    Abstract The paper presents experimental data on the effect of different concentrations of the salts of cadmium, copper, and zinc on the adaptive potentialities of the most common opportunistic microflora in the Volga-Caspian region. The findings suggest that natural microorganisms (Aeromonas and Citrobacter) show a high metal resistance that seems to be associated with steadily increased background levels of these metals in the waters of the Volga-Caspian basin.

    Abstract In the article there are presented the results of a study of halotolerance of enterobacteria isolated from water and fish in the Volga River delta, which were dominant in the microbial landscape of these biotopes. Their halotolerance was studied by means of inoculation of daily pure cultures of meat--peptone broth with 3, 7 and 10% of sodium chloride and incubation at 37°C. At that all studied microflora was found to have significant indices of halotolerance with a predominance of aquatic isolates. At that in 3,0 and 7,0 % NaCl concentrations were very similar, but in 10% NaCl broth the halotolerance of water strain was almost 2.0 times higher than the in fish ones. Among isolated Enterobacteriaceae most halophilic bacteria were from the Proteus group, Citrobacteria and Enterobacteria. The halotolerance of isolated enterobacteria in hydroecosystem of the Volga River delta had seasonal specificity and dynamics. There was established the dynamic growth of this sign in the water and fish strains from spring to autumn. Thus, the analysis of the obtained results of the long-term conducted study has shown the wide dissemination of conditionally pathogenic enterobacteria in water and fish, and their high halotolerance and the ability of some of them to remain viable up to salt production. It gives grounds to recommend the necessity of the introduction of them into the normative documentation in the implementation ofthe sanitary-microbiological control of not only raw materials but also ready salted fish production.

    Abstract ANRIL (Antisense Non-coding RNA in the INK4 Locus, also known as CDKN2B-AS1) – 3.8-kb long non-coding RNA transcribed from the antisense strand of INK4b-ARF-INK4a gene cluster. It is known that ANRIL overexpression is associated with development of oncological pathologies of different localization. In addition, there are a number of studies devoted to role of ANRIL genetic polymorphism in emergence and progression of tumors, including tumors of genitourinary system.The aim of the study was to establish a possible association between rs4977574 ANRIL gene polymorphism and clear cell renal cell carcinoma (CCRCC) development in representatives of Ukrainian population which are smokers and non-smokers. Object and methods. Whole venous blood of 101 patients with CCRCC (42 women and 59 men) and 100 patients without oncopathology history (34 women and 66 men) was used in the study. DNA from blood white cells was extracted using GeneJET Whole Blood Genomic DNA Purification Mini Kit (Thermo Fisher Scientific, USA). Genotyping of rs4977574 ANRIL gene polymorphic locus was performed using real-time polymerase chain reaction (real-time PCR) method in the presence of TaqMan assay C_31720978_30. The mathematical data were processed using the SPSS software package (version 17.0). P values < 0.05 were considered as statistically significant. Results. It was found that difference in rs4977574-genotype distribution between patients with CCRCC and control persons was absent in general group (P = 0.216). At the same time, the statistical analysis stratified by smoking showed that both in non-smokers and smokers rs4977574-genotypes frequency also did not differ significantly between comparison groups (P = 0.511 and P = 0.099, respectively). However, after adjusting for age, gender, body mass index, and smoking habits statistically significant association between rs4977574 ANRIL gene polymorphism and risk of kidney cancer development was detected in smokers subjects under superdominant inheritance model (P = 0.043).     Abstract HOX antisense intergenic RNA (HOTAIR), the well-studied long noncoding RNA (lnRNA), realizes the regulation of cell cycle genes expression at the epigenetic and transcriptional levels. The results of experiments showed that HOTAIR promotes epithelial-mesenchymal transition (EMT) during the renal-cell carcinoma (RCC) development. Moreover, HOTAIR knockdown inhibits proliferation and invasion of RCC cells. At the same time the role of HOTAIR gene polymorphisms in kidney cancer development is unknown. The aim of the study was to perform a case-control study on representatives of both genders in order to assess the possible link between HOTAIR rs1899663 gene polymorphism and kidney cancer development in Ukrainian population. Object and methods. 141 unrelated Ukrainian patients (42 female, 59 male) with clear cell renal cell carcinoma (CCRCC) and 100 healthy subjects (34 female, 56 male) were enrolled to case-control study. HOTAIR rs1899663 polymorphism genotyping was performed using polymerase chain reaction-restriction fragment length polymorphism analysis (PCR-RFLP). The mathematical processing of obtained results was done using Statistical Package for Social Science software (SPSS, version 17.0, Chicago, IL, USA). All statistical tests were two-sided, P <0.05 was considered significant. Results. It was shown that ratio of HOTAIR rs1899663 GG-, GT- and TT-genotypes between CCRCC patients and control subjects did not differ (P = 0.207). Comparison of rs1899663 genotype frequencies in groups of different sex demonstrated no significant difference between CCRCC patients and control group in women (P = 0.067) and men (P = 0.248). The results of logistic regression analysis showed the association between HOTAIR rs1899663 polymorphic locus and CCRCC occurrence in female subjects under superdominant model of inheritance.     Abstract In the article there are presented the results of a study of antibiotic resistance of Enterobacteriaceae isolated from water of Volga River delta. Found that all the investigated microflora was established to have acquired multiple antibiotic resistance in dependence on the season of the year and the inhabited area. Minimal antibiotic resistance of Enterobacteriaceae was registered to Tobramycin and Chloramphenicol, the maximal resistance--to Benzylpenicillinum and Ampicillin. The data indicate to the significant persistence of antibiotic-resistant Enterobacteriaceae in hydroecosystems of Volga River delta which can be used as markers of sanitary-epidemiological and ecological trouble of the freshwater hydroecosystems.

    Abstract The results of γ-glutamyl carboxylase gene rs2592551 polymorphism determining in 170 patients with ischemic atherothrombotic stroke and 124 subjects without acute cerebrovascular disease (control group) have been evaluated. Obtained results revealed that rs2592551 polymorphism was related to ischemic stroke in Ukrainian population. The risk for this disease in patients with T/T genotype was higher than in major C-allele carriers (odds ratio (OR) = 3.117; 95% confidence interval (CI) = 1.016-9.566; P = 0.047). After dividing patients into subgroups, formed by the presence of certain risk factors for atherosclerosis, similar association has been established for women and non-smokers. At the same time, the heterozygous genotype (C/T) in females had significantly protective effect against ischemic stroke development when compared to C/C and T/T genotypes (OR = 0.460; 95% CI 0.213-0.994; P = 0.048). Statistical significance of these results persisted even after adjustment for age, body mass index, smoking and hypertension.

    Abstract The goal of this article is to discuss diagnostic criteria that are directly observable and at the same time allow to judge with sufficient reliability whether learning is activity-based not only in its project, but also in its implementation - in a cooperative child-adult action. Only one criterion is proposed here: children's initiative aimed at discovering and appropriating new concepts. In an attempt to elucidate the blurred meaning of such terms as “activity-based education”, “student as an agency (initiator) of learning activity” the authors suggest revising the subject matter behind these words, and for this purpose to visit the lessons and highlight the events that manifest children's learning initiative. The evidence from the literacy lessons taught according to the Elkonin’s ABC primer shows how the guesses and questions by individual students reveal to the observer that the students relate the concept mastered here and now and the concepts that belong to the same system but have not been discussed as yet in the classroom. Such transcending beyond the scope of the task as initially set by the teacher, discloses children's efforts to construct the concepts that outstrip the teacher's plan by several hours, weeks, and even months. As a rule, such initiatives are accompanied by expressed emotions of joy, surprise, pleasure, inspiration, interest, signifying their personal meaning for children.

    Abstract What are the conditions for the intersection and mutual enrichment of two separate lines of development of conceptual thinking – those of everyday and scientific notions? We assume that such intersection would require at least two conditions. The first one is well known: it is modeling of essential properties of the subject under study. Educational models are providing hand-on actions of the young student with a theoretical content of the notion. The second condition is the motivational support for constructing educational models. This condition is not fulfilled as evidenced by the lack of students’ initiative in creating and using the educational models at the early stages of the introduction of scientific concepts. We expected the conceptual characters of educational games to complement scaffolding for bringing the initial concepts into the systematic school curricula?. On the one hand, these conceptual characters act according to the logic of the concept, on the other hand, they exist as the fairy-tale heroes. Our hypothesis is supported by the evidence from the formative experiments in grade one. When the conceptual characters became part of the lessons, learning became impregnated with feelings, imagination and initiative; in particular, the first-graders were building the learning models without any suggestions from their teacher. The assessment of these students showed that the introduction of the initial concepts with the help of conceptual characters significantly affected the very concepts thus developed; in particular, it promoted their attribution to the essential properties of the subject under study.

    Abstract On the basis of long-term (1995–2010 and 2013–2014) microbiological monitoring of water and pike perch in the various water areas of the Volga Delta, four dominant groups of bacteria from the family Enterobacteriaceae and genera Aeromonas, Flavobacterium, and Pseudomonas were identified. The results of studies on the species composition of water microflora and fish from natural populations and artificial breeding are summarized. The prevalence of potentially pathogenic microflora in the water and fish over the indicative ones has been shown. Water temperature influences the seasonal cycles of microorganisms and their pathogenicity factors, which determine their fast adaptation to living in different ecological niches.

    Abstract The paper gives data obtained over many years on the quantity of bacteria of the genus Aeromonas in the hydroecosystems of the delta of the Volga and Caspian Sea and in commercial fishes (sturgeon (Acipenseridae), pike perch (Stizostedion), carp (Cyprinus carpio), silurus (Silurus glanis), etc.). It provides a detailed analysis of their seasonal trends and spatial distribution, markers of their pathogenicity, and antibiotic resistance. The findings suggest that Aeromonas is a causative agent of sapronoses and meets the criteria for natural focal infections. They show a wide range of tolerance to the abiotic factors of hydroecosystems, as well as mixotrophy and autrophic nutrition in the environment and they are of great sanitary and hygienic importance.
