Abstract Hypoxia-inducible factor (HIF) is a nuclear protein that plays a crucial role in oxygen homeostasis through its transcriptional activity and thousands of target gene profiles. Through transcriptional and post-transcriptional regulation, the downstream target genes of HIF can trigger multiple pathological responses in the body, including energy metabolism, cytopenia, and angiogenesis. There are three distinct subtypes of HIF: HIF-1, HIF-2, and HIF-3. HIF-1 is a significant regulator of the cellular response to hypoxia, and the balance between its production and degradation is critical for this response. As hypoxia is linked to several disorders, understanding HIF can open up novel avenues for the treatment of many diseases. This review describes the regulatory mechanisms of HIF-1 synthesis and degradation and the clinical significance of the hypoxia-inducible factor pathway in lung injury, kidney disease, hematologic disorders, and inflammation-related diseases.

Abstract A radical initiator-free defunctionalization reaction of alkyl isocyanides with a hydrosilane has been established through C-N bond cleavage under catalyst-free visible light irradiation. Various alkyl isocyanides participated in the defunctionalization with tris(trimethylsilyl)silane under blue light irradiation at room temperature, delivering the reduced products in good yields with high chemoselectivity.

Abstract Biomolecular condensates formed by multicomponent phase separation play crucial roles in diverse cellular processes. Accurate assessment of individual-molecule contributions to condensate formation and precise characterization of their spatial organization within condensates are crucial for understanding the underlying mechanism of phase separation. Using molecular dynamics simulations and graph theoretical analysis, we demonstrated quantitatively the significant roles of cation-π and π-π interactions mediated by aromatic residues and arginine in the formation of condensates in polypeptide systems. Our findings reveal temperature and chain length-dependent alterations in condensate network parameters, such as the number of condensate network layers, and changes in aggregation and connectivity. Notably, we observe a transition between assortativity and disassortativity in the condensate network. Moreover, polypeptides W, Y, F, and R consistently promote condensate formation, while the contributions of other charged and two polar polypeptides (Q and N) to condensate formation depend on temperature and chain length. Furthermore, polyadenosine and polyguanosine can establish stable connections with aromatic and R polypeptides, resulting in the reduced involvement of K, E, D, Q, and N in phase separation. Overall, this study provides a distinctive, precise, and quantitative approach to characterize the multicomponent phase separation.

Abstract As the primary domain of ideological and political education in higher education institutions, ideological and political courses must align with principles rooted in human psychology and education. Integrating educational psychology into ideological and political teaching in universities enhances the scientific, targeted, and forward-thinking nature of such education. The burgeoning exploration of knowledge graph applications has extended to machine translation, semantic search, and intelligent question answering. Diverging from traditional text matching, the knowledge spectrum graph transforms information acquisition in search engines. This paper pioneers a predictive system for delineating the relationship between educational psychology and ideological and political education in universities. Initially, it extracts diverse psychological mapping relationships of students, constructing a knowledge graph. By employing the KNN algorithm, the system analyzes psychological characteristics to effectively forecast the relationship between educational psychology and ideological and political education in universities. The system's functionality is meticulously detailed in this paper, and its performance is rigorously tested. The results demonstrate high accuracy, recall rates, and F1 values. The F1 score can reach 0.95enabling precise sample classification. The apex of the average curve for system response time peaks at approximately 2.5 s, maintaining an average response time of less than 3 s. This aligns seamlessly with the demands of practical online teaching requirements. The system adeptly forecasts the relationship between educational psychology and ideological and political education in universities, meeting response time requirements and thereby fostering the scientific and predictive nature of ideological and political teaching in higher education institutions.

Abstract Heat engines used to output useful work have important practical significance, which, in general, operate between heat baths of infinite size and constant temperature. In this paper, we study the efficiency of a heat engine operating between two finite-size heat sources with initial temperature difference. The total output work of such heat engine is limited due to the finite heat capacity of the sources. We firstly investigate the effects of different heat capacity characteristics of the sources on the heat engine's efficiency at maximum work (EMW) in the quasi-static limit. Moreover, it is found that the efficiency of the engine operating in finite-time with maximum power of each cycle is achieved follows a simple universality as η=ηC/4+OηC2, where ηC is the Carnot efficiency determined by the initial temperature of the sources. Remarkably, when the heat capacity of the heat source is negative, such as the black holes, we show that the heat engine efficiency during the operation can surpass the Carnot efficiency determined by the initial temperature of the heat sources. It is further argued that the heat engine between two black holes with vanishing initial temperature difference can be driven by the energy fluctuation. The corresponding EMW is proved to be ηMW=2-2.

Abstract AbstractConsumers’ different aesthetic experience affects their choice of products, so how to construct product design features space based on consumers’ aesthetic experience, especially how to integrate traditional cultural elements into modern product design, has become one of the most effective ways to improve product design quality.Taking the innovative design of traditional Chinese Miao’s patterns on canvas bags as an example, this paper constructs the mapping relationship between consumers’ aesthetic experience and product feature space on the basis of analyzing the feature elements of Chinese Miao traditional patterns “Jiuqu River flower”, as pronounced in Chinese.Using word cloud semantic tool, this paper analyzes the interview results of three consumer groups, namely, fashion youth, intellectuals and housewives, and obtains the aesthetic expression of three consumer groups on the innovative design of traditional patterns on canvas bags. Based on this, the feature elements of traditional pattern extraction and redesign of “Jiuqujiang River Flower” are used. Three canvas bag styles containing traditional cultural elements were constructed for different perceptual word pairs. Finally, the 7-level semantic difference quantization method of Kansei Engineering was used to quantitatively evaluate the effectiveness of the spatial mapping method of aesthetic experience and product features. The final results showed that the modern product quality of creative design can be effectively improved by integrating with the traditional Chinese cultural elements, on the basis of making the mapping from aesthetic experience to product feature space, so that to meet the consumers’ aesthetic needs to the most.

Abstract Background: Sex determining region Y-box 2 (SOX2) can promote squamous cell carcinoma (SSC) because it regulates the migration and invasion of several different types of squamous carcinoma cells. However, few studies have examined the prognostic value of SOX2 and its effect on the epithelial-mesenchymal transition (EMT) in esophageal SSC (ESCC), a cancer characterized by high invasion and rapid metastasis.
Aim: To verify the relationship of SOX2 and the EMT in ESCC and determine the prognostic value and significance of SOX2 and protein markers of the EMT in ESCC.
Methods: One hundred and eighty-five postsurgical ESCC patients were retrospectively examined. Immunohistochemistry was used to detect SOX2, E-cadherin, and vimentin in ESCC tissues. The chi-square test was used to determine the relationships of the expression of these proteins with clinical data. Kaplan-Meier survival curves were used to evaluate factors associated with overall survival (OS).
Results: SOX2 and vimentin had high expression in ESCC tissues and correlated with the depth of local carcinoma invasion. SOX2 expression had positive correlations with tumor size, vimentin expression, and the EMT, and a negative correlation with E-cadherin expression. Expression of SOX2 and vimentin had negative correlations with OS. SOX2 expression was an independent prognostic risk factor for poor OS in patients with ESCC.
Conclusion: SOX2 expression was an independent risk factor for OS in patients with ESCC and its expression had a positive correlation with the expression of vimentin, a classic marker of the EMT. SOX2 promoted the migration and invasion of ESCC, and this may related to its effect on vimentin in promoting the EMT.
