    Abstract We highlight the critical role of data in developing sustainable combustion technologies for industries requiring high-density and localized energy sources. Combustion systems are complex and difficult to predict, and high-fidelity simulations are out of reach for practical systems because of computational cost. Data-driven approaches and artificial intelligence offer promising solutions, enabling renewable synthetic fuels to meet decarbonization goals. We discuss open challenges associated with the availability and fidelity of data, physics-based numerical simulations, and machine learning, focusing on developing digital twins capable of mirroring the behavior of industrial combustion systems and continuously updating based on newly available information.

    Abstract For patients with fulminant liver failure and end-stage liver disease, liver transplantation remains the only effective treatment. Over the years, as a result of the ageing population, the average age of liver transplant donors and recipients has increased and currently about one quarter of patients receiving transplantation in the United States are above the age of 65. Recently, a study reported that patients aged 65 years or older had lower one-year survival compared to a younger cohort. Herein, we express our opinion about this interesting publication.

    Abstract Personality disorders can influence and, along with cognitive deficits, compromise the quality of life of patients with epilepsy. This study evaluated personality traits and disorders in patients with frontal (FLE) or temporal lobe epilepsy (TLE) using the Millon Clinical Multiaxial Inventory-III with the aim to determine prevalent personality profiles. The results demonstrate the presence of particularly pronounced personality traits and disorders with prevalence of histrionic and obsessive-compulsive personality profiles, respectively, in FLE and TLE. These profiles may be related to different effects of pathophysiological and clinical aspects.

    Abstract Osteoarthritis (OA) is a degenerative and progressive joint disease. When all three compartments are involved, end-stage OA is treated with a total knee arthroplasty (TKA). Unicompartmental knee arthroplasty (UKA) is a primary treatment for isolated osteoarthritis. UKA has a quicker recovery time than TKA, as well as less morbidity and more tissue sparing. At the time of surgery, 17% of patients have a tricompartmental disease and most patients with a Kellegren-Lawrence grade >3 have an intact anterior cruciate ligament (ACL). Conventional TKA sacrifices the ACL. Patients with concurrent medial and lateral osteoarthritis and a functional ACL may receive a primary bi-unicondylar arthroplasty. Combined partial knee arthroplasty (CPKA) is an established practice either in bicompartmental femoro-tibial OA or in OA progression after UKA, with the addition of another UKA. A conversion of a lateral UKA to a tricompartmental joint replacement has been reported in the literature. In our case report, we describe a one-stage hypoallergenic tricompartmental UKA, with improved clinical score and no sign of early failure at the last follow-up.

    Abstract A fundamental hindrance to building data-driven reduced-order models (ROMs) is the poor topological quality of a low-dimensional data projection. This includes behavior such as overlapping, twisting, or large curvatures or uneven data density that can generate nonuniqueness and steep gradients in quantities of interest (QoIs). Here, we employ an encoder-decoder neural network architecture for dimensionality reduction. We find that nonlinear decoding of projection-dependent QoIs, when embedded in a dimensionality reduction technique, promotes improved low-dimensional representations of complex multiscale and multiphysics datasets. When data projection (encoding) is affected by forcing accurate nonlinear reconstruction of the QoIs (decoding), we minimize nonuniqueness and gradients in representing QoIs on a projection. This in turn leads to enhanced predictive accuracy of a ROM. Our findings are relevant to a variety of disciplines that develop data-driven ROMs of dynamical systems such as reacting flows, plasma physics, atmospheric physics, or computational neuroscience.

    Abstract Building the inner layer of our blood vessels, the endothelium forms an important line communicating with deeper parenchymal cells in our organs. Previously considered passive, endothelial cells are increasingly recognized as key players in intercellular crosstalk, vascular homeostasis, and blood fluidity. Comparable to other cells, their metabolic function strongly depends on mitochondrial health, and the response to flow changes observed in endothelial cells is linked to their mitochondrial metabolism. Despite the direct impact of new dynamic preservation concepts in organ transplantation, the impact of different perfusion conditions on sinusoidal endothelial cells is not yet explored well enough. This article therefore describes the key role of liver sinusoidal endothelial cells (LSECs) together with their mitochondrial function in the context of liver transplantation. The currently available ex situ machine perfusion strategies are described with their effect on LSEC health. Specific perfusion conditions, including perfusion pressure, duration, and perfusate oxygenation are critically discussed considering the metabolic function and integrity of liver endothelial cells and their mitochondria.

    Abstract Adverse childhood experiences (ACEs) refer to negative events of childhood. Research has demonstrated relationships between ACEs and adult mental and physical difficulties. Fewer studies have examined potential moderators of these relationships. This study assessed the role of character strengths in the relationship between ACEs and negative physical and mental health outcomes in adulthood. An online sample of 1,491 adults completed questionnaires measuring character strengths, exposure to ACEs, and aspects of physical/mental health. Results replicated previous findings that ACEs and character strengths are each meaningfully related to health outcomes. Gratitude and self-regulation were generally indicative of better health outcomes, kindness and appreciation of beauty of poorer outcomes. Character strengths remained meaningful correlates of adult behavioral and emotional health even when controlling for ACEs. Character strengths did not moderate the relationship between ACEs and health, suggesting that character strengths do not mitigate the effects of ACEs, but do make an independent contribution to physical and mental health.

    Abstract Liver transplantation is the only lifesaving procedure for children with end-stage liver disease. The field is however heterogenic with various graft types, recipient age and weight and underlying diseases. Despite recently improved overall outcomes and the expanded use of living donors, waiting list mortality remains unacceptable particularly in small children and infants. Based on the known negative effect of elevated donor age, higher body mass index, and prolonged cold ischemia time, the number of available donors for pediatric recipients is limited. Machine perfusion has regained significant interest in the adult liver transplant population during the last decade. Ten randomized controlled trials are published with an overall advantage of machine perfusion techniques over cold storage regarding post-operative outcomes, including graft survival. The concept of hypothermic oxygenated perfusion (HOPE) was the first and only perfusion technique used for pediatric liver transplantation today. In 2018 the first pediatric candidate received a full-size graft donated after circulatory death with cold storage and HOPE, followed by a few split liver transplants after HOPE with an overall limited case number until today. One series of split procedures during HOPE was recently presented by colleagues from France with excellent results, reduced complications, and better graft survival. Such early experience paves the way for a more systematic use of machine perfusion techniques for different graft types for pediatric recipients. Clinical reports of pediatric liver transplants with other perfusion techniques are awaited. Strong collaborative efforts are needed to explore the effect of perfusion techniques in this vulnerable population impacting not only the immediate posttransplant outcome, but the development and success of an entire life.

    Abstract The last decade has been notable for increasing high-quality research and dramatic improvement in outcomes with dynamic liver preservation. Robust evidence from numerous randomized controlled trials has been pooled by meta-analyses, providing the highest available evidence on the protective effect of machine perfusion (MP) over static cold storage in liver transplantation (LT). Based on a protective effect with less complications and improved graft survival, the field has seen a paradigm shift in organ preservation. This editorial focuses on the role of MP in LT and how it could become the new "gold standard". Strong collaborative efforts are needed to explore its effects on long-term outcomes.
