Abstract ABSTRACT: The aim of the present study was to evaluate the post-vaccinal reaction to two lentogenic vaccine strains of Newcatle disease virus (NDV) and a recombinant turkey herpesvirus (rHVT) vaccine expressing the fusion glycoprotein of NDV in broiler chickens through histomorphometric and histopathologic analyses of the trachea. The experiment involved 245 chicks housed in randomized blocks with three different enclosures under controlled conditions of temperature, light and ventilation. Each enclosure represented a vaccine strain and was divided into groups according to the administration route. Each block also had its own control group composed of unvaccinated birds. The vaccine strains PHY.LMV.42 (PL42) and La Sota (LS) were selected according to the Intracerebral Pathogenicity Index (ICPI) and the rHVT-NDV Serotype 3 strain (ST3) was selected for representing non-NDV infection. At two, four, seven, 14 and 21 days post vaccination, fragments from the middle third of the trachea were collected and submitted to routine histological processing. For the histomorphometric analysis, the slides were photographed, and the thickness of the tracheal mucosa was measured. Statistical analysis involved two-way ANOVA and Tukey’s post-hoc test with a 5% significance level. For the histopathological evaluation, lesions were described as to the degree of intensity and distribution. At four and 14 days post vaccination with the LS strain administered by the ocular route, the means of thickening of the tracheal mucosa (20.85±7.31μm and 26.97±5.50μm, respectively) were significantly higher (p<0.05) than for all other strains, which was related to the severe histopathological lesions found in this group, characterized by hyperemia, hyperplasia of the mucous glands, moderate deciliation and multifocal lymphohistiocytic inflammatory infiltrate. At 21 days, broiler chickens vaccinated with the ST3 strain showed more discrete lesions and less thickening of the tracheal mucosa (23.23±7.62μm; p<0.05) in comparison with other ...

Abstract When evaluating published studies on the production of platelet-rich plasma (PRP), it is possible to observe that there is no consensus on when blood should be collected and processed. Some perform blood collection before surgery, prior to the administration of drugs, fluids, or any surgical trauma, while others collect blood during the perioperative period. The present objective was to determine the best moment for PRP production based on the platelet count. Blood samples were collected from 20 healthy female dogs at two distinct moments: before surgery, prior to anesthesia and fluid therapy (M0), and later, in the operation room, after administration of the anesthetics and intravenous fluids (M1). To obtain PRP, blood was centrifuged at 1200 rpm for ten minutes, then at 1600 rpm for ten minutes. Results showed that the blood collected at M1 had a 29% decrease in platelet count compared to M0 (highly significant statistical difference). It is recommended that blood collection, which is an essential step for producing platelet-rich plasma, shall be done during the pre-operative period.

Abstract ABSTRACT: This work aimed to evaluate the effect of platelet-rich plasma (PRP) on advancement skin flaps in dogs regarding improvement of vascularization, with focus on increasing its viable area, since there are reports that it is a potential angiogenesis stimulator. The experimental group was composed of eight adult bitches, in which two advancement skin flaps were made in the ventral abdominal region. No product was applied in the control flap (CF), while PRP was used in the contralateral flap, called treated flap (TF). The areas were clinically evaluated every two days until the 7th postoperative day regarding skin color and presence of necrosis. At 10 days, both flaps were removed and submitted to histological examination and blood vessel morphometry. The vessels counted in each group were statistically analyzed by the F-test at 1% probability. Results showed no significant difference in macroscopic changes in the wound, or CF and TF vascularization, thus suggesting that PRP gel did not improve advancement skin flap angiogenesis in bitches under the experimental conditions in which this research was developed.

Abstract ABSTRACT: The nurse shark, Ginglymostoma cirratum (Bonnaterre, 1778) is one of the most studied species of elasmobranchs. However, the knowledge of their reproductive biology is still relatively rare, particularly in the western South Atlantic. This study aimed to describe the morphology of the uterus and the ovary of G. cirratum, based on specimens caught off at the Fortaleza/CE coast, northeast Brazil. Samples were collected from September 2012 to June 2013, from regular landings of artisanal fishing, which commercialize this species freely. A total of ten females were collected. The methodologies followed for analyzing the ovaries and uterus of those females included both macroscopic and histological analysis. G. cirratum has internal type ovary morphology, with invaginations of connective tissue, which defines compartments and separate oocyte groups in ovigerous lots. The epithelium lining the ovary changes from simple columnar ciliated in the area without ovigerous lots, which turns into a simple cubic epithelium in the coating portion of the epigonal organ where ovarian tissue is absent. The uterine mucosa has secretory cells denoted by Alcian Blue staining, indicating the production of mucopolysaccharides, even in immature individuals. This lecithotrophic shark has a uterine vascularized mucosa that is one characteristic of viviparous elasmobranch species.

Abstract ABSTRACT: Equine coital exanthema is a venereal infectious disease poorly reported in horses in Brazil and was never described in the northeastern region of the country. This work aims to describe the clinical and pathological aspects of an outbreak of equine coital exanthema caused by equid alphaherpesvirus 3, occurred in a herd of horses at the semiarid region of the State of Rio Grande do Norte. Main clinical signs consisted of anorexia, hiporexia, fibrinous or purulent secretion in the penis mucosa and vagina. Two mares presented mild to minimal lesions that consisted of scars in the mucosa of the vagina and in the perivulvar region. In a stallion the disease consisted of severe, multifocal, umbilicated-exanthematous ulcers of approximately 1cm in diameter on the penis mucosa. Other areas where ulcers and crusts were focally observed included the skin of the scrotum and on the lips and mucocutaneous junctions of the oral cavity. Histologically, the main lesion consisted of multifocal severe ulcerative and fibrinous necrotizing balanoposthitis and mild multifocal necrotizing, lymphocytic dermatitis in the lips and scrotum. The equide alphaherpesvirus 3 DNA was amplified in blood samples and penis mucosa using the PCR technique. This is the first report of molecular diagnosis of equine coital exanthema affecting horses in northeastern Brazil. Further studies should be carried out in order to investigate the epidemiology and the importance of this herpetic disease in the country.

Abstract ABSTRACT: This study aimed to perform phytochemical analysis and to test the antimicrobial activity of the crude hydroalcoholic extract obtained from the leaves of Sphagneticola trilobata. Classes of secondary metabolites present in the extract were identified through phytochemical screening using analytical thin-layer chromatography. Antimicrobial activity was evaluated by testing cultures of Staphylococcus aureus, S. epidermidis, Staphylococcus spp., Escherichia coli, Serratia marcescens, Enterococcus faecalis, Pseudomonas aeruginosa, Salmonella Typhimurium, and Klebsiella pneumoniae isolated from human skin and those of Staphylococcus spp. isolated from dog skin using the broth microdilution method. In the phytochemical screening, classes of anthracenic derivatives and mono-, sesqui-, and diterpenes were identified. Colorimetric analysis showed total phenol and total flavonoid contents of 21.7 ± 0.009 mg of gallic acid equivalents per gram of sample and 0.23 ± 0.005 mg of catechin equivalents per gram of sample, respectively. Microbiological analysis revealed that the hydroalcoholic extract of S. trilobata exhibited antimicrobial activity against cultures of Staphylococcus spp., E. coli, S. marcescens, and E. faecalis isolated from human skin and those of Staphylococcus spp. isolated from dog skin. Thus, crude hydroalcoholic extract of leaves of S. trilobata contained flavonoids and terpenoids as secondary metabolites, which contributed to its antimicrobial activity against skin bacteria isolated from different sources.

Abstract The tongue is regarded as a key organ for bats, since the specialized characters are used for taxonomic and systematic purposes, cladistic studies, and elucidation of patterns of eating habits. This study aimed to characterize morphologically the tongue in the species Artibeus lituratus, Artibeus planirostris, Artibeus obscurus, and Dermanura cinerea, and to report the first evidence of melanin in the lingual epithelium of bats. We used 20 specimens collected in the municipalities of Vitória de Santo Antão and Sirinhaém, in Pernambuco State, Brazil. The tongues were extracted and submitted to routine histological techniques, stained with hematoxylin and eosin (HE) and Fontana-Masson. In the four species, the keratinized stratified pavement epithelium was observed, but with the presence of melanin granules on the basal layer only in the species A. lituratus, A. planirostris, and A. obscurus, evidenced through the Fontana-Masson staining technique. The melanin found in the tongue of these species is characterized as the first record for the order Chiroptera and it may be related to oral asepsis, and/or inhibition of toxic substances observed in plants. Therefore, there is a need for further studies, aiming to improve our knowledge of the function of pigment in this organ. Keywords: Artibeus; Dermanura; melanin; bats; lingual morphology.
