    Abstract Abstract Background Acute kidney injury (AKI) is a serve and harmful syndrome in the intensive care unit. Comparing to the patients with AKI stage 1/2, the patients with AKI stage 3 have higher in-hospital mortality and risk of progression to chronic kidney disease. The purpose of this study is to develop a prediction model that predict whether patients with AKI stage 1/2 will progress to AKI stage 3. Methods Patients with AKI stage 1/2, when they were first diagnosed with AKI in the Medical Information Mart for Intensive Care, were included. We used the Logistic regression and machine learning extreme gradient boosting (XGBoost) to build two models which can predict patients who will progress to AKI stage 3. Established models were evaluated by cross-validation, receiver operating characteristic curve, and precision–recall curves. Results We included 25,711 patients, of whom 2130 (8.3%) progressed to AKI stage 3. Creatinine, multiple organ failure syndromes were the most important in AKI progression prediction. The XGBoost model has a better performance than the Logistic regression model on predicting AKI stage 3 progression. Thus, we build a software based on our data which can predict AKI progression in real time. Conclusions The XGboost model can better identify patients with AKI progression than Logistic regression model. Machine learning techniques may improve predictive modeling in medical research.

    Abstract The low retention rate and lack of long-term blood donors are still worldwide problems, and the fostering and retaining of more blood donors has posed an urgent problem that requires a solution. The purpose of this study was to explore the role of functional motivation and blood donor stage in the future intention to donate blood. Three sites in China provided 252 blood donors who filled out a questionnaire including the volunteer function inventory, their future intention to donate blood, number of blood donations and demographic information. Based on a functional motivation approach, we investigated the motivations that influence unpaid blood donors’ future intention through qualitative interview methods. The results showed that blood donor stage moderated the relationship between values motivation and future intention to donate blood. More specifically, the predictive effect of values motivation on future intention decreased gradually from new donors to regular donors. Our study found that the effect of various motivations on blood donors’ intention was conditional; this effect changed with the development of blood donor behavioral stages. The results are significant in practice for establishing effective intervene strategies in maintaining blood donors.

    Abstract Microinjection molding is a novel frontier polymer processing strategy different from conventional ones. In this paper, three different cavity-sizes of micro-mold tools were firstly fabricated, and the influences of micro-mold cavity dimension on the phase morphology structure, crystallization and orientation, and mechanical performance of the microinjection molded polylactic acid (PLA)/polycaprolactone (PCL) blend microparts were carefully investigated accordingly. The results show that the reduction of the cavity size would result in much higher shear stress field and cooling temperature gradient, which is advantageous to the fibrillation and orientation of PCL-dispersed phase. Consequently, with decreasing the micro-mold cavity dimension from length 26 mm to 15 mm, the interfacial compatibility is improved, significantly increasing number of PCL fibers with smaller diameter are in situ formed in PLA matrix and their orientation degree also obviously increases, which is verified by SEM and 2D-WAXD measurements. The Differential Scanning Calorimetry (DSC) analysis shows that the decrease in cavity dimension causes the enhancement of PLA crystallization property due to shear-induced crystallization, which is reflected by the decreasing PLA cold crystallization temperature and increasing PLA crystallinity (almost doubling that of conventional macropart). As a result, the dynamic/static mechanical property measurements exhibit that with decreasing the cavity size, the storage modulus, and the loss modulus of PLA/PCL blend micropart increase, and the corresponding tensile strength, elongation at break, and Young’s modulus also present an obviously increasing tendency. The related investigations would provide some new spaces and insights for realization of high-performance of PLA/PCL blend micropart.

    Abstract Based on the self-determination theory (SDT), this study used a mixed-methods (i.e., quantitative and qualitative approaches) design to explore the role of basic psychological need satisfaction (BPNS) played in sustained volunteering. Quantitative analysis of 803 college student volunteers revealed that competence and relatedness need satisfaction had significant associations with sustained volunteering, while autonomy need satisfaction did not. Furthermore, latent profile analyses identified five profiles of BPNS: low (Profile 1), relatively low (Profile 2), moderate (Profile 3), low autonomy-high competence and relatedness (Profile 4), and high (Profile 5). Volunteers in Profile 4 and Profile 5 reported higher sustained volunteering than those in other profiles. Subsequent qualitative synthesis of interview data from 33 college student volunteers found that competence need satisfaction (45.58%) was mentioned most frequently among the factors promoting sustained volunteering, then followed by relatedness (27.43%) and autonomy need satisfaction (11.06%). These findings highlight the important role of BPNS, especially competence and relatedness need satisfaction, in promoting college students’ long-term volunteering.

    Abstract Background. Erythronium japonicum Decne (Liliaceae) is an early spring ephemeral with an underground initial floral differentiation stage. The flowering mechanism is crucial in ornamental plants due to the associated economic value. Therefore, this study is aimed at exploring the metabolic landscape during floral differentiation, including flower primordium, perianth, stamen, and the pistil differentiation period, in E. japonicum coupled with a conjoint analysis of the metabolome and transcriptome. Using ultraperformance liquid chromatography-tandem mass spectrometry, we identified 586 metabolites from 13 major metabolite classes. Comparative metabolomics between different floral developmental stages revealed several abundant metabolites during the respective phases. Upaccumulation of p-coumaroylputrescine, scopoletin, isorhoifolin, cosmosiin, genistin, and LysoPC 15 : 0 emphasized the significance of these compounds during flower development. Furthermore, previously identified DEGs, viz., EARLY FLOWERING 3, Flowering locus K, PHD finger-containing protein, and zinc finger SWIM domain-containing protein for floral differentiation, depicted a high correlation with lipid, flavonoid, and phenolics accumulation during floral developmental stages. Conclusions. Together, the results improve our interpretation of the underground floral development in E. japonicum.

    Abstract Erythronium japonicum Decne (Liliaceae) flowers in early spring after overwintering. Its sexual reproduction process includes an underground development process of floral organs, but the underlying molecular mechanisms are obscure. The present study is aimed at exploring the transcriptional changes and key genes involved at underground floral developmental stages, including flower primordium differentiation, perianth differentiation, stamen differentiation, and pistil differentiation in E. japonicum. Multistage high-quality transcriptomic data resulted in identifying putative candidate genes for underground floral differentiation in E. japonicum. A total of 174,408 unigenes were identified, 28,508 of which were differentially expressed genes (DEGs) at different floral developmental stages, while only 44 genes were identified with conserved regulation between different stages. Further annotation of DEGs resulted in the identification of 270 DEGs specific to floral differentiation. In addition, ELF3, PHD, cullin 1, SE14, ZSWIM3, GIGNATEA, and SERPIN B were identified as potential candidate genes involved in the regulation of floral differentiation. Besides, we explored transcription factors with differential regulation at different developmental stages and identified bHLH, FAR1, mTERF, MYB-related, NAC, Tify, and WRKY TFs for their potential involvement in the underground floral differentiation process. Together, these results laid the foundation for future molecular works to improve our understanding of the underground floral differentiation process and its genetic regulation in E. japonicum.

    Abstract Abstract. Background:. Differential diagnosis of active tuberculosis (ATB) and latent tuberculosis infection (LTBI) has been a challenge for clinicians in high TB burden countries. The purpose of this study was to improve the accuracy of differential diagnosis of ATB and LTBI by using fluorescent immunospot (FluoroSpot) assay to detect specific Th1 cell immune responses. The novel mycobacterium tuberculosis (MTB) latency-associated antigens Rv1733c and synthetic long peptides derived from Rv1733c (Rv1733c SLP) were used based on virulence factors early secreting antigen target-6 (ESAT-6) and culture filtrate protein-10 (CFP-10). Methods:. Fifty-seven ATB cases, including 20 pathogen-confirmed ATB and 37 clinically diagnosed ATB, and 36 LTBI cases, were enrolled between January and December 2017. FluoroSpot assay was used to detect the interferon γ (IFN-γ) and interleukin 2 (IL-2) secreted by the specific T cells after being stimulated with MTB virulence factors ESAT-6 and CFP-10, MTB latency-associated antigens Rv1733c and Rv1733c SLP. The receiver operating characteristic (ROC) curve was used to define the best cutoff value of latency-associated antigens in the use of differentiating ATB and LTBI. The sensitivity, specificity, predictive value, and likelihood ratio of ESAT-6 and CFP-10-FluoroSpot combined with latency-associated antigen in the differential diagnosis of ATB and LTBI were also calculated. Results:. Following the stimulation with Rv1733c and Rv1733c SLP, the frequency of single IL-2-secreting T cells stimulated by Rv1733c SLP had the largest area under the ROC curve, which was 0.766. With a cutoff value of 1 (spot-forming cells [SFCs]/2.5 × 105 peripheral blood mononuclear cells) for frequency, the sensitivity and specificity of distinguishing ATB from LTBI were 72.2% and 73.7%, respectively. ESAT-6 and CFP-10-FluoroSpot detected the frequency and proportion of single IFN-γ-secreting T cells; the sensitivity and specificity of distinguishing ATB from LTBI were 82.5% and 66.7%, respectively. ...

    Abstract The thermal degradation behaviors and reaction kinetics of medical waste infusion bag (IB) and nasal oxygen cannula (NOC) were investigated under inert atmosphere with the heating rates of 5, 10, 15, and 25 K·min −1 . Ozawa–Flynn–Wall (OFW), Kissinger–Akahira–Sunose (KAS), and Friedman were employed to estimate the activation energy. Coats–Redfern and Kennedy–Clark methods were adopted to predict the possible reaction mechanism. The results suggested that the reaction mechanism of IB pyrolysis was zero-order, and that of NOC pyrolysis was concluded that zero-order for the first stage and three-dimensional diffusion Jander equation for the second stage. Based on the kinetic compensation effect, the reconstructed reaction models for IB and NOC pyrolysis were elaborated by introducing adjustment functions. The results indicated that the reconstructed model fitted well with the experimental data. The results are helpful as a reference and provide guidance for the determination of IB and NOC degradation behaviors and the simulation of parameters.
