    Abstract Rosa rugosa Purple branch is a rare continuously flowering variety of R. rugosa and also is one of main cultivar varieties of R. rugosa in China. Although R. rugosa has been successfully used in the field of cosmetics, there are also many reports on its chemical components. The chemical composition and application value in cosmetics of R. rugosa Purple branch are still limited. The purpose of this study was to investigate the chemical components in the petals of R. rugosa Purple branch as well as evaluate their skincare and antibacterial potentials through DPPH radical scavenging, tyrosinase inhibition, elastase inhibition and antibacterial bioassays. Two new compounds, isorhamnetin-3-O-(2"-O-galloyl)-β-D-glucopyranoside (1), kaempferol-3-O-β-D-glucopyranosyl-(1→2)-(6''-O-trans-p-coumaroyl)-β-D-glucopyranoside (2) and thirteen known compounds (3-15) were isolated and identified from the petals by bioassay-guided fractionation. All the compounds were isolated and reported from R. rugosa Purple branch for the first time, in which twelve of them are discovered from R. rugosa for the first time. The tyrosinase inhibitory activities of 1, 2, rosarugoside B (3), kaempfero-3-O-β-D-glucopyranosyl(1→2)-β-D-glucopyranoside (4), astragalin-2'',6''-di-O-gallate (8), quercetin-3-O-β-D-glucopyranosyl(1→2)-β-D-glucopyranoside (10), 2-phenylethyl-β-D-glucopyranoside (13), 2-phenylethyl-6-O-α-L-arabinofuranosyl-β-D-glucopyranoside (14), and 2-phenylethyl-D-rutinoside (15) were firstly reported here, in which 8 showed strong tyrosinase inhibitory activity with IC₅₀ of 187.32 ± 0.18 μg/mL. There were eight compounds (1, 6-12) showed strong antioxidant activity, with the scavenging rates of DPPH radical ranging from 88.59% to 94.30% and the IC₅₀ ranging from 4.22 to 13.85 μg/mL, among them, 1 and quercetin-3-O-(6''-O-Z-p-coumaroyl)-β-D-glucopyranoside (11) exhibited antioxidant activity for the first time. 3 exhibited moderate elastase inhibitory activity with IC₅₀ of 148.67 ± 0.11 μg/mL. In addition, seven compounds (2, 5-8, 10-11) showed varying degrees of antibacterial activity. These results indicated that R. rugosa Purple branch had anti-aging, whitening and antibacterial activities, which can be further applied to the development of functional cosmetics.

    Abstract To achieve sensitive detection of low-content microRNA, photoelectrochemical/electrochromic dual-mode sensor with intrinsically low background signal has been developed, but the two detection modules are usually designed with a series-connected structure, which may cause signal interference and thus affect the detection reliability. To solve the above problems, a decoupled dual-mode bioassay for sensitive miRNA-21 detection with high reliability is constructed in this work, by selecting two capacitors to realize parallel amplification for the two detection modules, supplemented with a 3D DNA nanoring photoelectrode signal amplification strategy. The complete decoupling of the two detection modes, photoelectrochemical and electrochromic, as well as the use of digital multimeter, improves the reliability and accuracy of the sensor, and also frees it from dependence on electrochemical workstation, making detection more intuitive and faster. With simple structure, low cost, good reproducibility, high sensitivity, and easy operation, the capacitor-parallel-amplified decoupled photoelectrochemical/electrochromic dual-mode bioassay has broad application prospect in on-site point-of-care detection of diseases and low-cost clinical diagnosis. The design idea of decoupled dual-mode detector can also be extended to the construction of other dual-mode methods.

    Abstract A uniform illumination over a screen is crucial for terahertz imaging. As such, conversion from a Gaussian beam to a flattop beam becomes necessary. Most of the current beam conversion techniques rely on bulky multi-lens systems for collimated input and operate in the far-field. We present a single metasurface lens to efficiently convert a quasi-Gaussian beam from the near-field region of a WR-3.4 horn antenna to a flattop beam.     Abstract The objective of this study was to evaluate the efficacy and safety of Chinese patent medicine compared with western medicine in the treatment of Alzheimer's disease using the Network Meta-analysis. This study retrieved relevant studies from 7 databases, and the retrieval time was from the establishment of each database to June 2022. After the screening, data extraction, and quality assessment, 47 studies were finally analyzed, involving 11 Chinese patent medicines. The results demonstrated that Chinese patent medicine intervention was superior to oral western medicine treatment in improving the patient's condition as assessed by the Mini-mental State Examination (MMSE), Activities of Daily Living (ADL), effective rate, and Alzheimer's Disease Assessment Scale-Cognitive section (ADAS-Cog). Particularly, the effect of Chinese patent medicine combined with western medicine intervention was prominent. Meanwhile, Chinese patent medicine intervention in AD did not significantly increase the risk of adverse reactions. The results of the Network Meta-analysis demonstrated that Chinese patent medicine combined with western medicine had statistically significant differences in the MMSE score, ADL score, effective rate, and ADAS-Cog score, compared with both western medicine alone and Chinese patent medicine alone. In terms of adverse reactions, the difference between Chinese patent medicine intervention and simple oral western medicine was statistically significant. The results of further ranking probability analysis demonstrated that Chinese patent medicine combined with western medicine intervention ranked first in terms of MMSE, ADL, effective rate, and ADAS-Cog. Additionally, oral Chinese patent medicine intervention alone ranked first in reducing adverse reactions. In the funnel plots of the MMSE, ADL, and effective rate, most studies were symmetrically distributed on both sides of the midline, where small sample effects and publication bias might exist to some extent. However, this conclusion still needs to be combined with clinical syndrome differentiation and treatment, and more large-sample, multi-center, high-quality studies are needed for further verification.

    Abstract Porcine epidemic diarrhea virus (PEDV) is a re-emerging enteropathogenic coronavirus that causes high mortality in neonatal piglets. The addition of trypsin plays a crucial role in the propagation of PEDV, but also increases the complexity of vaccine production and increases its cost. Previous studies have suggested that the S2' site and Y976/977 of the PEDV spike (S) protein might be the determinants of PEDV trypsin independence. In this study, to achieve a recombinant trypsin-independent PEDV strain, we used trypsin-dependent genotype 2 (G2) PEDV variant AJ1102 to generate three recombinant PEDVs with mutations in S (S2' site R894G and/or Y976H). The three recombinant PEDVs were still trypsin dependent, suggesting that the S2' site R894 and Y976 of AJ1102 S are not key sites for PEDV trypsin dependence. Therefore, we used AJ1102 and the classical trypsin-independent genotype 1 (G1) PEDV strain JS2008 to generate a recombinant PEDV carrying a chimeric S protein, and successfully obtained trypsin-independent PEDV strain rAJ1102-S2'

    Abstract Porcine epidemic diarrhea virus (PEDV), a re-emerging enteropathogenic coronavirus, has become the predominant causative agent of lethal diarrhea in piglets, resulting in huge economic losses in many countries. Furthermore, the rapid variability of this virus has increased the emergence of novel variants with different pathogenicities. In this study, 633 fecal samples collected from diarrheic piglets in China during 2017-2019 were analyzed, and 50.08% (317/633) of these samples were PEDV-positive. The full-length spike (S) genes of 36 samples were sequenced, and a genetic evolution analysis was performed. The results showed that thirty S genes belonged to the GII-a genotype and six S genes belonged to the GII-b genotype. From the PEDV-positive samples, one strain, designated ECQ1, was successfully isolated, and its full-length genome sequence was determined. Interestingly, ECQ1 is a recombinant PEDV between the GII-a (major parent) and GII-b (minor parent) strains, with recombination occurring in the S2 domain of the S gene. The pathogenicity of ECQ1 was assessed in 5-day-old piglets and compared with that of the strain EHuB2, a representative of GII-a PEDV. Although both PEDV strains induced similar fecal viral shedding in the infected piglets, ECQ1 exhibited lower pathogenicity than did EHuB2, as evidenced by reduced mortality and less severe pathological changes in the intestines. These data suggest that PEDV strain ECQ1 is a potential live virus vaccine candidate against porcine epidemic diarrhea.
