    Abstract The purpose of study is to evaluate associations of polymorphism of genes encoding components of renin-angiotensin system and indices of echocardiography in examined patients with rheumatic heart disease (RHD). The sampling consisted of 128 patients with RHD and average age 58,96±0,34 years. The echocardiography was implemented using Philips Affinity 50 machine. The genetic typing was carried out according polymorphic markers Thr174Met, Met235Thr and А1166С by polymerase chain reaction in real-time with electrophoretic scheme of detecting the result of "SNP-EXPRESS". The heterozygosis of Thr174Met results in larger dilatation of left sections of heart. The Thr174Thr homozygotes characterized by large linear dimensions of right heart. The Thr235Thr homozygotes characterized by minimal parameters reflecting hypertrophy of left ventricle of heart. There was no significant difference in echocardiography indices in patients with A1166A polymorphism. The mutations in codon 235 and 174 did not affect distance of 6-minute walk test. The statistically significant difference in the distance of 6-minute walk test was obtained only for А1166С: the minimum distance indicators is in the group of A1166C heterozygotes - 291,46 (273,83-309,09) meters and the maximum distance in С1166С homozygotes - 357,20 (309,21-405,19) meters. In patients with RHD, Thr174Met heterozygosis can lead to dilatation of left heart camera, while homozygosis for Thr174Mr leads to dilatation of right heart camera and left ventricular hypertrophy. The Met235Thr heterozygosis, on the contrary, are characterized by minimal sizes of left atrium and right ventricle. Polymorphism of the A1166C gene encoding angiotensin type I receptor had practically no effect on echocardiography indices in patients with RHD.

    Abstract Objective - to outline the basic ideas about the conditions and results of the burning of human corpses and the potential possibilities of a forensic medical examination of the burned remains. It is impossible to burn the corpse of an adult to a state of only homogeneous ash not only on the fire, but also in special crematorium chambers. After the burning of the corpse, ash and bone fragments remain in a state of black, gray and white heat. The state of the cremated remains allows, at least, establishing the number of incinerated corpses, their species and anatomical affiliation. Reconstruction of the conditions of corpse burning is possible only within the framework of a comprehensive forensic examination with the participation of specialists in the field of fire-technical affairs and forensic medical experts. The exclusive competence of specialists in the field of fire-technical affairs is the verification of the conditions of combustion (type and volume of fuel, technical features of combustion, duration of cremation, etc.).

    Abstract To search for effective and safe methods aimed at reducing the frequency of surgical treatment for nodular thyroid pathology. 1999-2019 literature review conducted using NLM databases. There is more and more evidence in the literature on the efficacy and safety of minimally invasive treatments for thyroid nodular disease. Based on the literature data, it was revealed that the most effective technique for minimally invasive treatment is laser thermal ablation of thyroid nodules under visual control. This technique can be considered an alternative to surgical intervention in the treatment of benign thyroid nodules and, in most cases, leads to a decrease or complete disappearance of the nodule, clinical manifestations, or cosmetic defects. Unlike surgical treatment, it is practically devoid of the likelihood of formidable complications and, as a rule, passes without any threatening consequences for the patient. While the use of laser thermal ablation is expanding, the international medical community has begun to incorporate this technique into treatment guidelines and is making efforts to disseminate it in general clinical practice. Since the method of laser thermal ablation can be considered an effective alternative to surgical intervention for the treatment of benign thyroid nodules, it is necessary to widely introduce it into the general clinical practice in Russia and algorithms for providing medical care to patients with nodular goiter. The introduction of imaging-guided laser thermal ablation into clinical practice and its inclusion in the algorithms for the provision of medical care to patients with thyroid nodular pathology will significantly improve the quality and availability of medical care for patients with thyroid nodular pathology.

    Abstract The ability of β-CD to form inclusion complexes with ibuprofen (IBU) and at the same time to make a two-phase system with citric acid was explored in the present study for achieving improved solubility and dissolution rate of IBU. Mechanical milling as well as mechanical milling combined with thermal annealing of the powder mixtures were applied as synthetic methods. Solubility and dissolution kinetics of the complexes were studied in compliance with European Pharmacopoeia (ICH Q4B). β-CD and citric acid (CA) molecules were shown to interact by both ball milling (BM), thermal annealing, as well as BM with subsequent annealing. Complexes were also formed by milling the three compounds (β-CD, CA and IBU) simultaneously, as well as by a consecutive first including IBU into β-CD and then binding the formed β-CD/IBU inclusion complex with CA. As a result, ternary β-CD/IBU/CA complex formed by initial incorporation of ibuprofen into β-CD, followed by successive formation of a two-phase mixture with CA, exhibited notably improved dissolution kinetics compared to the pure ibuprofen and slightly better compared to the binary β-CD/IBU system. Although the addition of CA to β-CD/IBU does not significantly increase the solubility rate of IBU, it must be considered that the amount of β-CD is significantly less in the ternary complex compared to the binary β-CD/IBU.

    Abstract The plasma membrane Pma1 H+-ATPase of the yeast Saccharomyces cerevisiae contains conserved residue Asp739 located at the interface of transmembrane segment M6 and the cytosol. Its replacement by Asn or Val (Petrov et al. (2000) J. Biol. Chem., 275, 15709-15716) or by Ala (Miranda et al. (2011) Biochim. Biophys. Acta, 1808, 1781-1789) caused complete blockage of biogenesis of the enzyme, which did not reach secretory vesicles. It was proposed that a strong ionic bond (salt bridge) could be formed between this residue and positively charged residue(s) in close proximity, and the replacement D739A disrupted this bond. Based on a 3D homology model of the enzyme, it was suggested that the conserved Arg811 located in close proximity to Asp739 could be such stabilizing residue. To test this suggestion, single mutants with substituted Asp739 (D739V, D739N, D739A, and D739R) and Arg811 (R811L, R811M, R811A, and R811D) as well as double mutants carrying charge-neutralizing (D739A/R811A) or charge-swapping (D739R/R811D) substitutions were used. Expression of ATPases with single substitutions R811A and R811D were 38-63%, and their activities were 29-30% of the wild type level; ATP hydrolysis and H+ transport in these enzymes were essentially uncoupled. For the other substitutions including the double mutations, the biogenesis of the enzyme was practically blocked. These data confirm the important role of Asp739 and Arg811 residues for the biogenesis and function of the enzyme, suggesting their importance for defining H+ transport determinants but ruling out, however, the existence of a strong ionic bond (salt bridge) between these two residues and/or importance of such bridge for structure-function relationships in Pma1 H+-ATPase.
