    Abstract ABSTRACTBackground This review aimed to systematically evaluate the immunogenicity and safety of the candidate Ebola virus vaccine (EVV).Methods We searched five databases for randomized controlled trials (RCTs) evaluating the effects of EVV on healthy adults. The primary outcomes were relative risk (RR) of sero-conversion or sero-response of EVV in healthy adults between the groups that received EVV and the controls.Results Twenty-nine RCTs (n = 23573) were included. There was a significant difference in RR of sero-conversion of EVV (RR 13.18; 95% CI 11.28–15.41; I2 = 33%; P < 0.01) between the two groups. There was a significant difference in RR of adverse events (AEs) of EVV (RR 1.49; 95% CI 1.27–1.74; I2 = 88%; P < 0.01), although no difference in RR of serious AE (SAE) between the two groups. Subgroup analysis showed that there was no significant difference in RR of AEs for DNAEBO, EBOV-GP, MVA, and rVSVN4CT1 vaccines, compared with controls.Conclusions The DNAEBO, EBOV-GP, MVA, and rVSVN4CT1 vaccines are likely to be safe and immunogenic, tending to support the vaccination against Ebola disease. These findings should provide much-needed evidence for public health policy makers to develop preventive measures based on disease prevalence features and socio-economic conditions.

    Abstract Perilla frutescens (PF) leaf is a traditional Chinese medicine and food with beneficial effects on allergic asthma. We sought to elucidate the active compounds, the targets, and underlying mechanisms of PF leaf in the treatment of allergic asthma by using experimental pharmacology and network pharmacology. An OVA-allergic asthma murine model was constructed to evaluate the effect of PF leaf on allergic asthma. And the network pharmacology and western blotting were performed to evaluate its underlying mechanisms in allergic asthma. PF leaf treatment significantly improved the lung function of OVA model mice and mitigated lung injury by significantly reducing of OVA-specific immunoglobulin E in serum, and interleukin 4, interleukin 5 and tumor necrosis factor alpha in the bronchoalveolar lavage fluid. 50 core targets were screened based on 8 compounds (determined by high performance liquid chromatography) through compound-target- disease network. Furthermore, MAPK signaling pathway was identified as the pathway mediated by PF leaf with the most potential against allergic asthma. And the WB results showed that PF leaf could down-regulate the expression of p-ERK, p-JNK and p-p38, which was highly consistent with the predicted targets and pathway network. In conclusion, this study provides the evidence to support the molecular mechanisms of PF leaf on the treatment of allergic asthma using network pharmacology and in vivo experiments.

    Abstract Objective. To investigate the effects of total phenolic acid on meningeal microcirculation in mice. Methods. A total of 84 healthy mice were randomly divided into the blank group, the model group, the positive control Western medicine group, the positive control Chinese medicine group, and the large, medium, and small doses of the total phenolic acid group, with 12 rats in each group. The corresponding drug was given to the group once a day for one week, and the litter was changed at 9 : 00 on the 6th day, and the water fasted. On the 7th day, the groups continued to be administered and weighed. After 1 h, the percentage of decrease in the perfusion of the mice was measured. Results. Compared with the blank group, the percentage of perfusion decreased significantly in the model group, indicating that the model was successful. Compared with the model group, the nimodipine group, the Naoluotong group, and the high-dose succulent total phenolic acid group were perfused. The percentage of decrease was significantly decreased (P<0.01), and the percentage of perfusion decreased in the middle and small doses of the total phenolic acid group (P<0.05), indicating that the percentage of mice perfused decreased by the administration of each group. Conclusion. Total phenolic acid can reduce the percentage of mean perfusion of microcirculation in animals and reduce brain damage.

    Abstract Objective. To observe the effect of total glycosides of Xiebai on chronic unpredictable stimuli in rats. Changes of serotonin (5-HT), dopamine (DA), norepinephrine (NE), serum corticosterone (CORT), adrenocorticotropic hormone (ACTH), organ index, and histopathology in brain tissue, to explore the treatment and characteristics of total white glucosides in depression model, were prepared by different stimuli in 21 days. Methods. Depression models were replicated by 21 days of chronic mild stimulation in rats. Finally, the levels of CORT and ACTH in the serum of rats with depression model and the levels of NE, DA, and 5-HT in brain homogenates were determined. The thymus and spleen wet weight of each group were measured, and the organ index was calculated. The thymus and pituitary were observed under the light microscope. Pathological changes of the hypothalamus, adrenal gland, and spleen and behavioral indexes of rats in each group were determined. Results. The results of the experiment showed that 21 days of chronic unpredictable stimulation significantly improved the behavioral response of the model group, and the levels of corticosterone and adrenocorticotropic hormone in the serum and the indicators in the brain homogenate significantly changed, affecting the relevant tissues. The index changes and the results of the combination can prove that this modeling method can be used for the preparation of depression models. The total white glucoside group can significantly improve the above effects, demonstrating that total glucosides of glucoside have a good therapeutic effect on the rat model of depression. Conclusion. The chronic unpredictability of the total dose of Xiebai glycosides can significantly improve the levels of NE and DA and the levels of serum CORT and ACTH in the brain homogenate of depressive model rats and improve the immune function of the body.

    Abstract Objective: To observe the effect of Wuweizi syrup on brain aging mice model induced by d-galactose in mice. Methods: 72 mice were randomly divided into blank group, model group, naokangling group and low, middle and high dose Wuweizi syrup group. After 3 days of adaptive feeding, except for the blank group, the rest mice were subcutaneously (sc) injected with d-galactose 1.25 g/kg at neck back to replicate the aging model. Sc injection was given for 40 consecutive days, and corresponding drugs were given by gavage on the 11th day of modeling. On the 39th day, the dark experiment was carried out to measure the learning ability of mice. After the last administration for 2 h, brain, liver, thymus and spleen were taken and weighed to calculate the organ index. Blood was taken and CAT, GSH and SOD levels in whole blood were determined. Some brain and liver tissues were taken to prepare homogenates, and MDA levels in tissue homogenates and plasma were measured. Brain, liver, thymus and spleen were stained with HE, and pathological sections were made for observation under light microscope. Results: Compared with the blank group, in the model group, the incubation period of mice was significantly reduced and the number of shuttle back and forth was significantly increased (P < 0.01); Spleen and brain indexes decreased significantly (P < 0.05), thymus indexes were decreased significantly, and liver indexes were increased significantly (P < 0.01); The levels of CAT, GSH and SOD in whole blood were significantly decreased (P < 0.01); MDA levels in liver homogenates, brain homogenates and plasma were significantly increased (P < 0.01); Pathological changes of brain, liver, thymus and spleen were observed under microscope.     Abstract Objective: To observe the intervention effect of total flavonoid of ilex pubesceus on animal models of cerebral ischemic tolerance. Methods: A rat model of global-focal cerebral ischemic tolerance was established by blocking bilateral common carotid artery blood flow and occluding left middle cerebral artery using thread-occlusion method. After the first operation, the Ginaton group and large-dosage, medium-dosage and small-dosage groups of total flavonoid of ilex pubesceus were given intragastric administration of corresponding drugs. The sham-operated group, pretreatment model group and ischemia-reperfusion group were given intragastric administration of the same volume of normal saline, 1 time a day, and administrated for 4d. At 24 h after the second operation, the neurological deficit was assessed, the whole blood viscosity, plasma viscosity, iNOS activity as well as NO level, IL-1β content and TNF-α content in the brain tissue of the rats were determined, and the morphological changes of brain tissue of the rats were observed by HE staining. Results: All the rat models of cerebral ischemic tolerance were established successfully. The total flavonoid of ilex pubesceus can obviously or significantly reduce the neurological deficit score, whole blood viscosity and plasma viscosity, obviously or significantly increase the NO level in the brain tissue of the rats, and significantly reduce the pathological damage of brain tissue of the rats. But compared with the ischemia-reperfusion group, the total flavonoid of ilex pubesceus can significantly or obviously increase the iNOS activity, IL-1β content and TNF-α content in the brain tissue of the rats.

    Abstract Objective: To observe the effect of common clinical drug Shuangjin Lian mixture on rats with oral ulcer and discuss its mechanism. Methods: Sodium carboxymethyl cellulose caused leukocyte aggregation in rats, observed the anti-inflammatory effect of Shuangjin mixture. 3 mm * 3 mm size ulcer surface in the oral cavity of rats was caused by 90% phenol solution, to observe the therapeutic effect and anti-inflammatory effect of Shuangjinlian mixture on rats with oral ulcers. Result: The low, middle and high dose Shuangjinlian mixture can inhibits the accumulation of white blood cells caused by sodium carboxymethyl cellulose in rats significantly (P < 0.01). And reduce the degree of edema and hyperaemia around the ulcer tissue significantly (P < 0.01), improve the ulcer healing probability, and reduce the level of TNF-α, VEGF levels and increase IL-2 level in the serum of rats with oral ulcers significantly (P < 0.01). Pathological examination showed that the lesion of ulcer tissue in each treatment group was obviously alleviated. Conclusion: Shuangjinlian mixture had anti-inflammatory effect and was effective for the prevention and treatment of oral ulcer in rats.     Abstract Purpose: Explore the effect of Baihe Dihuang powder on chronic stress depression rat models. Methods: Chronic stress depression rat models were established with different stimuli for 21 days. At the same time, the drug was administered for 21 consecutive days. The animals were weighed once a week after the start of the formal experiment. On the second day after the end of drug administration, conduct sugar water consumption test and open-filed box experiment, and conduct behavioral observation; At the end of behavioral testing, blood was taken from the eyeball and plasma was separated to measure MDA level and erythrocyte SOD activity; Take brain for homogenate, then measure the contents of 5-HT, NE and DA in brain tissue homogenate; Take the thymus and spleen, stained with 10% formalin fixation, embedding and HE staining, then use microscope to observe the histopathological changes. Results: Chronic stress depression rats model replicated successfully. Each group of given drugs could increase the weight, the consumption of sugar water, and improve the behavioral score, increase erythrocytes SOD activity and decrease MDA level of plasma, increase the content of 5-HT, NE and DA of brain homogenate, and improve the pathological changes of thymus and spleen of chronic stress depression model animals. Conclusion: Chronic stress depression rat model replicates successfully. Baihe Dihuang powder can interfere chronic stress depression rats model through different action pathways.     Abstract Aims: Explore the effect of compound lobelia oral liquid on acute pharyngitis rabbits model caused by ammonium hydroxide, evaluate its curative effect. Methods: Acute pharyngitis rabbit model was prepared by ammonia spray at pharyngeal. After 1 h of the last administration, score the pharyngeal tissue symptoms of rabbits under the condition of naked eyes (score according to the score criteria). Then, take blood from the marginal auricular vein for routine blood tests and serum TNF-α level measure. Take pharyngeal tissue for HE staining, and observe its pathological changes under the microscope. Results: Compared with the blank group, the pharyngeal tissue symptom score and serum TNF-α level in the model group were significantly increased (P < 0.01). The results of blood routine showed that the number of white blood cells and lymphocytes in the model group increased significantly (P < 0.01), and neutrophilic granulocyte percentage decreased significantly (P < 0.01). The infiltration of inflammatory cells in the pharynx tissue was obvious and pathological changes were observed. Compared with the model group, large, middle and small doses of compound lobelia oral liquid group could reduce the scores of pharyngeal tissue symptoms in rabbits with acute pharyngitis induced by ammonia. The large and middle dose compound lobelia oral liquid group could significantly reduce serum TNF-α level and neutrophilic granulocyte percentage, significantly increase the number of white blood cells and lymphocytes (P < 0.01); The small dose compound lobelia oral liquid group could significantly reduce the number of white blood cells and lymphocytes (P < 0.01), there was an increasing tendency to neutrophils and a decreasing tendency to serum TNF-α, but no statistical significance. The large, middle and small doses of compound lobelia oral liquid group all could improve the pathological changes of pharyngeal tissue inflammatory infiltration.
