    Abstract An activatable probe able to detect RONS level underlying the development of rheumatoid arthritis (RA) would be far-reaching for the diagnosis and drug efficacy assessment of RA. Despite more and more evidence suggests that ONOO⁻ is an important signaling molecule participating in the RA disease, only rare fluorescent probes can detect ONOO⁻ in this disease with satisfying performance. To this end, we designed and synthesized a novel activatable AIE fluorescent probe (DPPO-PN) for the detection of ONOO⁻ levels in RA. The probe linearly responds to 0–10 μM ONOO⁻ with a significant far-red fluorescence enhancement at 632 nm in 30 s (F/F₀ = 161-fold, LOD = 10 nM) upon the excitation of 490 nm, elucidating excellent sensing abilities for ONOO⁻ in cuvettes. Moreover, the fluctuations of intracellular ONOO⁻ levels caused by adscititious adding or stimulant provoking could be real-time captured and visualized with this probe by confocal imaging. Further, the intravital imaging of ONOO⁻ in LPS-induced and CFA-induced RA mouse models also can be achieved with the aid of the probe, substantiating the burst of ONOO⁻ in the RA process. Therefore, this work not only offers an auxiliary tool for the diagnosis of RA but also would benefit our understanding of the ONOO⁻‘s roles in RA.

    Abstract The inherent complexity of the fluid flow in subsurface systems brings potential inevitable uncertainty in their characterization. Computationally intensive high-dimensional inversion problems often emerge in solving the fluid flow problems of various scenarios, which required to be probed. To improve the efficiency of solving such problems, surrogate strategies are widely used to quantify the uncertainty of underground multiphase flow models. In this paper, a deep learning surrogate model is developed for predicting the time-dependent dynamic multiphase flow in a two-dimensional (2D) channelized geological system. The surrogate model is combined with a residual U-net and an autoregressive strategy, which considers the output at the previous time step as input and predict the output at the current time step. The residual U-net has a symmetric network structure similar to U-net and contains extra residual units. The rich skip connections in the network can promote information dissemination and achieve better prediction performance with fewer parameters. We demonstrated the performance of the autoregressive residual U-net (AR-Runet) for predicting the migration of solute transport in heterogeneous 2D binary model. The result shows the AR-Runet surrogate model can provide an accurate approximation of saturation and pressure fields at different times. We also have demonstrated that with the autoregressive strategy this network can achieve similar predict results with relatively less training data. The performance of the AR-Runet network is also compared with the autoregressive Dense net (AR-Dense). The findings indicate that the AR-Runet can provide effective measures for developing surrogate model and uncertainty analysis in dynamic multiphase flow predictions of subsurface systems.

    Abstract The layer-dependent Chern number (C) in MnBi_{2}Te_{4} is characterized by the presence of a Weyl semimetal state in the ferromagnetic coupling. However, the influence of a key factor, namely, the exchange coupling, remains unexplored. This study focuses on characterizing the C=2 state in MnBi_{2}Te_{4}, which is classified as a higher C state resulting from the anomalous n=0 Landau levels (LLs). Our findings demonstrate that the exchange coupling parameter strongly influences the formation of this Chern state, leading to a competition between the C=1 and 2 states. Moreover, the emergence of odd-even LL sequences, resulting from the breaking of LL degeneracy, provides compelling evidence for the strong exchange coupling strength. These findings highlight the significance of the exchange coupling in understanding the behavior of Chern states and LLs in magnetic quantum systems.

    Abstract Dual-channel fluorescent probes could respond to a specific target and emit different wavelengths of fluorescence before and after the response. Such probes could alleviate the influence caused by the variation of the probe concentration, excitation intensity, and so on. However, for most dual-channel fluorescent probes, the probe and fluorophore faced spectral overlap, which reduced sensitivity and accuracy. Herein, we introduced a cysteine (Cys)-responsive and near-infrared (NIR) emissive AIEgen (named TSQC) with good biocompatibility to dual-channel monitor Cys in mitochondria and lipid droplets (LDs) during cell apoptosis through wash-free fluorescence bio-imaging. TSQC can label mitochondria with bright fluorescence around 750 nm, and after reacting with Cys, the reaction product TSQ could spontaneously target LDs with emissions around 650 nm. Such spatially separated dual-channel fluorescence responses could significantly improve detection sensitivity and accuracy. Furthermore, the Cys-triggered dual-channel fluorescence imaging in LDs and mitochondria during apoptosis induced by UV light exposure, H₂O₂, or LPS treatment is clearly observed for the first time. Besides, we also report here that TSQC can be used to image subcellular Cys in different cell lines by measuring the fluorescence intensities of different emission channels. In particular, TSQC shows superior utility for the in vivo imaging of apoptosis in acute and chronic epilepsy mice. In brief, the newly designed NIR AIEgen TSQC can respond to Cys and separate two fluorescence signals to mitochondria and LDs, respectively, to study Cys-related apoptosis.
