    Abstract Whey is a protein complex derived from milk, being a functional food with multiple health benefits. In this paper, the antigenotoxic potential of fresh bovine whey (FBW) in onion (Allium cepa) meristematic roots exposed to Quizalofop-P-tefuryl (QPt) herbicide was evaluated using the Allium assay. Firstly, the Allium cepa meristematic roots obtained after a short germination of 24 hours in distilled water were subjected to a pre-treatment with FBW in three different concentrations (500, 1000 and 2000 µl/L) for 24 hours. After that, there was a post-treatment with QPt herbicide (100, 500 and 1000 µl/L), for another 24 hours. All variants were tested alongside a negative control (onion root tips in distilled water) and a positive control (onion root tips treated with 500 µl/L QPt). The genotoxic effects of QPt were observed in all treatment variants, through the low rate of mitosis and through the induction of a large number of chromosomal and nuclear abnormalities (bridges, laggards, rings and strap nuclei). On the other hand, the fresh bovine whey improved the mitotic activity and reduced the index of chromosomal aberrations in variable percentages, in all treatment variants. These results suggest the cytoprotective potential of FBW against the cytotoxic and genotoxic effects of the tested herbicide. Although the mechanism of antigenotoxicity is unknown, it seems plausible that the whey protein acts as a blocking agent by chemical or physical interaction with the QPt components. Nevertheless, additional studies are needed to determine with certainty this potential.

    Abstract The chemical compounds from extracts of three Ranunculaceae species, Aconitum toxicum Rchb., Anemone nemorosa L. and Helleborus odorus Waldst. & Kit. ex Willd., respectively, were isolated using the HPLC purification technique and analyzed from a bioinformatics point of view. The classes of compounds identified based on the proportion in the rhizomes/leaves/flowers used for microwave-assisted extraction and ultrasound-assisted extraction were alkaloids and phenols. Here, the quantifying of pharmacokinetics, pharmacogenomics and pharmacodynamics helps us to identify the actual biologically active compounds. Our results showed that (i) pharmacokinetically, the compounds show good absorption at the intestinal level and high permeability at the level of the central nervous system for alkaloids; (ii) regarding pharmacogenomics, alkaloids can influence tumor sensitivity and the effectiveness of some treatments; (iii) and pharmacodynamically, the compounds of these Ranunculaceae species bind to carbonic anhydrase and aldose reductase. The results obtained showed a high affinity of the compounds in the binding solution at the level of carbonic anhydrases. Carbonic anhydrase inhibitors extracted from natural sources can represent the path to new drugs useful both in the treatment of glaucoma, but also of some renal, neurological and even neoplastic diseases. The identification of natural compounds with the role of inhibitors can have a role in different types of pathologies, both associated with studied and known receptors such as carbonic anhydrase and aldose reductase, as well as new pathologies not yet addressed.

    Abstract Dyes used in the food industry are an important class of food additives and are often used to make processed food more visually appealing, especially to children. The purpose of this paper was to evaluate the cytotoxic effect of Sunset Yellow (SY) and Brilliant Blue (BB) food dyes on the cell cycle of onion (Allium cepa L). The dye doses were 5, 10 and 20 mg SY/100ml distilled water respectively 15, 30 and 60 mg BB/100ml distilled water, at the exposure times of 24 hours in root tip cells of onion. Cytogenetic tests reveal a decrease of the mitotic index and the occurrence of various cellular aberrations following food dyes treatments. Decrease in mitotic index and increase of cellular aberrations it was concentration dependent. Types of cellular aberrations were varied; thus, were observed cells with irregular kinetics of chromosomes, ring chromosomes, laggard and sticky chromosomes and cells with micronuclei. Exposure of onion roots tips to both food dyes showed a large number of cells in prophase and low number of cells in anaphase at all treatment doses. The obtained results suggest caution in the consumption of foods that contain these two types of dyes and finding healthier alternatives for colouring of some food products like sweets, in attractive colours for consumers, especially for children.

    Abstract Used for hundreds of years in traditional medicine and ethnomedicine, the species of the genus Aconitum are the subject of many contemporary studies that focus on the biological and pharmacological functions of the active principles isolated (and purified) from extracts obtained from plants roots and/ or aerial parts. Well known for its strong neurotoxicity, aconitine and some other diterpenoid alkaloids extracted from Aconitum species or their derivatives have shown significant anticancer and apoptotic, analgesic, antimicrobial, antifungal, antidiarrheal and antioxidant potentials, as well as a high effectiveness in treating rheumatic fever, joint pains and some endocrine conditions. Strating from the toxic potential of these plants, phytochemical, pharmacological and toxicological studies are highly needed for improving their quality and safe usage.

    Abstract Climate conditions clearly influence the concentration of important substances in grapes, generating chemical reactions that will determine the final wine aromas. Three different regions were chosen to cover the most important viticultural areas from Romania. The aim of this study was to highlight, for the first time, the volatile profile for two Romanian white wines, Feteasca regala and Feteasca alba, from three different vineyards (Silagiu, Aiud, and Sarica Niculițel). The results showed that wine’s aromatic profile was directly proportional with the area of origin for the grapes, directly correlated to the climate. The obtained values for alcohols, esters, aldehydes, and terpenoid compounds were also related to the oenoclimatic aptitude index, a significant accumulation of aroma compounds being observed mainly for the Feteasca regala wine. A total of 17 superior alcohols were evidenced within the two types of wines, among them, 2-phenyl ethanol being distinguished by its higher level in all samples, varying from 7692 up to 11,783 µg/L. Together with some aromatic esters, it offers one of the most pleasant aromas, resembling rose flavour. Of all the acids found in wines, the succinic acid has the most intense flavour, tasting somehow bitter and salty, imprinting to wine a certain “juiciness” and “vinosity”. Diethyl succinate was one of the main esters in all six samples, with concentrations from 777 up to 1200 µg/L. Also, two terpenoid compounds and two aldehydes were found in all samples. The data obtained from PCA evaluation suggested that there is a significant variance among wine varieties. Nevertheless, hierarchical clustering was applied to explain the relationship between the six samples of wines, the smallest clusters that included Silagiu and Aiud winegrowing regions suggesting an increased similarity of the compositional profile.

    Abstract In the present study, the water quality was determined through cytogenotoxic endpoints by Allium cepa L. assay. Water samples were collected from nine sites selected according to the pollution level and large water discharge points into the Karaçomak River, Turkey. All of the tested water samples had a significant mitodepressive effect on the root tip cells of A. cepa L. They induced a significant increase in chromosomal aberrations like stickiness, anaphase bridges, laggards, and nuclear anomalies, i.e. binucleate cells, giant cells in vitro. It is suggested that the findings of the present study are very important to mitigate the environmental impacts caused by industrial operations on human and ecological health.

    Abstract The current study presents the research on the cytogenetic potential of water samples taken from the Argeş River, Romania. The water samples were collected from 7 sites selected dependant on the anthropic impact to which they were subjected. The genotoxic and cytotoxic potentials of the water samples were evaluated using Allium cepa L. as a test organism. The statistical interpretation of results indicated a significant reduction of the mitotic index and an important increase in the frequency of chromosomal and mitotic aberrations for most of the water samples taken for study.
