    Abstract Enzyme-linked immunospot (ELISPOT) is one of the most important methods to measure the number of specific cells by detecting protein secretion at a single-cell level. However, traditional ELISPOT based on enzyme-substrate color development can only detect one target. Therefore, scientists developed multiple-target ELISPOT based on enzyme-substrate coloring. Besides, FluoroSPOT that can detect 2-4 fluorescent signals are developed. Nevertheless, the maximum detection targets of multiple-target ELISPOT and FluoroSPOT are around 4, and the signal amplification system can be further optimized. Fluorescence-based oligo-linked immunospot (FOLISPOT), which utilized DNA-barcoded antibodies to provide a highly multiplexed method with signal amplification, was developed to detect multiple targets simultaneously. In this method, multiple targets can be detected in one round and multiple rounds of detection can be conducted, and thus a large number of targets can be detected. Besides, signal amplification is achieved by DNA complementary pairing and modular orthogonal DNA concatemers, and thus cells secreting limited amounts of proteins can be detected. According to the studies, FOLISPOT can detect more spots than ELISPOT and can detect targets that are undetectable by ELISPOT. Furthermore, FOLISPOT can be utilized to detect more than 6 targets, by allowing sequential detection of multiple targets in one round and sequential detection in multiple rounds.

    Abstract Display rules specify socially appropriate facial expressions in a given situation. However, managing emotions for such a social adaption sometimes leads to deleterious psychological outcomes. Given that people nowadays rely on emojis to express emotions online, the present study investigated (1) whether display rules exist in emoji communications and (2) how emotion management using emojis is associated with psychological well-being. Prior studies have demonstrated the effects of context on the frequency of emoji use. However, the intensity and type of expression may differ, even if emojis are used at the same frequency. The current study thus investigated whether emotional expressions and the types of emojis used are adjusted to contexts similar to facial displays. As many as 1,289 Japanese participants typed emojis in response to Internet chats and reported the intensity of their emotional expressions. The contexts of the chats varied depending on the target of use, the emotional value of contexts, and private or public settings. The results showed that, similar to facial displays, individuals expressed emotions through emojis more with those closely related, more in positive contexts than in negative contexts, and more in private than in public contexts. When the expressions were intense, individuals used emojis consistent with the emotional value of the context. Upon attenuating the expressions, this study found that individuals tended to use euphemistic emojis and sent smiling emojis in negative contexts to manage the expressions. Moreover, expressing emotions with emojis was associated with subjective well-being, whereas managing emotions with emojis was weakly associated with depressive symptoms. Together, this study indicates the existence of display rules for emojis, calling for future research on the psychological impact of online emotion norms.

    Abstract The purpose of the article is to determine the role of critical thinking and unconscious competence in the implementation of effective communication during group discussions, debates and dialogues. The necessary conditions for creating an effective educational environment conducive to the development of students' conversational skills are demonstrated. An educational experiment was conducted with the participation of 75 third-year students from the School of [BLINDED], [BLINDED], during which the students practiced both critical thinking and unconscious competence in the process of group discussions. The conducted survey at the beginning and end of training, was determined the degree of use of critical thinking skills and unconscious competence in the process of finding answers to arguments during debates, discussions and disputes. The results of the surveys showed that in the process of speaking, critical thinking skills are used more often (79%) than unconscious competence (21%), but at the same time, students considered that unconscious competence (81%) is more effective in debates and discussions, than critical thinking (19%). It was concluded that critical thinking skills are easier and faster to learn to participate in a constructive discussion than the skills of unconscious competence, the development of which must take place in an authentic learning environment for a longer period. The results of the study confirmed that the participation of students in the conversation class increased their ability to analyze, critically evaluate, argue, unconsciously respond and understand the interlocutor. Therefore, it is important to invest additional efforts and create conditions for open, flexible and comfortable communication of students using modern pedagogical methods aimed at developing students' thinking skills of a higher order. The findings can be useful in the field of language teaching, psychology, and linguistics, as well as become the basis for the development of new curricula using collective discussions.

    Abstract How the transformation of trade patterns affects carbon emissions is an important topic in the field of trade and the environment. Based on the panel data of Chinese cities from 2000 to 2016, this paper uses fixed effect model, instrumental variable model and threshold estimation model to investigate the impact of the transformation of export trade patterns on carbon emissions and its internal mechanism. We find that during the investigation period, the transformation of export trade pattern has a significant "U"-shaped nonlinear impact on carbon emissions, and the research results still hold after a series of robustness checks and instrumental variable estimation; through the mechanism analysis of the threshold effect, it is found that in the process of the impact of trade patterns transformation on carbon emissions, there are double thresholds for energy structure transition; the nonlinear impact of export trade pattern transformation on carbon emissions has significant heterogeneity in different regions, city categories, and different periods; the mechanism analysis results show that industrial structure upgrading, industrial co-agglomeration, and green technology innovation are the main paths that the transformation of export trade patterns affects carbon emissions. Under the dual goals of fulfilling China's commitment to "carbon peak in 2030 and carbon neutrality in 2060" and implementing corporate social governance to promote green and sustainable development, this paper will provide suggestions for realizing the goal of low-carbon emission reduction from the perspective of the transformation of export trade patterns.
