    Abstract Background: Hepatocellular carcinoma (HCC) is a common clinical malignant tumor of the digestive system. Hu-Qi-Zheng-Xiao (HQZX) decoction has been clinically found to prolong the survival of patients with hepatocellular carcinoma and improve the quality of patients' survival, but its antitumor biological mechanism is still unclear.
    Methods: A nude mouse hollow fiber hepatocellular carcinoma model was constructed to analyze the in vivo efficacy of HQZX decoction against 7 different hepatocellular carcinoma cells. The subcutaneous graft tumor model was again validated. In vitro, the effect of HQZX decoction on the growth and metastasis of the cell line with the highest growth inhibition was evaluated. The cell line with the best efficacy response screened was again used to construct a hollow fiber hepatocellular carcinoma model and hollow fiber conduit cells were extracted to detect the expression of HIF-1α, VEGF, EMT-related molecules, LCSCs-related molecules, and to observe the density of the subcutaneous vascular network of hollow fiber conduits. The liver metastasis model of splenic injection was constructed to observe the effect of HQZX decoction on tumor metastasis.
    Results: The hollow fiber hepatocellular carcinoma model was evaluated for the efficacy of HQZX decoction, and it was found to have the highest growth inhibition of LM3-luc cells. In vitro, the CCK8 assay revealed that HQZX decoction could inhibit tumor migration and invasion and promote apoptosis. In addition, the mechanism study of extracting cells from hollow fiber tubes found that HQZX decoction could inhibit metastasis-associated HIF-1α, VEGF, EMT-related molecules, and LCSCs-related molecules expression. capillary network around subcutaneous fiber tubes was reduced in the HQZX decoction gavage group of mice. It inhibited tumor metastasis in nude mice.
    Conclusions: HQZX decoction inhibited the growth of a variety of hepatocellular carcinoma cells. HQZX decoction suppressed the expression of metastasis-associated VEGF, EMT-related molecules, and LCSCs-related molecules and inhibited tumor angiogenesis and growth and metastasis, which may be related to the inhibition of the HIF-1α signaling pathway. It reveals that HQZX decoction may be a promising herbal compound for anti-HCC therapy, and also reveals the accurate feasibility of the hollow fiber hepatocellular carcinoma model for in vivo pharmacodynamic evaluation and mechanism study.

    Abstract This research was designed to explore the variation characteristics of soil C:N:P stoichiometry and enzyme activity in the Qilian Mountains different grassland types. Thus, 7 grassland types (Upland meadow: UM, Alpine meadow: AM, Temperate steppe: ST, Alpine steppe: AS, Temperate Desert Steppe: TDS, Temperate Desert: TD, Alpine desert: AD) of Qilian Natural Reserve were selected to analyze the variation characteristics of soil enzyme activities and stoichiometry of different grassland types and its relationship with environmental factors. The study indicated that the C/N, C/P, and N/P of different grasslands ranged from 5.08 to 17.35, 2.50 to 72.29, and 0.53 to 4.02.The ranking of different types grassland for the C/N was TS ≥ AM ≥ UM ≥ AS ≥ TDS > AD > TD, and the changing pattern of C/P and N/P is similar to that of C/N. The ranking of different types grassland for the urease enzyme activity was UM ≈AS > AD ≈TDS ≈TS ≈AM > TD, and TS ≈AM ≈UM ≈AS ≈AD > TDS > TD for alkaline phosphatase enzyme activity, and AS ≈AM ≈TS ≈TDS≥UM ≥TD ≈AD for catalase enzyme activity. Based on N/P ratio and RDA analysis, nitrogen was the main factor limiting the grassland productivity, and pH, TN, SOC, Richness index and Simpson diversity index were the main environmental factors affecting the soil C:N:P stoichiometry and enzyme activities. Cluster analysis showed that 7 grassland types were clustered into three categories. In conclusion, the stoichiometric characteristics and soil enzyme activities of different grasslands vary with grassland types. Nitrogen was the main factor limiting the grasslands productivity, and pH, TN, SOC, Richness index and Simpson diversity index were the main environmental factors affecting the soil C:N:P stoichiometry and enzyme activities, and the grassland Qilian Mountain can be managed in the ecological district according to the clustering results. The results of this study can provide data support and theoretical guidance for the scientific management and ecological protection of grassland in Qilian Mountains Reserve.

    Abstract Background: The phenotype of peripheral neuropathy (PN) associated with glial fibrillary acidic protein-immunoglobulin G (GFAP-IgG) has not been well described.
    Objectives: The aim of this study was to report the frequency, clinical, and electrophysiological characteristics of PN in GFAP-IgG-positive patients.
    Design: This study is a single-center retrospective observational study.
    Data sources and methods: GFAP-IgG-positive patients with PN were retrospectively identified from the Huashan Hospital Autoimmune Encephalitis Cohort between 2017 and 2021. Eight patients who presented with PN from other published studies were also included in the analysis. The clinical and electrophysiological characteristics of GFAP-IgG-related PN were described.
    Results: A total of 21 (31%) patients (7 females, 14 males;
    Conclusion: Peripheral nerves could be involved in autoimmune GFAP astrocytopathy. Predominant motor axonal neuropathy mainly involving the lower extremities is the most common PN phenotype in this disorder. GFAP-IgG-related PN is responsive to immunotherapy.

    Abstract Hepatocellular carcinoma (HCC) is the predominant pathological type of liver cancer. Several therapeutic treatments, including sorafenib and regorafenib, have only modestly improved survival in patients with HCC. The aim of this study was to investigate the expression profiles and the regulation of competitive endogenous RNAs (ceRNAs) of the sorafenib-related target genes in HCC. Based on clinical information and expression profiles of HCC clinical samples from The Cancer Genome Atlas (TCGA) and Gene Expression Omnibus (GEO) databases, shared differentially expressed genes (DEGs) were analyzed and identified. Sorafenib-associated DEGs (SADs) were obtained by intersecting the DEGs with the sorafenib target genes from SuperTarget database. The expression patterns of SADs were verified in the Oncomine database. The biological functions of the SADs were annotated by gene set enrichment analysis (GSEA). In addition, a ceRNA network associated with SADs was constructed. Long non-coding RNAs (lncRNAs) in network that were significantly associated with overall survival were identified as prognosis of patients by Cox regression analysis. Finally, the expression levels of prognostic genes in HCC tissues and cell lines were verified using qRT-PCR. Gene expression differential analysis yielded a total of 146 common DEGs were obtained, including 21 upregulated and 125 downregulated DEGs. Among them, ten SADs were detected to be differentially expressed between tumor and normal tissues, including AXL, CYP2C19, CYP2C8, CYP2C9, CYP3A4, FGFR2, GMNN, PDGFRA, and TTK. GSEA analysis grouped them into three categories by function. The first category (CYP2C19, CYP2C8, CYP2C9 and CYP3A4) and second category (GMNN, TTK and EGER2) had the opposite roles in the enriched terms and pathways, while the third class (AXL and PDGFRA) has enrichment terms and pathways that intersect with those of the first and second categories. A ceRNA network associated with SADs was also constructed including 49 lncRNAs, 14 miRNAs, and 8 mRNAs. Three of these lncRNAs, SNHG7, GAS5 and HCP5, were found upregulated in HCC tissues and to be independent predictors in HCC patients. Significant correlations were found in expression between the prognostic lncRNAs and SADs. Ten SADs were systematically identified using expression data from HCC and normal tissues from TCGA and GEO datasets. GSEA analysis provided us with insight into the function of SADs. In the future, we will continue to explore the mechanisms of coordinated regulation of SADs-related prognostic lncRNAs and SADs at the ceRNA axis level and their potential functions in the development of HCC.

    Abstract Potential natural vegetation (PNV), the final successional stage of vegetation, plays a key role in ecological restoration, the design of nature reserves, and development of agriculture and livestock production. Meteorological data from historical and current periods including the last inter-glacial (LIG), last glacial maximum (LGM), mid Holocene (MH) periods and the present day (PD), plus derived data from 2050 and 2070, in conjunction with the Comprehensive and Sequential Classification System (CSCS) model, were used to classify global PNV. The 42 classes of global PNV were regrouped into 10 groups to facilitate analysis of spatial changes. Finally, spatio-temporal patterns and successional processes of global PNV as well as the response to climate changes were analysed. Our study made the following five conclusions. (1) Only one missing class (IA1 frigid-extrarid frigid desert, alpine desert) arose in periods of LIG, MH, 2050, and 2070 for global PNV. (2) The frigid-arid groups were mainly distributed in higher latitudes and elevations, but temperate-humid groups and tropical-perhumid groups occurred in middle and low latitudes, respectively. Temperate zonal forest steppe, warm desert, savanna and tropical zonal forest steppe increased, while six other groups decreased. (3) The conversion from temperate zonal forest steppe to tundra and alpine steppe from LIG to LGM occupied the largest area, indicating a drastic shift in climate and the associated response of terrestrial vegetation sensitive to climate change. (4) The CSCS could be used to simulate the long-term succession of global PNV. (5) As a consequence of global warming, forests shifted to the northern hemisphere and Tibet, areas with much higher latitude and elevation. The PNV groups with greater shift distance revealed the more serious effects of global climate change on vegetation.
