    Abstract In vitro culture and generation of highly specialized goblet cells is still a major challenge in conjunctival 3D in vitro equivalents. A model comprising all physiological factors, including mucus-secreting goblet cells has the potential to act as a new platform for studies on conjunctival diseases. We isolated primary conjunctival epithelial cells and fibroblasts from human biopsies. 3D models were generated from either epithelial layers or a combination of those with a connective tissue equivalent. Epithelial models were investigated for marker expression and barrier function. Full-thickness models were analyzed for goblet cell morphology and marker expression via immunofluorescence and quantitative real-time PCR. Simple epithelial models cultured at the air-liquid interface showed stratified multi-layer epithelia with pathologic keratinization and without goblet cell formation. The combination with a connective tissue equivalent to generate a full-thickness model led to the formation of a non-keratinized stratified multi-layer epithelium and induced goblet cell differentiation. In our model, a high resemblance to natural conjunctiva was achieved by the combination of conjunctival epithelial cells with fibroblasts embedded in a collagen-hydrogel as connective tissue equivalent. In the future, our conjunctival in vitro equivalent enables the investigation of goblet cell differentiation, conjunctival pathologies as well as drug testing.

    Abstract Products containing chemicals with eye irritation potential need to be labeled with the respective hazard symbol. To avoid the testing of numerous dilutions of chemicals on animals, their labeling is directed by a theoretical approach. In this report, a previously described in vitro tissue model of the cornea based on human epithelial cells was used for eye irritation testing of dilutions. As a sensitive and non-destructive method to analyze the barrier function of the epithelium, impedance spectroscopy was applied. Moreover, the morphology and viability of the epithelial models were assessed. We tested four chemicals that, neatly, cause severe damage to the eye: tetrahydrofuran, acetic acid, diethylethanolamine, and benzalkonium chloride. With our test method, we were able to determine the concentrations of the chemicals which are critical for the integrity of the cornea. The threshold was < 0.1% for the most and > 5% for the least toxic substance. The described test system is not only an alternative for animal models but also for the theoretical examination of the hazard potential of diluted chemicals. By using the advantages of tissue engineering and non-destructive analysis tools, we can achieve more precise and safer labeling of the eye irritation potential of products.

    Abstract Long-term storage and cryopreservation of biological tissues is a global challenge in the fields of regenerative medicine, tissue engineering and implants, because of the formation of large ice crystals that may damage cellular membranes and extracellular matrix (ECM) of collagen. In our studies different thawing mechanisms were tested to achieve more uniform warming of 3D in vitro tissue models, prepared from magnetic nanoparticles (NPs)-modified 3D electrospun nanofiber fleeces and fibroblasts. 3D tissue models frozen in liquid nitrogen were defrosted either with water bath or with radio-frequency (RF) inductive heating of the magnetic NPs and their morphology was compared to that of a non-frozen tissue model using a transmission Mueller microscope and thin sections of all three types of tissue models. Our results demonstrate a sensitivity of the polarimetric parameters obtained with the differential decomposition of Mueller matrices to small changes in sample morphology caused by the different thawing methods. A detailed statistical analysis proved the statistical significance of the experimental data from the three groups of all tissue models.
