Abstract Inflammasomes are key components of the innate immunity system that trigger the inflammatory response. Inappropriate activity of the inflammasome system has been linked to onset and perpetuation of inflammation in atherosclerotic plaques and cardiovascular disease. Low-to-moderate beer consumption is inversely associated with cardiovascular event presentation, while high levels of alcohol intake are associated with increased cardiovascular risk. Although fermented beverages have been suggested to exert their beneficial effects through their anti-oxidant and anti-inflammatory properties, little is known regarding the capacity of beer to modulate innate immunity cell responses. To this aim, primed or activated THP-1 macrophages were conditioned with human serum obtained from a prospective two-arms longitudinal crossover study to investigate the effect of a moderate and regular daily intake of beer, either alcohol-free or traditional, in the regulation of TLR-mediated inflammatory responses in healthy but overweight individuals. Conditioned macrophages with serum obtained after four-week intervention with alcohol-free beer significantly reduced the transcription of pro-inflammatory interleukins such as IL-1β and TNF. The serum of traditional beer consumers did not exhibit the same capacity as the serum of alcohol-free beer consumers to reduce gene expression of pro-inflammatory interleukins; however, serum from traditional beer consumers showed a regulatory effect at the protein level by significantly decreasing the intracellular protein levels of pro-IL-1β in primed macrophages and preventing cleaved-IL-1β protein release.

Abstract ROCK2 is a protein involved in the restructuring of the cytoskeleton in cell adhesion and contractibility processes. miR-138-5p and miR-455-3p regulate Rock2 expression, cell proliferation, migration, and invasion in different experimental cell models. However, their participation in the cytoarchitecture and mobility of B16F1 melanoma cells exposed to 5-Br-2'-dU is partially known. This work aimed to analyze ROCK2 and miRs 138-5p and 455-3p expression associated with morphological and mobility changes of B16F1 mouse melanoma cells exposed to the thymidine analog 5-Bromo-2'-deoxyuridine (5-Br-2'-dU). We observed an increase (2.2X n = 3, p < 0.05) in the cell area, coinciding with an increase in cell diameter (1.27X n = 3, p < 0.05), as well as greater cell granularity, capacity for circularization, adhesion, which was associated with more significant polymerization of F-actin, collapsed in the intermediate filaments of vimentin (VIM), and coinciding with a decrease in migration (87%). Changes coincided with a decrease in Rock2 mRNA expression (2.88X n = 3, p < 0.05), increased vimentin and a reciprocal decrease in miR-138-5p (1.8X), and an increase in miR-455-3p (2.39X). The Rock2 kinase inhibitor Y27632 partially rescued these changes. These results suggest ROCK2 and VIM regulate the morphological and mobility changes of B16 melanoma cells after exposure to 5-Br-2'-dU, and its expression may be reciprocally regulated, at least in part, by miR-138-5p and miR-455-3p.

Abstract Patients admitted for acute coronary syndrome (ACS) usually have high cardiovascular risk scores with low levels of high-density lipoprotein cholesterol (HDL-C) and high low-density lipoprotein cholesterol (LDL-C) levels. Here, we investigated the role of lipoprotein functionality as well as particle number and size in patients with a first-onset ACS with on-target LDL-C levels. Ninety-seven patients with chest pain and first-onset ACS with LDL-C levels of 100 ± 4 mg/dL and non-HDL-C levels of 128 ± 4.0 mg/dL were included in the study. Patients were categorized as ACS and non-ACS after all diagnostic tests were performed (electrocardiogram, echocardiogram, troponin levels and angiography) on admission. HDL-C and LDL-C functionality and particle number/size by nuclear magnetic resonance (NMR) were blindly investigated. A group of matched healthy volunteers ( n = 31) was included as a reference for these novel laboratory variables. LDL susceptibility to oxidation was higher and HDL-antioxidant capacity lower in the ACS patients than in the non-ACS individuals. ACS patients had lower HDL-C and Apolipoprotein A-I levels than non-ACS patients despite the same prevalence of classical cardiovascular risk factors. Cholesterol efflux potential was impaired only in the ACS patients. ACS-STEMI (Acute Coronary Syndrome—ST-segment-elevation myocardial infarction) patients, had a larger HDL particle diameter than non-ACS individuals (8.4 ± 0.02 vs. 8.3 ± 0.02 and, ANOVA test, p = 0.004). In conclusion, patients admitted for chest pain with a first-onset ACS and on-target lipid levels had impaired lipoprotein functionality and NMR measured larger HDL particles. This study shows the relevance of HDL functionality rather than HDL-C concentration in ACS patients.

Abstract PURPOSE: A wide number of research efforts have been carried out over the last decades to assess and improve system sustainability. Different methods and tools are increasingly implemented in product systems; nevertheless its application to services and other systems, in which products and services are combined, is rather limited. In this work, an approach to improve the sustainability of systems composed of products and services (P + S systems) is proposed and implemented in two case studies. METHODS: The sustainability improvement of P + S systems is addressed integrating different methods and approaches. The flows between system approach is used to identify links between all systems involved. The life cycle sustainability assessment (LCSA) method is applied to simultaneously evaluate environmental, economic and social aspects of sustainability with a life cycle perspective. In addition, sustainable design principles are taken into account to propose effective improvement strategies. The systematic development of four phases is required to carry out the integration of these approaches: (i) identification of links between involved systems; (ii) sustainability assessment of the P + S system; (iii) application of sustainability strategies; and (iv) sustainability results comparison. RESULTS AND DISCUSSION: Two different P + S systems are analysed. First, sustainability improvements in the service provided by a clothing boutique are proposed, taking into account that the store where the service is developed and the clothes acquired by the service provider are product systems involved in the service operation. Next, the sustainability of an itinerary exhibition is investigated. Products required for the exhibition space layout as well as the service used to transport all materials between different locations are taken into account. In each case, sustainability indicators obtained in both initial and alternative systems are compared, and variations detected due to strategies applied are analysed. CONCLUSIONS: Sustainability of P + S systems is evaluated using a set of quantitative indicators, the relative incidence of each system is determined and more sustainable alternatives are projected. Thus, this research supposes an advance towards the sustainable design of complex systems composed of products and services, since effects due to the interconnection between systems are studied and criteria to carry out sustainability-oriented decision making are provided.

Abstract Objective: The aim of this 2-arm parallel study was to evaluate the alignment efficiency of heat-activated nickel-titanium (NiTi-TE) and superelastic nickel titanium (NiTi-PSE) archwires over the first 3 months of orthodontic treatment and compare these groups.
Setting and sample population: Randomized, double-blind, controlled, single-center trial in 52 patients with fixed orthodontic appliances from an orthodontic graduate program in the permanent dentition and moderate crowding in the lower arch.
Material and methods: Patients were randomly allocated to one of two interventions: NiTi-TE and NiTi-PSE archwires, 0.014-inch (3M Unitek™, CA, USA) with a follow-up period of 3 months. The primary outcome was the alignment efficiency determined by the reduction in Little's irregularity index (mm), measured in three points, T0: before the start of orthodontic treatment, T1: 1 month later, T2: 2 months later, T3: 3 months later. Data were analyzed using independent sample t tests and repeated measures ANOVA.
    Results: 52 patients (NiTi-TE
Conclusions: NiTi-TE and NiTi-PSE wires of 0.014-inch were similar in their clinical efficiency for the resolution of crowding during the first 3 months of orthodontic treatment.

Abstract The development of product–service systems (PSS) is currently considered a promising solution to the challenge of sustainability. Nevertheless, the sustainability of these systems has not been systematically assessed and there is a need to develop more guiding principles. In this work, an approach based on the flows between product and service systems is used to facilitate both the definition of PSS boundaries and the identification of links between the systems involved. In addition, the life cycle sustainability assessment (LCSA) method is applied to simultaneously quantify environmental, economic and social impacts. Two cases are analysed. First, the production process of cow´s milk, in which a veterinary service is required, is studied using data measured from a dairy farm. Next, the sustainability of a clothing retail service taking, into account that a construction product is involved in its creation stage, is evaluated. In each PSS specific life cycle, stages are analysed, a functional unit referred to both products and services is defined, and quantitative indicators are selected to assess each sustainability dimension. The category of workers is selected to evaluate social aspects. The relative incidence of each system is evaluated and the impacts of different factors on the PSS sustainability are analysed.

Abstract The sustainability improvement of museums and exhibitions is a recent concern for multiple organisations. The application of sustainability criteria is one of the most important strategies of innovation in design activities, products, and service systems. This study analyses the sustainability of two alternatives to an itinerant cultural exhibition service. The exhibition travels to 12 destinations over 3 years and is within a space of 300 m 2 . In the first alternative, the contents are printed and exposed on a physical medium, and in the second, audiovisual media projects the contents on the walls. Life cycle sustainability assessment is applied to evaluate the impacts in the environmental dimension and the economic and social dimensions. The calculation of indicators, such as the greenhouse gas emissions, total costs, and working time, which are referred to each sustainability dimension, is conducted. A descriptive, comparative study was performed to identify the impact factors with a higher incidence. The results demonstrate that the audiovisual exhibition is more sustainable than the printed exhibition, with a difference of 8.7%, 7%, and 6.6% in GWP 100 , C E , and T W indicators, respectively.

Abstract Product and service development based on sustainable criteria is one of the poles of innovation in design activity. Public policies, consumer preferences, and corporate social responsibility lead to the growing importance of applying sustainability principles in the current design process. However, to make this practice widespread, sustainability assessment must be included in the curricula of bachelor’s and master’s degrees in design, so that the graduates have the precise tools to use when they start their professional careers. Furthermore, sustainable design methods have mainly been applied to improve only the environmental behavior of products. In this work, a methodology that simultaneously evaluates environmental, economic, and social aspects was applied to project more sustainable designs of products and services. This approach was implemented in higher education to develop Bachelor’s and Master’s degree final projects by design engineering students. Collaboration with different companies and institutions allowed the study of a number of cases. The production process of a cash management machine, the service provided by a public nursery school, and the development of an itinerary exhibition were addressed. In each case, product and service requirements were analyzed, sustainability indicators were obtained, and more sustainable designs could be proposed. This experience is also part of a global strategy at the University of Zaragoza to support the implementation of the UN’s Sustainable Development Goals, enhancing sustainability education. The intention of this paper is to present a methodology for more sustainable design, and examples of its application that other teachers can easily follow when teaching design of products or services.
