    Abstract Gizzerosine [2-amino-9-(4-imidazolyl)-7-azanonanoic acid] is a toxic amino acid formed from histamine and lysine at high temperatures, and may be present in foodstuffs (e.g., fishmeal and meat-bone meal) for animals including cats and dogs. Here we developed a simple, rapid, sensitive, specific, and automated method for the analysis of gizzerosine in foodstuffs by high-performance liquid chromatography (HPLC) involving pre-column derivatization with o-phthaldialdehyde (OPA) in the presence of N-acetylcysteine (instead of the usual 2-mercaptoethanol or ethanethiol reagent). OPA reacted immediately (within 1 min) with gizzerosine in an autosampler at room temperatures (e.g., 20-25 °C), and their derivative was directly injected into the HPLC column. The highly fluorescent gizzerosine-OPA derivative was well separated from the OPA derivatives of all natural amino acids known to be present in physiological fluids (e.g., plasma), proteins and foodstuffs, and was detected at an excitation wavelength of 340 nm and an emission wavelength of 450 nm. The total time for chromatographic separation (including column regeneration) was 20 min per sample rather than 40 min and longer in previous HPLC methods. The detection limit for gizzerosine was at least 6 pmol/ml in an assay solution (HPLC vial) or at least 0.09 pmol per injection into the HPLC column. The analysis of gizzerosine was linear between 1 and 100 pmol per injection. When gizzerosine was extracted from foodstuffs, its detection limit was at least 875 pmol/g foodstuff or at least 0.21 mg/kg foodstuff. Our routine HPLC technique does not require any cleanup of samples or the OPA derivatization products (including the OPA-gizzerosine adduct), and is applicable for the analysis of gizzerosine in both foodstuffs and animal tissues.

    Abstract To mitigate systematic errors in magnetic resonance fingerprinting (MRF), the precomputed dictionary is usually computed with minimal granularity across the entire range of tissue parameters. However, the dictionary grows exponentially with the number of parameters increase, posing significant challenges to the computational efficiency and matching accuracy of pattern-matching algorithms. Existing works, primarily based on convolutional neural networks (CNN), focus solely on local information to reconstruct multiple parameter maps, lacking in-depth investigations on the MRF mechanism. These methods may not exploit long-distance redundancies and the contextual information within voxel fingerprints introduced by the Bloch equation dynamics, leading to limited reconstruction speed and accuracy. To overcome these limitations, we propose a novel end-to-end neural network called the Local and Global Vision Transformer (LG-ViT) for MRF parameter reconstruction. Our proposed LG-ViT employs a multi-stage architecture that effectively reduces the computational overhead associated with the high-dimensional MRF data and the transformer model. Specifically, a local Transformer encoder is proposed to capture contextual information embedded within voxel fingerprints and local correlations introduced by the interconnected human tissues. Additionally, a global Transformer encoder is proposed to leverage long-distance dependencies arising from shared characteristics among different tissues across various spatial regions. By incorporating MRF physics-based data priors and effectively capturing local and global correlations, our proposed LG-ViT can achieve fast and accurate MRF parameter reconstruction. Experiments on both simulation and in vivo data demonstrate that the proposed method enables faster and more accurate MRF parameter reconstruction compared to state-of-the-art deep learning-based methods.

    Abstract Over the past few decades, unbiased approaches such as genetic screening and protein affinity purification have unveiled numerous proteins involved in DNA double-strand break (DSB) repair and maintaining genome stability. However, despite our knowledge of these protein factors, the underlying molecular mechanisms governing key cellular events during DSB repair remain elusive. Recent evidence has shed light on the role of non-protein factors, such as RNA, in several pivotal steps of DSB repair. In this review, we provide a comprehensive summary of these recent findings, highlighting the significance of ribosomal RNA (rRNA) as a critical mediator of DNA damage response, meiosis, and mitosis. Moreover, we discuss potential mechanisms through which rRNA may influence genome integrity.
