    Abstract Bacterial cells that stop growing but maintain viability and the capability to regrow are termed dormant and have been shown to transiently tolerate high concentrations of antimicrobials. Links between tolerance and cellular energetics as a possible explanation for the tolerance, have been investigated and have produced mixed and seemingly contradictory results. Because dormancy merely indicates growth arrest, which can be induced by various stimuli, we hypothesize that dormant cells may exist in a range of energetic states that depend on the environment. To energetically characterize different dormancies, we first induce them in a way that results in dormant populations and subsequently measure both of their main energy sources, the proton motive force magnitude and the concentration of ATP. We find that different types of dormancy exhibit characteristic energetic profiles that vary in level and dynamics. The energetic makeup was associated with survival to some antibiotics but not others. Our findings portray dormancy as a state that is rich in phenotypes with various stress survival capabilities. Because environmental conditions outside of the lab often halt or limit microbial growth, a typologization of dormant states may yield relevant insights on the survival and evolutionary strategies of these organisms.

    Abstract One of the recurring issues in forensic document analysis is determining the chronological order of line-crossing sequences. This seemingly simple but tricky task allows forensic laboratories to demonstrate the real order of events. The aim of this work is to analyse black ink/toner intersections using the digital microscope Hirox RH-2000. Five types of pens (ballpoint, gel, rollerball, fountain and fineliner) as well as five different laser printers were used to create the samples. Forensic examiners were tasked with assessing toner fractures, the bronzing effect, and gaps. The findings were analysed to determine the importance of each characteristic for each type of pen. Results indicate that observing the bronzing effect at high magnification (700x) is crucial for establishing the sequence order of application. In this regard, the fineliner pen is considered the most challenging type of pen to analyse in a black ink/toner intersection. The method was validated through a blind test. Based on this, a flow chart has been developed to assist the Forensic Document Examiners (FDEs) in their evaluations. The flow chart emphasises the significance of combining multiple observations and enables a more objective analysis for trained FDEs.

    Abstract Running inevitably causes the animal trunk to undulate. The consequential head rotations have to be stabilised in space for a steady gaze and an accurate sense of self-motion for balance. The ecology and anatomy of the species determine the necessity to stabilise the head in yaw, pitch, and roll direction. Terrestrial lizards, running with a sprawled body posture, are especially prone to undulations in the horizontal (yaw) plane. Measurements on an experimental oscillation platform show that Acanthodactylus pardalis lizards stabilise their head less in pitch direction (54% stabilisation) than in yaw and roll direction (66% and 64% stabilisation, respectively). Because we performed these experiments in darkness, the lizards based their head stabilisation on vestibular information. Hence, we hypothesised that their vestibular system is less sensitive in pitch direction than in yaw and roll direction. Yet, this was not confirmed by a detailed Fluid-Structure Interaction model of the membranous labyrinth, which showed that not pitch sensitivity (88% of yaw sensitivity), but roll sensitivity (73% of yaw sensitivity) is the lowest. So why is the head stabilisation in darkness almost as good in roll direction as in yaw direction? While this may be due to neurological nonlinearities, it seems worth noticing that the moment of inertia is lowest in roll direction due to the elongated head shape. Hence, less torque is needed to stabilise a head rotation in roll direction than in the other two directions.

    Abstract We describe a phenotypic antibiotic susceptibility testing (AST) method that can provide an eightfold speed-up in turnaround time compared with the current clinical standard by leveraging advances in microscopy and single-cell imaging. A newly developed growth plate containing 96 agarose pads, termed the multipad agarose plate (MAP), can be assembled at low cost. Pads can be prepared with dilution series of antibiotics. Bacteria are seeded on the pads and automatically imaged using brightfield microscopy, with a fully automated segmentation pipeline quantifying microcolony formation and growth rate. Using a test set of nine antibiotics with very different targets, we demonstrate that accurate minimum inhibitory concentration (MIC) measurements can be performed based on the growth rate of microcolonies within 3 h of incubation with the antibiotic when started from exponential phase. Faster, reliable and high-throughput methods for AST, such as MAP, could improve patient care by expediting treatment initiation and alleviating the burden of antimicrobial resistance.

    Abstract Background: The Health Impact Assessment (HIA) is a procedure with the aim to protect the populations exposed to the impacts deriving from the establishment or upgrading of large industrial enterprises, i.e. large combustion plants (>300 MWth). In Italy a guideline for the HIA procedure has been published in compliance with the 2014/52/EU Directive on the Environmental Impact Assessment (EIA) requirements. Italian HIA procedure. An ecotoxicological approach has been included for the first time in the HIA procedure with the aim to detect toxic effects caused by unknown not-monitored contaminants or mixtures in the ecosystem components affected by the potential emissions, discharges and releases of large industrial enterprises. Ecotoxicology plays an important bridge role between environment and human health in the scoping and monitoring step of the HIA procedure with a key function of early warning system and screening. The aim of this paper is to present the Italian experience in the first three years of the application of the new approach, proposing recommendations on specific case studies. Conclusion and future perspective. 80% of enterprises, that applied HIA, have delivered a robust, integrated and detailed documentation in relation to the ecotoxicological assessment, this positive feedback will generate environmental and human health benefits to the areas where the plants are established.
