    Abstract In modern scientific research, the objective is often to identify which variables are associated with an outcome among a large class of potential predictors. This goal can be achieved by selecting variables in a manner that controls the the false discovery rate (FDR), the proportion of irrelevant predictors among the selections. Knockoff filtering is a cutting-edge approach to variable selection that provides FDR control. Existing knockoff statistics frequently employ linear models to assess relationships between features and the response, but the linearity assumption is often violated in real world applications. This may result in poor power to detect truly prognostic variables. We introduce a knockoff statistic based on the conditional prediction function (CPF), which can pair with state-of-art machine learning predictive models, such as deep neural networks. The CPF statistics can capture the nonlinear relationships between predictors and outcomes while also accounting for correlation between features. We illustrate the capability of the CPF statistics to provide superior power over common knockoff statistics with continuous, categorical, and survival outcomes using repeated simulations. Knockoff filtering with the CPF statistics is demonstrated using (1) a residential building dataset to select predictors for the actual sales prices and (2) the TCGA dataset to select genes that are correlated with disease staging in lung cancer patients.

    Abstract Interpretable regression models are important for many application domains, as they allow experts to understand relations between variables from sparse data. Symbolic regression addresses this issue by searching the space of all possible free form equations that can be constructed from elementary algebraic functions. While explicit mathematical functions can be rediscovered this way, the determination of unknown numerical constants during search has been an often neglected issue. We propose a new multi-objective memetic algorithm that exploits a differentiable Cartesian Genetic Programming encoding to learn constants during evolutionary loops. We show that this approach is competitive or outperforms machine learned black box regression models or hand-engineered fits for two applications from space: the Mars express thermal power estimation and the determination of the age of stars by gyrochronology.

    Abstract Background: Pronation deformity in patients with cerebral palsy can have a major impact on upper limb functionality. There is lack of consensus in the literature about the preferred surgical technique to address this deformity.
    Study aim: To evaluate and synthesize the outcome of different surgical techniques for pronation deformity in patients with cerebral palsy.
    Methodology: The databases MEDLINE and Embase were searched for publications up to December 2021. Articles were considered eligible for inclusion when the included patients had a pronation deformity caused by cerebral palsy and results of surgical intervention for pronation deformity were examined. Evaluation of the quality of the retrieved study was conducted using the MINORS tool. Meta-analysis was not possible due to the heterogeneity of interventions and reported outcomes.
    Results: Nineteen studies, involving 475 patients and eight different techniques were included. All studies reported gain of active supination in most patients. The effect of surgery on functional gain was less clear and there was a large heterogeneity of reported functional outcome measures. There were 46 reported complications. Overall quality of study design was poor, illustrated by the average MINOR score of 6.9/16. Overall, there is a high risk of bias due to poor internal and external validity of the studies.
    Conclusion: Despite positive reports on gain in supination and functionality after most procedures addressing pronation deformity in CP patients, no conclusions can be drawn concerning the preferred technique due to the low quality of the evidence.

    Abstract Introduction: The AOP-Wiki is the main platform for the development and storage of adverse outcome pathways (AOPs). These AOPs describe mechanistic information about toxicodynamic processes and can be used to develop effective risk assessment strategies. However, it is challenging to automatically and systematically parse, filter, and use its contents. We explored solutions to better structure the AOP-Wiki content, and to link it with chemical and biological resources. Together, this allows more detailed exploration, which can be automated.
    Materials and methods: We converted the complete AOP-Wiki content into resource description framework (RDF) triples. We used >20 ontologies for the semantic annotation of property-object relations, including the Chemical Information Ontology, Dublin Core, and the AOP Ontology.
    Results: The resulting RDF contains >122,000 triples describing 158 unique properties of >15,000 unique subjects. Furthermore, >3500 link-outs were added to 12 chemical databases, and >7500 link-outs to 4 gene and protein databases. The AOP-Wiki RDF has been made available at https://aopwiki.rdf.bigcat-bioinformatics.org.
    Discussion: SPARQL queries can be used to answer biological and toxicological questions, such as listing measurement methods for all Key Events leading to an Adverse Outcome of interest. The full power that the use of this new resource provides becomes apparent when combining the content with external databases using federated queries.
    Conclusion: Overall, the AOP-Wiki RDF allows new ways to explore the rapidly growing AOP knowledge and makes the integration of this database in automated workflows possible, making the AOP-Wiki more FAIR.
