    Abstract Transfer RNA (tRNA) modifications impact the structure and function of tRNAs, thus affecting the efficiency and fidelity of translation. In the opportunistic pathogen Pseudomonas aeruginosa translational regulation plays an important but less defined role in adaptation to changing environments. In this study, we have explored tRNA modifications in P. aeruginosa through LC-MS/MS approaches. Neutral loss scanning (NLS) demonstrated the potential to identify previously unknown modifications, whereas multiple reaction monitoring (MRM) was able to detect modifications with high specificity and sensitivity. In this study, the MRM-based external calibration method allowed for quantification of the four canonical and 32 modified ribonucleosides, out of which 21 tRNA modifications were quantified in the total tRNA pool of P. aeruginosa PA14. We also purified the single tRNA isoacceptors tRNA-ArgUCU, tRNA-LeuCAA, and tRNA-TrpCCA and determined their specific modification patterns, both qualitatively and quantitatively. Deeper insights into the nature and dynamics of tRNA modifications in P. aeruginosa should pave the way for further studies on post-transcriptional gene regulation as a relatively unexplored molecular mechanism of controlling bacterial pathogenicity and mode of growth.

    Abstract Pseudomonas aeruginosa is a major cause of nosocomial infections and also leads to severe exacerbations in cystic fibrosis or chronic obstructive pulmonary disease. Three intertwined quorum sensing systems control virulence of P. aeruginosa, with the rhl circuit playing the leading role in late and chronic infections. The majority of traits controlled by rhl transcription factor RhlR depend on PqsE, a dispensable thioesterase in Pseudomonas Quinolone Signal (PQS) biosynthesis that interferes with RhlR through an enigmatic mechanism likely involving direct interaction of both proteins. Here we show that PqsE and RhlR form a 2:2 protein complex that, together with RhlR agonist N-butanoyl-L-homoserine lactone (C4-HSL), solubilizes RhlR and thereby renders the otherwise insoluble transcription factor active. We determine crystal structures of the complex and identify residues essential for the interaction. To corroborate the chaperone-like activity of PqsE, we design stability-optimized variants of RhlR that bypass the need for C4-HSL and PqsE in activating PqsE/RhlR-controlled processes of P. aeruginosa. Together, our data provide insight into the unique regulatory role of PqsE and lay groundwork for developing new P. aeruginosa-specific pharmaceuticals.

    Abstract On January 1, 2015 a statutory minimum wage was introduced in Germany. The Arbeitsmarktspiegel shows several important developments of the German labour market since its introduction. While overall employment in Germany continues to show a positive trend, the number of exclusively marginal part-time employees drops markedly at the turn of the year. This effect is strongest in industries and regions with a low average wage level. A little more than half of the decrease can be explained by persons changing directly into exclusively employment subject to social security. Yet, the data also show an increased amount of exits out of the labour market or at least out of the group of persons covered by the data. Transitions into unemployment play a rather secondary role in explaining the decline of marginal employment. The number of employees receiving additional benefits according to Social Code Book II slightly decreases after the introduction of the minimum wage. For this group, both the transitions into employment without benefit receipt as well as into exclusive benefit receipt are slightly increased.

    Zum 1. Januar 2015 wurde in Deutschland ein gesetzlicher Mindestlohn eingeführt. Der Arbeitsmarktspiegel beschreibt vor dem Hintergrund dieser Einführung wichtige Entwicklungen auf dem deutschen Arbeitsmarkt. Während die Gesamtbeschäftigung in Deutschland weiterhin einem positiven Trend folgt, sank die Zahl der ausschließlich geringfügig Beschäftigten zum Jahreswechsel 2014/2015 deutlich. Dieser Rückgang war in Branchen und Regionen mit niedrigem durchschnittlichem Lohnniveau am höchsten. Etwas mehr als die Hälfte des Rückgangs ist dadurch zu erklären, dass die betroffenen Personen direkt in ausschließlich sozialversicherungspflichtige Beschäftigung übergingen. Daneben verließen aber auch vermehrt Personen den Arbeitsmarkt bzw. tauchen in den Daten nicht länger auf. Übergänge in reine Arbeitslosigkeit spielen bei der Erklärung insgesamt eine eher untergeordnete Rolle. Die Zahl der Beschäftigten mit Bezug von SGB-II-Leistungen geht nach der Einführung des Mindestlohns leicht zurück. In geringem Umfang steigen zum Jahreswechsel die Übergänge sowohl in reine Beschäftigung ohne weiteren Leistungsbezug als auch in reinen Leistungsbezug unter Wegfall der Beschäftigung.

    Abstract Die zweite Ausgabe des Arbeitsmarktspiegels beschreibt anhand aktueller Daten bis einschließlich Januar 2016 wichtige Entwicklungen auf dem deutschen Arbeitsmarkt vor dem Hintergrund der Einführung des gesetzlichen Mindestlohns in Deutschland. Die Gesamtbeschäftigung steigt in 2015 weiter an. Die Zahl der ausschließlich geringfügig Beschäftigten, welche zum Jahreswechsel 2014/2015 saisonbereinigt um etwa 93.000 Personen sank, weist bis Ende des dritten Quartals 2015 eine rückläufige Tendenz auf. In den Wintermonaten könnte der Tiefpunkt dieser negativen Entwicklung erreicht sein. Ausgabe 2 des Arbeitsmarktspiegels zeigt erstmals, dass der Großteil der Übergänge aus ausschließlich geringfügiger in ausschließlich sozialversicherungspflichtige Beschäftigung im selben Betrieb erfolgt. Für beschäftigte SGB-II-Leistungsbezieher zeigt sich, dass nach Abklingen der ersten Reaktion auf die Einführung des Mindestlohns im Verlauf des Jahres 2015 die Anzahl der Übergänge in Beschäftigung ohne Leistungsbezug weiter zunimmt. Den Sprung aus der Bedürftigkeit von staatlichen Leistungen schaffen dabei vor allem sozialversicherungspflichtig beschäftigte Leistungsbezieher. Detailliert untersucht werden außerdem Beschäftigungsentwicklungen differenziert nach Geschlecht und Altersgruppen. Die erste Ausgabe des Arbeitsmarktspiegels erschien im Januar 2016 als IAB-Forschungsbericht 01/2016 (vom Berge et al., 2016). Dieser Forschungsbericht stellt eine aktualisierte und inhaltlich modifizierte Version dar.

    The second edition of the Arbeitsmarktspiegel describes important developments on the German labour market since the introduction of the minimum wage in Germany with latest data until January 2016. Overall employment continues to increase in 2015. The number of exclusively marginal employees, which decreased by a seasonally adjusted estimate of 93,000 persons at the turn of the year 2014/2015, shows a declining trend by the end of the third quarter of 2015. The bottom of this negative trend could be reached by winter. For the first time the second edition of the Arbeitsmarktspiegel shows that most of the transitions from exclusively marginal employment to exclusively employment covered by social security took place in the same establishment. With regard to employees receiving additional benefits according to Social Code Book II, the number of transitions into employment without benefit receipt further increases in the course of 2015 after their initial reaction following the introduction of the minimum wage. Especially those benefit recipients whose parallel employment is subject to social security are able to end eligibility for government benefits. Furthermore, the development of employment is examined in more detail by sex and age groups. The first edition of the Arbeitsmarktspiegel was released as IAB-Forschungsbericht 01/2016 (vom Berge et al., 2016) in January 2016. This research report represents an updated and substantially modified version.
