    Abstract Municipal and winery wastewater can provide a valuable irrigation source in regions where water accessibility is problematic or sustainable disposal of waste is essential. It is imperative, however, that when used for irrigation, water conservation benefits are not compromised by a decline in soil and plant health. To date, published literature investigating the use of wastewaters for grapevine production is restricted to a limited set of studies. Globally, wastewater usage within the horticultural sector, including winegrape production, is increasing. It is necessary, therefore, to better understand the short‐ and long‐term implications of such practice. Although wastewaters may contain a range of organic and inorganic pollutants, this review focuses primarily on specific issues associated with high salt loading that is typical under wastewater irrigation. Managing salt, in particular sodium (Na+) and potassium (K+), in wastewater will be necessary in order to maintain the soil physical, chemical and biological health in the long term. Soil structural degradation resulting from a high concentration of exchangeable monovalent cations is of major concern where wastewater is being used for irrigation. This review will address (i) the effect of wastewater irrigation on vine performance and grape quality; (ii) chemical and physical changes in soils irrigated with municipal and winery wastewater; and (iii) management practices that may assist in ameliorating vineyard soils abundant in monovalent cations as a result of wastewater irrigation.

    Abstract 1. Optimal partitioning theory (OPT) suggests that plants should allocate biomass to the organ that acquires the most limiting resource. An implied assumption of this is that there are trade-offs in allocation between leaf, stem and root functions. 2. Recently, an alternative approach, allometric biomass partitioning theory (APT), was developed to predict how plants should divide their metabolic production between leaves, stems and roots, based on the constraints of body size. APT predicts that, for an allometrically ideal plant, leaf mass should scale to the 3/4th power of body size, and stem and root mass should scale isometrically to body size. 3. In this study, we combine OPT with APT by investigating biomass partitioning not accounted for by allometric constraints across broad environmental gradients. 4. Intraspecific variability in biomass allocation shows correlations with environmental factors that would be predicted by OPT. However, interspecifically, these patterns either do not appear or are greatly reduced. 5. Our study suggests that, after size is accounted for, intraspecific residual variation in biomass allocation may be partially explained by environmental factors in a manner consistent with OPT. However, the particular patterns vary between species, obscuring large-scale patterns. 6. In summary, differences due to environmental variability can be incorporated with allocation patterns related to total mass to help understand how plants should allocate biomass in response to changes in both size and environment.

    Abstract The biochemistry of cancer cells diverges significantly from normal cells as a result of a comprehensive reprogramming of metabolic pathways. A major factor influencing cancer metabolism is hypoxia, which is mediated by HIF1α and HIF2α. HIF1α represents one of the principal regulators of metabolism and energetic balance in cancer cells through its regulation of glycolysis, glycogen synthesis, Krebs cycle and the pentose phosphate shunt. However, less is known about the role of HIF1α in modulating lipid metabolism. Lipids serve cancer cells to provide molecules acting as oncogenic signals, energetic reserve, precursors for new membrane synthesis and to balance redox biological reactions. To study the role of HIF1α in these processes, we used HCT116 colorectal cancer cells expressing endogenous HIF1α and cells in which the hif1α gene was deleted to characterize HIF1α-dependent and independent effects on hypoxia regulated lipid metabolites. Untargeted metabolomics integrated with proteomics revealed that hypoxia induced many changes in lipids metabolites. Enzymatic steps in fatty acid synthesis and the Kennedy pathway were modified in a HIF1α-dependent fashion. Palmitate, stearate, PLD3 and PAFC16 were regulated in a HIF-independent manner. Our results demonstrate the impact of hypoxia on lipid metabolites, of which a distinct subset is regulated by HIF1α.
