    Abstract As one of the edible jellyfish species, Rhopilema esculentum (Kishinouye, 1891) is a traditional fishery resource and an important economic aquaculture species in China. However, facing the current situation of natural resources exhaustion and problems of breeding population frequent disease, quantity, and quality of seedlings in artificial breeding cannot satisfy the market demand. Temperature and salinity have been considered to play crucial roles in regulating R . esculentum asexual reproduction. This study examined the combined effects by exposing post-preserved R . esculentum podocysts (preserved at 2 ± 1 °C for more than 12 months) to three variable temperatures (simulated temperatures increasing from different starting dates of 14.5 °C on 1 April, 18 °C on 1 May, and 23.2 °C on 1 July, respective to natural levels) and three salinities (20, 25, and 30). Podocyst excystment, the start time of strobilation, duration of strobilation, and cumulative ephyra numbers were tested for 45 days and transfer rates from podocysts to ephyrae were analyzed to assess the most optimal combination of temperature and salinity. The results showed that podocyst excystment and ephyrae production occurred in all treatments. Higher temperature and lower salinity significantly facilitated the podocyst excystment and accelerated the start time of strobilation ( p < 0.05). Significantly greater ephyra numbers were produced with lower salinity (20 and 25) and temperatures increasing from 18 °C on 1 May to natural levels ( p < 0.05). There were significant interactions between temperature and salinity on the cumulative ephyra numbers and transfer rates from podocysts to ephyrae ( p < 0.05). These results suggested that R . esculentum podocysts for long-term preservation at low temperature could be recycled. Temperature and salinity regulation can affect the number and time of R. esculentum seedlings to achieve high production and satisfy the market demand for real-time seedling supply.     Abstract Designing good recirculating aquaculture systems (RASs) is challenging in shrimp aquaculture. In this study, two sets of RASs were constructed using sea cucumber nursery tanks for rearing Litopenaeus vannamei . Recirculating aquaculture was supported by key technologies such as sewage collection and aeration systems adapted to the rectangular tanks and technologies for the removal of sewage, shrimp shells, and dead individuals. Six-hundred and eighty-five thousand juveniles were selected for rearing in the newly constructed RASs, where the average stocking density was 1013 shrimp/m 3 . During the recirculating aquaculture period of 53 days, the water temperature of the tanks was 24–31 °C, the salinity was 25–32‰, the pH was 6.4–8.2, the DO was ≥ 4.9 mg/L, the concentration of total ammonia nitrogen (TAN) was maintained between 0.17 and 4.9 mg/L, the concentration of nitrite nitrogen (NO 2 -N) was between 0.12 and 4.7 mg/L, and the total number of Vibrio bacteria remained between 330 and 9700 cfu/mL. At the end of the experiment, the final average weight of individual shrimp was 13.43 g, and the average yield reached 12.92 kg/m 3 . The great improvement in growth performance marks a breakthrough in RAS technology of shrimp, and it supports the use of an innovative methodology for the retrofitting and utilization of idle sea cucumber nursery tanks.

    Abstract A recirculating aquaculture system (RAS) can reduce water and land requirements for intensive aquaculture production. However, a traditional RAS uses a fixed circulation flow rate for water treatment. In general, the water in an RAS is highly turbid only when the animals are fed and when they excrete. Therefore, RAS water quality regulation technology based on process control is proposed in this paper. The intelligent variable-flow RAS was designed based on the circulating pump-drum filter linkage working model. Machine learning methods were introduced to develop the intelligent regulation model to maintain a clean and stable water environment. Results showed that the long short-term memory network performed with the highest accuracy (training set 100%, test set 96.84%) and F1-score (training 100%, test 93.83%) among artificial neural networks. Optimization methods including grid search, cuckoo search, linear squares, and gene algorithm were proposed to improve the classification ability of support vector machine models. Results showed that all support vector machine models passed cross-validation and could meet accuracy standards. In summary, the gene algorithm support vector machine model (accuracy: training 100%, test 98.95%; F1-score: training 100%, test 99.17%) is suitable as an optimal variable-flow regulation model for an intelligent variable-flow RAS.

    Abstract Sheng Jing Decoction (SJD), as a traditional Chinese medicine prescription, is mainly be used to treat male infertility. However, the pharmacological functions and molecular mechanisms of SJD are poorly understood. In this study, we investigated the functions of SJD on spermatogenesis and sperm motility and explored the potential mechanisms involved. Here, we demonstrated that high, medium, and low doses of SJD are effective in restoring the impairments of the whole body and testicular tissue by cyclophosphamide inducing and to rescue the damage of testicular tissue cells including Sertoli cells and germ cells. SJD can partly restore the decrease in sperm concentration, sperm vitality, sperm motility, and normal sperm morphology rate in oligozoospermic mouse models. Ki67 staining analyses confirm SJD can promote testicular tissue cell proliferation. Real-time RT-PCR analyses also reveal that SJD can upregulate the expression of proliferation-associated gene Lin28a and differentiation-associated genes Kit, Sohlh2, and Stra8. SJD can also reduce the impairment of mitochondrial membrane potential (MMP) and sperm plasma membrane integrity by cyclophosphamide inducing. Our results reveal that SJD is effective in improving both sperm quantity and quality by increasing the sperm concentration, sperm vitality, sperm motility, and normal sperm morphology rate. SJD can promote spermatogenesis by upregulating the expression of the proliferation-associated gene Lin28a and the differentiation-associated genes (Kit, Sohlh2, and Stra8). SJD can sustain MMP and sperm plasma membrane integrity to increase sperm motility.

    Abstract Objective: To investigate the feasibility and advantages of total auricular reconstruction with single big expander at dual plane through incision at remnant ear.
    Methods: 52 patients with microtia were treated. Through incision at remnant ear, one big expander (150 ml) was implanted in the post-auricular area beneath the superficial facial in the hairless area, and above the superficial fascia in the area with hair. In the second stage, total auricular was reconstructed with autologous cartilage framework and expanded skin. Skin graft was not necessary. Another 32 paitents with single big expander above the superficial fascia were used as control. The downward shift distance of expanders, the appearance of auriculocephalic sulcus and complication were recorded in order to assess the effect of these two methods.
    Results: The downward shift distance of expanders in the dual plane group (0.7 ± 0.3) cm were lower than that in the control group (1.3 ± 0.4) cm, showing significant difference (P < 0.05). After a follow-up period of 6-12 months, shallow auriculocephalic sulcus was presented in 5 patients of the dual plane group and 12 patient of the control group. The shallow auriculocephalic sulcus was cut to make it deeper and covered with full skin graft.
    Conclusions: Dual plane skin expansion could reduce the downward shift distance of expander. Adequate skin on the upper part of expander could make satisfactory auriculocephalic sulcus.

    Abstract Abstract Background Over-activation of phosphatidylinositol 3-kinase (PI3K)-AKT-mammalian target of rapamycin (mTOR) signaling pathway is one of important mechanisms to promote castration resistant prostate cancer, the final stage of prostate cancer (PCa). Dysregulation of PP1-meditaed AKT dephosphorylation might contribute to such an event but is not fully understood. As a newly identified tumor suppressor, MIIP exerts its role in various types of cancer but has not been investigated in PCa. Results We first demonstrated that overexpression of migration and invasion inhibitory protein (MIIP) in human PCa cell lines suppresses their growth while knockdown of MIIP does the opposite in vitro. Although MIIP has no effect on the expression of AR and its target genes or the nuclear translocation of AR in AR-positive PCa cells, MIIP overexpression significantly inhibits activation of AKT-mTOR pathway in both AR- positive and negative PCa cells whereas knockdown of MIIP enhances AKT-mTOR signaling. Using Western blot, immunofluorescence co-localization and co-immunoprecipitation analysis, we found that MIIP interacts with PP1α via its C-terminal part but does not affect its protein level. Importantly, silence of PP1α reversed the inhibitory effect of MIIP on AKT phosphorylation and cell growth in PCa cell lines, while MIIP∆C, which is incapable of interacting with PP1α, loses MIIP’s effect, suggesting that MIIP exerts its roles via interaction with PP1α. Further, MIIP overexpression inhibits the growth of both AR- positive and negative PCa xenograft in nude mice. Finally, immunohistochemical staining of PCa tissue microarray showed that MIIP expression level is downregulated in PCa and negatively correlated with Gleason score of PCa. Conclusion We discovered that MIIP is a novel suppressor of oncogenic AKT-mTOR signaling in PCa by facilitating PP1-meditaed AKT dephosphorylation. Our study further emphasized the tumor suppressive role of MIIP and illustrated a novel mechanism.
