hemocytes, participates in encapsulation. Additionally, co-immunoprecipitation experiments reveal HaCTL3 interacts with Haβ-integrin, and knockdown of Haβ-integrin leads to reduced encapsulation of HaCTL3-coated beads. These results indicate that Haβ-integrin serves as a hemocytic receptor of HaCTL3 during the encapsulation reaction. Furthermore, we demonstrate that 20-hydroxyecdysone (20E) treatment dramatically induces the expression of HaCTL3, and knockdown of the 20E receptor (EcR)/ultraspiracle (USP), abrogates this response. Overall, this study provides the first evidence of the presence of a hemocytic receptor (Haβ-integrin), that interacts with the PRR HaCTL3 to facilitate encapsulation reaction in insects and demonstrates the regulation of this process by the steroid hormone ecdysone.

    Abstract Growth-blocking peptide (GBP) is an insect cytokine that stimulates plasmatocyte adhesion, thereby playing a critical role in encapsulation reaction. It has been previously demonstrated that GBP-binding protein (GBPB) is released upon oenocytoid lysis in response to GBP and is responsible for subsequent clearance of GBP from hemolymph. However, current knowledge about GBPB is limited and the mechanism by which insects increase GBPB levels to inactivate GBP remains largely unexplored. Here, we have identified one GBP precursor (HaGBP precursor) gene and two GBPB (namely HaGBPB1 and HaGBPB2) genes from the cotton bollworm, Helicoverpa armigera. The HaGBP precursor was found to be predominantly expressed in fat body, whereas HaGBPB1 and HaGBPB2 were mainly expressed in hemocytes. Immunological analyses indicated that both HaGBPB1 and HaGBPB2 are released from hemocytes into the plasma during the wandering stage. Additionally, 20-hydroxyecdysone (20E) treatment or bead challenge could promote the release of HaGBPB1 and HaGBPB2 at least partly from oenocytoids into the plasma. Furthermore, we demonstrate that the N-terminus of HaGBPB1 is responsible for binding to HaGBP and suppresses HaGBP-induced plasmatocyte spreading and encapsulation. Overall, this study helps to enrich our understanding of the molecular mechanism underlying 20E mediated regulation of plasmatocyte adhesion and encapsulation via GBP-GBPB interaction.

    Abstract BACKGROUND: Wolbachia are obligate endosymbiotic bacteria that infect numerous species of arthropods and nematodes. Wolbachia can induce several reproductive phenotypes in their insect hosts including feminization, male-killing, parthenogenesis and cytoplasmic incompatibility (CI). CI is the most common phenotype and occurs when Wolbachia-infected males mate with uninfected females resulting in no or very low numbers of viable offspring. However, matings between males and females infected with the same strain of Wolbachia result in viable progeny. Despite substantial scientific effort, the molecular mechanisms underlying CI are currently unknown. METHODOLOGY/PRINCIPAL FINDINGS: Gene expression studies were undertaken in Drosophila melanogaster and D. simulans which display differential levels of CI using quantitative RT-PCR. We show that Hira expression is correlated with the induction of CI and occurs in a sex-specific manner. Hira expression is significantly lower in males which induce strong CI when compared to males inducing no CI or Wolbachia-uninfected males. A reduction in Hira expression is also observed in 1-day-old males that induce stronger CI compared to 5-day-old males that induce weak or no CI. In addition, Hira mutated D. melanogaster males mated to uninfected females result in significantly decreased hatch rates comparing with uninfected crosses. Interestingly, wMel-infected females may rescue the hatch rates. An obvious CI phenotype with chromatin bridges are observed in the early embryo resulting from Hira mutant fertilization, which strongly mimics the defects associated with CI. CONCLUSIONS/SIGNIFICANCE: Our results suggest Wolbachia-induced CI in Drosophila occurs due to a reduction in Hira expression in Wolbachia-infected males leading to detrimental effects on sperm fertility resulting in embryo lethality. These results may help determine the underlying mechanism of CI and provide further insight in to the important role Hira plays in the interaction of Wolbachia and its insect host.

In this study, we identified a SRP gene from the cotton bollworm, Helicoverpa armigera, namely Ha-SRP, and studied its precise roles in insect immunity. Expressions of Ha-SRP were upregulated in H. armigera larval hemocytes after injection of Escherichia coli. When the expression of Ha-SRP in H. armigera larval hemocytes was inhibited by dsHa-SRP injection, the transcription of prophenoloxidase genes in hemocytes was repressed, phenoloxidase activity in bacteria-challenged larval hemolymph was significantly decreased, and nodule formation in bacteria-injected larvae was reduced. More importantly, RNAi-treated insects infected with E. coli showed higher bacterial growth in hemolymph compared with infected controls. These results suggest that Ha-SRP gene plays importance roles in H. armigera innate immunity, possibly by mediating prophenoloxidase activation and nodulation response.
