Abstract This study found significant differences between the standard length distributions of humpback and redfin culter and between Mongolian culter and topmouth culter. A stable isotope analysis (δ13C and δ15N) was used to investigate the interspecies differences between the feeding habits and trophic niches of four carnivorous species of Cultrinae fish. The results showed that the δ13C and δ15N values were significantly correlated with the standard length and species. However, the δ13C values of humpback and redfin culter were not significantly different, suggesting that these two fish species had similar food sources. The δ15N values of Mongolian culter and topmouth culter were the highest, suggesting that they occupied a higher trophic level and that animal prey was more important in their diets. Moreover, variations in δ13C and δ15N indicated considerable niche overlap and interspecific competition among the four species. We also evaluated trophic niches, diversity, redundancy, and evenness utilizing isotopic niche metrics, and we estimated asymmetrical niche overlaps. The analysis revealed that four carnivorous species of Cultrinae displayed similar trophic niche sizes and trophic diversity. The trophic traits of topmouth culter clearly defined them as a trophic generalist in terms of the inter-individual variability in their isotopic niches. A significant finding was that the average niche overlap between them was as high as 64.02%; topmouth culter had the greatest trophic overlap with redfin culter (95.52%) and humpback (90.38%), followed by Mongolian culter onto redfin culter (85.32%), indicating that topmouth culter and Mongolian culter benefit the most from the food supply in the habitat, or they are more competitive in the presence of limited resources.

Abstract The Yangtze finless porpoise ( Neophocaena asiaeorientalis ) is the sole freshwater subspecies of Neophocaena phocaenoides , and there is a lack of data on its transcriptome. In this study, we applied RNA-seq technology to assemble, de novo, a transcriptome and analyzed differential expressed genes (DEGs). About 6 Gb of clean data was generated for the Yangtze finless porpoise blood ( n = 6) through de novo sequencing. In total, 151,211 unigenes were generated and a total of 119,039 of these unigenes (78.72%) were functionally annotated when searched for within the NCBI Nr, SwissProt, GO, COG, and KEGG databases. Diverse and extensive expressed gene catalogs were sampled for the Yangtze finless porpoise. DESeq2 was used to analyze the differential expression genes (DEGs) obtained from the assembled transcriptome. The results indicated that DEGs have close relationships with the Yangtze finless porpoise’s development, evolution and adaptation. Further, we found that genes involved in cetacean TAG synthesis might directly explain the molecular basis of cetacean blubber thickening. These transcriptome data will assist in understanding molecular mechanisms of Yangtze finless porpoise adaptation.

Abstract The global climate and environmental variability can induce fish adaptive strategy change and form a corresponding complementary mechanism subsequently. Because of climate fluctuation, human activities, and water conservancy projects, it has been reported that natural fish resources of the silver carp have been declining in a wavy trend from 1950. However, few studies have explored the potential effects of determining the relationship between the adult fish and fish larvae. Using the field survey method, this study analyzed the fisheries resource status of the adult silver carp and its early life stage in the Yangtze River. Results indicated that different geographic populations showed significant habitat dependence, and the adult silver carp tends to choose habitats with slow water flow, rich biological bait, and less human interference. What is more, its distribution pattern has regional and seasonal differences obviously. Additionally, redundancy analysis on the fish larvae showed that water temperature and water flow are the two most important factors influencing the fish larvae blooming. According to the generalized additive model (GAM), the hydrological factors that significantly influence the larvae abundance are water temperature, transparency, daily increasing rate of water level, and discharge (p < 0.05). Combining the historical and present research data, the results indicated that river and lake connected habitat is essential for the silver carp recruitment and migration. Future studies should focus on the complementary mechanism of silver carp both in the natural habitat and invasion waters. It is suggested that the spawning habitat should be well protected during the channel project and economic belt construction along the Yangtze River.

Abstract The temporal variation of sedimentary organic matters (SOM) in the East China Sea (ECS) is not fully assessed due to the complicated natural and anthropogenic processes. In this study, the compositions and distributions of multi-biomarker indicators (n-alkanes, n-alkanols and specific sterols) in a long sediment core collected from the Min-Zhe fined-grained sediment zone of the ECS were systematically studied to reveal variations of SOM sources and phytoplankton records over the last decades. The results of n-alkanes and sterols have demonstrated that terrestrial plants are the dominant SOM source, with the mean proportion of 67%. The temporal variation of terrestrial to marine biomarker ratios (TMBR) agree well with that of the dominant n-alkanes and terrigenous/aquatic ratios (TAR), suggesting that the multi-biomarker indicators yield a better assessment of the SOM source input. The regular variations of terrestrial OM and phytoplankton signals during the past decades record the interaction among marine ecological effects, anthropogenic activities and natural processes (i.e., extreme climate events). As a whole, the elevated productivity of marine phytoplankton since 1952 (mainly caused by enhanced nutrient inputs owing to the intensified human activities) and the decline in the period of 2005–2011 (likely influenced by the water impoundment of the Three Gorges Dam in 2003) were well reconstructed on the basis of brassicasterol (B) and dinosterol (D) indicators in this study. The B/(B + D) ratios also reveal the changes of community structure as a result of variations in the nutrient structure controlled by anthropogenic forcing. Notably, the anomalous peak in the abundance of n-alkanes with the even carbon predominance in about 1990 is identified for the first time, which may occur at that time of the first heavy rainfall after a long drought and is probably of the origin of biomass/fossil fuel combustion.

Abstract Although various geochemical and geophysical investigations have already indicated a great resource potential in the Xisha area of the South China Sea, the origin of organic matter and molecular evidence for tracing the migration of hydrocarbons from deep petroleum reservoirs are still lacking. In this study, systematic organic geochemical analyses, including bulk organic matter parameters and lipid biomarkers were performed for deep sediments from two cores. The C/N ratios and δ 13 C and δ 15 N values of organic matter in most of the samples, together with the maxima of short-chain n -alkanoic acids and mid-chain n -alkanols, high abundances of monounsaturated fatty acids C 18:1ω9 and C 22:1ω13 , jointly indicate the dominance of marine organic matter. n -Alkanes in sediments from core GMGS4-XH-W06B are characterized by small unresolved complex mixture (UCMs) humps, high odd/even predominance (OEP) and carbon preference index (CPI) values, clearly exhibiting characteristics of modern sediments. However, the sediments for core GMGS4-XH-W03B are featured with big UCMs, associated with OEP and CPI values around 1.0, showing signatures of petroleum hydrocarbons from high maturity sources. Considering the geologic background, the biomarker signatures are solid evidence for indicating the existence of underlying petroleum reservoirs, and may provide the valuable information for assessing the hydrocarbon resources in the Xisha area.

Abstract Estuary river–lake (ERL) complex is critical for the development of ichthyoplankton, which plays an essential role in the breeding, nursing, and recruitment of freshwater fish species. In this study, different ERL sections were selected to investigate the ichthyoplankton assemblage from the Changjiang River estuary to its Hukou section between May 2018 and August 2020. During the study period, fish larvae of 40, 35, 54, and 46 fish species were sampled in the Rugao (RG), Nanjing (NJ) (nearby estuary), Anqing (AQ), and Hukou (HK) (connected river–lake) sections in the Changjiang River, respectively. Results on ichthyoplankton surveys revealed fluctuating assemblage patterns in the different river sections; larval fish species composition presented ecological habitat-dependent characteristics, which supported the importance of ERL complex for ichthyoplankton development. Furthermore, the density of fish larvae was negatively correlated with water transparency but positively correlated with water temperature, the daily rate of water level increase, and runoff in the limitative range. A generalized additive model analysis indicated that the hydrological factors significantly affecting larval fish abundance are water temperature, transparency, the daily rate of water level increase, and runoff (p < 0.05). These results also indicated that river estuaries and river–lake connected complexes are essential for ichthyoplankton recruitment and migration. In combination with the historical record, ichthyoplankton assemblage’s patterns in the Changjiang River lower reaches were preliminarily elucidated, although additional works are needed such as assessing effects on their survival and recruitment.

Abstract The spatial distribution of fish individuals is affected by habitat conditions and species ecological characteristics, and it also reflects the longtime adaptation to habitat at the phenotypic and genotypic level. As a typical river-lake migratory fish species, the silver carp habitat selection was determined by its migration preference and genetic features. In this study, 15 microsatellite fluorescent markers combined with capillary electrophoresis were used to analyze the genetic diversity, genetic differentiation, and structure of nature silver carp populations in the Sanzhou (SZ), Hukou (HK), Anqing (AQ), Zhenjiang (ZJ), and Rugao (RG) sections of the Yangtze River. The results showed that 15 microsatellite loci exhibited medium to high polymorphisms. The overall genetic diversity in the Yangtze River was high, with the average value of Shannon’s information index ranging from 1.559 to 1.668. The numbers of alleles (Ne) ranged from 1.630 to 10.100. The expected heterozygosity (He, 0.690–0.721) was higher than observed heterozygosity (Ho, 0.598–0.646), and the genetic variation mainly originated from within the population (94.69%). However, the entire population was in the state of heterozygous deletion, and HK, RG populations encountered the risk of inbreeding risk (F > 1). Interestingly, there was a distinct genetic structure for the population in the HK section, which indicated that local population has occurred to the silver carp in this river section, and they may also possess aggregation characteristics specific to the river-lake-connected (RLC) habitat. The results mostly support the conclusion that the RLC habitat is essential for geographic population formation. The potential impact of special habitats on natural populations should be considered, and continuous surveys on population dynamics should be performed.

Abstract Abstract Background Coilia nasus oogenesis/spawning migration is a well-defined synchronous arrangement process. DnaJs are indispensable molecular chaperones for oogenesis process. However, how DnaJs involved the anadromous spawning migration mechanism is outstanding and plausible. Results In this regard, two DnaJs (Cn-DnaJa1 and Cn-DnaJb1) are cloned from the Coilia nasus’s ovary. Their structure both contains J domain, G/F domain and ZF domain. Their mRNA transcripts were found extensively expressed in all the sampled tissues and significantly highly in gonads, which probably mean that DnaJs involved in C. nasus’s gonad development basal metabolic processes. In the process of spawning migration, Cn-DnaJa1 and Cn-DnaJb1 mRNA transcripts were also expressed with significant differences during oogenesis with highest levels in the development phase, and maintaining high levels during the multiplication, mature and spawning phase. Further study showed that the DnaJa1and DnaJb1protein have high distribution in the onset phase and mainly distributed in the oocyte cytoplasm especially during the migration development phase’s. Conclusions This experiment study demonstrated that DnaJs participate in reproductive regulation during the spawning migration process in C. nasus and possibly play a vital role in the ovary development process. These findings also provided a base knowledge for further molecular mechanism study of spawning migration.

Abstract HSP90β1 (known as glyco-protein 96, GP96) is a vital endoplasmic reticulum (ER) depended chaperonin among the HSPs (heat shock proteins) family. Furthermore, it always processes and presents antigen of the tumor and keeps balance for the intracellular environment. In the present study, we explored the effect of tributyltin chloride (TBT-Cl) exposure on HSP90β1 expression in river pufferfish, Takifugu obscurus. The full length of To-HSP90β1 was gained with 2775 bp in length, with an ORF (open reading frame) encoding an 803 aa polypeptide. A phylogenetic tree was constructed and showed the close relationship to other fish species. The HSP90β1 mRNA transcript was expressed in all tissues investigated with higher level in the gill and liver. After the acute and chronic exposure of TBT-Cl, the To-HSP90β1 mRNA transcript significantly was up-regulated in gills. Moreover, the histology study indicated the different injury degree of TBT-Cl in liver and gill. Immunohistochemistry (IHC) staining results implied the cytoplasm reorganization after TBT-Cl stress and the function of immunoregulation for To-HSP90β1 to TBT-Cl exposure. All the results indicated that HSP90β1 may be involved in the resistance to the invasion of TBT-Cl for keeping autoimmune homeostasis.

Abstract Heat shock proteins play essential roles in basic cellular events. Spawning migration is a complex process, with significant structural and biochemical changes taking place in the adult gonad. To date, the molecular mechanisms underlying migration reproductive biology remain undetermined. In this regard, a full length HSP90AA1 comprising 2608 nucleotides from the anadromous fish Coilia nasus was characterized, encoding 742 amino acid (aa) residues with potential phosphorylation sites. HSP90AA1 mRNA transcripts were detected in all organs, especially in the gonad. Furthermore, the greatest transcript levels were found during the developmental phase, while the lowest levels were found during the resting phase. In addition, the strongest immunolabeling positive signal was found in the primary spermatocyte and oocyte, with lower positive staining in secondary germ cells, and a weak or absent level in the mature sperm and oocyte. Interestingly, HSP90AA1 was mainly located in the cytoplasm of germ cells. These results are important for understanding the molecular mechanism of anadromous migration reproductive biology. In combination with data from other fish species, the result of this present study may facilitate further investigations on the spawning migration mechanism.
