    Abstract The perception of orosensory stimuli, which includes flavor, can vary between individuals. These individual variations in oral sensations can be due to genetic factors and it would appear that they can predict food liking and consumption. The most studied source of variation is related to bitter taste perception associated with 6-n-propylthiouracil (PROP) responsiveness. In this context, humans can be classified as non-tasters (NT), medium tasters (MT) and supertasters (ST). Evidence suggests that genetic variation in bitter taste perception contributes to differences in the level of irritation caused by alcohol perception in solutions. The aim of this investigation was to study the bitter taste sensitivity among a group of mezcal consumers and its relationship with sensory perception and preference through PROP taster status. The tests were carried out in the state of Oaxaca in Mexico. A total of 83 mezcal consumers were classified by their PROP taster status and were asked to provide sensory descriptors for five mezcal samples and rate them according to the level of liking. The three-solution test was used to classify the subjects as NT, MT, and ST, while a Multiple Factor Analysis (MFA) was used to visualize the sensory descriptors provided by these three groups. The proportion of MT subjects was 16%, while the proportion of NT and ST was 34 and 51%, respectively. The MT provided higher liking ratings for at least three mezcal samples. According to MFA, the mezcal samples were organized in a similar configuration along the two dimensions. However, NT mentioned a limited number of simple terms (strong flavor, tasteless, burning in the mouth) to describe the samples, whereas ST used a more complex vocabulary (astringent, smoky, scratchy aftertaste). These data suggest that the preference for mezcal samples was similar for non-taster and supertasters, but there are indications that the sensory perception of mezcal differs between groups.

    Abstract We here report two new species of meiofaunal orbiniids belonging to the genus Questa Hartman, 1966, which have been discovered during benthic surveys conducted around the Azores and the Canary Islands. The new species, Q. manuelgerardi n. sp. and Questa ericnunezi n. sp., are described, figured and discussed in detail. Questa manuelgerardi n. sp., endemic from the Azores, is characterized by the lack of furcate chaetae and branchiae, and by the presence of two pairs of anal cirri. Questa ericnunezi endemic from the Canary Island, is defined by the presence of furcate chaetae, 12-18 pairs of branchiae, and two pairs of anal cirri. In addition, new records of the species Q. caudicirra Hartman, 1966 are provided. An identification key to the 12 known species of Questa is also included.

    Abstract Abstract: We report 8 children younger than 2 years who died from acute illicit fentanyl intoxications in Connecticut between 2020 and 2022.The Connecticut Office of the Chief Medical Examiner (CT OCME) investigates all unexpected, violent, and suspicious deaths in Connecticut. The CT OCME's electronic database was searched for fentanyl deaths by age. All underwent autopsies and toxicology testing.The ages ranged from 28 days to 2 years (mean age, 12 months). The causes of death involved acute fentanyl intoxications with 1 having xylazine, 1 having para-fluorofentanyl, and 1 having cocaine and morphine. All the manners of death were certified as homicide. The postmortem fentanyl blood concentrations ranged from 0.40 to 46 ng/mL. Most of the children were found unresponsive after being put to sleep. Three were co-sleeping with adults (2 in bed; 1 on a recliner). There was a known history of parental/caregiver drug abuse in 7 of 8 of the fatalities.We summarize the key investigative, autopsy, and toxicological findings. As illicit fentanyl use increases, there is a potential for infant exposure and death. The investigation and certification of these deaths and the role of intentional administration versus inadvertent exposure due to caregiver neglect in the context of the certification of the manner of death are described.

    Abstract Occupational exposure to airborne dust is responsible for numerous respiratory and cardiovascular diseases. Because of these hazards, air samples are regularly collected on filters and sent for laboratory analysis to ensure compliance with regulations. Unfortunately, this approach often takes weeks to provide a result, which makes it impossible to identify dust sources or protect workers in real time. To address these challenges, we developed a system that characterizes airborne dust by its spectro-chemical profile. In this device, a micro-cyclone concentrates particles from the air and introduces them into a hollow waveguide where an infrared signature is obtained. An algorithm is then used to quantitate the composition of respirable particles by incorporating the infrared features of the most relevant chemical groups and compensating for Mie scattering. With this approach, the system can successfully differentiate mixtures of inorganic materials associated with construction sites in near-real time. The use of a free-space optic assembly improves the light throughput significantly, which enables detection limits of approximately 10 µg/m

    Abstract Paraventricular thalamus (PVT) plays important roles in the regulation of emotion and motivation through connecting many brain structures including the midbrain and the limbic system. Although acetylcholine (ACh) neurons of the midbrain were reported to send projections to PVT, little is known about how cholinergic signaling regulates PVT neurons. Here, we used both RNAscope and slice patch-clamp recordings to characterize cholinergic receptor expression and ACh modulation of PVT neurons in mice. We found ACh excited a majority of anterior PVT (aPVT) neurons but predominantly inhibited posterior PVT (pPVT) neurons. Compared to pPVT with more inhibitory M2 receptors, aPVT expressed higher levels of all excitatory receptor subtypes including nicotinic α4, α7, and muscarinic M1 and M3. The ACh-induced excitation was mimicked by nicotine and antagonized by selective blockers for α4β2 and α7 nicotinic ACh receptor (nAChR) subtypes as well as selective antagonists for M1 and M3 muscarinic ACh receptors (mAChR). The ACh-induced inhibition was attenuated by selective M2 and M4 mAChR receptor antagonists. Furthermore, we found ACh increased the frequency of excitatory postsynaptic currents (EPSCs) on a majority of aPVT neurons but decreased EPSC frequency on a larger number of pPVT neurons. In addition, ACh caused an acute increase followed by a lasting reduction in inhibitory postsynaptic currents (IPSCs) on PVT neurons of both subregions. Together, these data suggest that multiple AChR subtypes coordinate a differential modulation of ACh on aPVT and pPVT neurons.

    Abstract Numerous experiments have shown that ocean acidification impedes coral calcification, but knowledge about in situ reef ecosystem response to ocean acidification is still scarce. Bahía Culebra, situated at the northern Pacific coast of Costa Rica, is a location naturally exposed to acidic conditions due to the Papagayo seasonal upwelling. We measured pH and pCO2 in situ during two non-upwelling seasons (June 2012, May–June 2013), with a high temporal resolution of every 15 and 30 min, respectively, using two Submersible Autonomous Moored Instruments (SAMI-pH, SAMI-CO2). These results were compared with published data from the 2009 upwelling season. Findings revealed that the carbonate system in Bahía Culebra shows a high temporal variability. Incoming offshore waters drive intra- and interseasonal changes. Lowest pH (7.8) and highest pCO2 (658.3 µatm) values measured during a cold-water intrusion event in the non-upwelling season were similar to those minimum values reported from upwelling season (pH = 7.8, pCO2 = 643.5 µatm), unveiling that natural acidification also occurs sporadically in the non-upwelling season. This affects the interaction of photosynthesis, respiration, calcification and carbonate dissolution and the resulting diel cycle of pH and pCO2 in the reefs of Bahía Culebra. During the non-upwelling season, the aragonite saturation state (Ωa) rises to values of > 3.3 and during the upwelling season falls below 2.5. The Ωa threshold values for coral growth were derived from the correlation between measured Ωa and coral linear extension rates which were obtained from the literature and suggest that future ocean acidification will threaten the continued growth of reefs in Bahía Culebra. These data contribute to building a better understanding of the carbonate system dynamics and coral reefs' key response (e.g., coral growth) to natural low-pH conditions, in upwelling areas in the eastern tropical Pacific and beyond.
