    Abstract The aquaculture industry is facing a growing need for a cost-effective, sustainable, and highly nutritious alternative to fish meal (FM) due to the increasing gap between supply and demand. This study investigated the effect of substituting (FM) with probiotics (Spirulina platensis) in fish feed on the survival, growth, and biochemical parameters of Oreochromis niloticus. Live tilapia fingerlings were stocked in 100-l glass aquaria. The feeding regimen consisted of four controls: commercial feed (C), formulated feed with FM (FMR0), and formulated feeds with 25% and 50% replacement of FM (FMR25, and FMR50), respectively. The trial groups included FMR25S5 (25% FM replacement with 5% Spirulina), FMR50S5 (50% FM replacement with 5% Spirulina), FMR25S10 (25% FM replacement with 10% Spirulina), and FMR50S10 (50% FM replacement with 10% Spirulina). The trial period lasted for 24 weeks. Weight gain (WG) was most pronounced in FMR50S10 (p < 0.05). The SGR (specific growth rate) was highest for FMR50S10, FMR50S5, and FMR50. A similar pattern was exhibited in the FMR50S10 group for length gain (LG). The protein efficiency ratio (PER) values were maximum in FMR50, FMR50S5, and FMR50S10. FMR25S10, FMR0, and FMR25 exhibited the highest feed intake (FI) (p < 0.05). Trial diets supplemented with Spirulina enhanced the activities of SOD, CAT, and MPO. FMR25S10 exhibited the highest GPx activity and appreciable flesh quality. Collectively, S. platensis enhanced growth and feed utilization and modulated biochemical endpoints in feeds.

    Abstract Bisphenol A(BPA) is a non-persistent, anthropic compound and continues to be ubiquitous in the environment. Since BPA is estrogenic in nature, its exposure induces adverse cellular responses associated with oxidative stress and inflammation. Hesperetin, a major flavonoid present in lemons and sweet oranges exhibits a broad spectrum of pharmacological effects attributed to its potent anti-inflammatory and antioxidant status. The current study reports the ‘green’ synthesis of nano hesperetin (NHsp) followed by characterization that included; Fourier Transform Infrared spectroscopy (FTIR), X-Ray Diffraction (XRD), Transmission Electron Microscopy (TEM), Scanning Electron Microscopy (SEM) and Dynamic Light Scattering (DLS). In accordance with the findings, the mean size of the synthesized nanoparticles was 139.9 nm with a PDI of 0.362. The morphotype was primarily spherical and rod shaped. This was followed by a comparative assessment of the ameliorative efficacy of bulk and nano hesperetin against BPA-induced hepatotoxicity in rat. The findings showed that post-exposure to 250 mg/kg of BPA for 21 days significantly enhanced the pro-inflammatory cytokines (IL-6, IL-8, IL-1β, TNF-α), in the serum with a concomitant upregulation in the mRNA expression of Nrf 2(Nuclear factor erythroid 2-related factor) factor in the liver. Treatment with both bulk hesperetin (Hst) and nano hesperetin (NHst) reduced the levels of cytokines and expression of hepatic Nrf2 factor. This was paralleled by the modulatory effects on the histology of liver and kidney which were reversed on treatment. Taken together, the nano hesperetin was more efficacious in alleviating the BPA-induced toxicity. Thus,hesperetin and its nano formulation could be potentially promising nutraceuticals in medicine and industry.

    Abstract The present study centered around the prospect of bio fabricating nanoparticles using Moringa oleifera leaf and investigating their therapeutic potential with an assessment of the antioxidant status. Synthesis of stable ‘green’ nanoparticles of Moringa leaf extract is successfully reported with an extensive characterization. The average particle size and polydispersity index (PDI) of the nanoparticles obtained by Dynamic light scattering (DLS) analysis were 141.6 nm and a polydispersity index (PDI) of 0.32 respectively. The UV–visible spectroscopy (UV–vis) revealed a strong red absorbance peak at 664 nm,characteristic of chlorophyll. Electron micrographs confirmed that particles were in the nano range and were spherical with minimal agglomeration. The constituent functional groups were assessed by Fourier transformed infrared spectroscopy (FTIR). The X-ray diffractograms of reflected both the amorphous and crystalline domains.Furthermore, the phytochemical screening showed a higher phenolic/ flavonoid content of the Moringa nanoparticles (NMo) in comparison to the bulk Moringa leaf extract(Mo). These results corresponded to a potent antioxidant status of the NMo which was assessed by three complementary antioxidant assays. The therapeutic potential of the nanoparticles was evaluated based on in vitro anti-diabetic assays and cytotoxicity assay on human cancer cell lines (MCF-7 and HepG-2). The key findings suggest a more profound therapeutic efficacy of NMo than the Moringa leaf extract.

    Abstract Objective: Arsenic (As) has been in the forefront of toxicological research due to its continual worldwide human exposure and its adverse effects including genotoxicity. The study assessed the potential ameliorative effect of Indian mustard seeds (Brassica juncea) and their nanoparticles against arsenic toxicity. Methods: Male Wistar rats were used for the exposure study. Group I was control and group V was exposed only to the nanoparticles. Groups II, III, and IV were exposed to 10 mg/kg sodium arsenite for two weeks. Groups III and IV were treated with mustard seeds and the nanoparticles (500 mg/kg) respectively. Results: Arsenic intoxication caused a marked increase in the levels of As in the brain. Additionally, oxidative damage was exhibited with significantly enhanced levels of serum 8-OHdG and hepatic p53 and pS15. The pictographs and the analyzed parameters of the COMET assay illustrated DNA damage with significantly increased values of tail moment and length. Treatment with mustard seeds and its nanoparticles markedly reversed the adverse effects of As toxicity, which is attributed to the presence of a gamut of constituent phytocompounds in the mustard seeds. Conclusion: Taken together the nano formulation was more efficacious and could be introduced as a potential nutraceutical to combat metal toxicity.

    Abstract Coming into the second year of the pandemic, the acute respiratory syndrome coronavirus 2 (SARS-CoV-2) and its variants continue to be a serious health hazard globally. A surge in the omicron wave, despite the discovery of the vaccines, has shifted the attention of research towards the discovery and use of bioactive compounds, being potential inhibitors of the viral structural proteins. The present study aimed at the green synthesis of zinc oxide (ZnO) nanoparticles with seed extracts of Nigella sativa and Pimpinella anisum —loaded nanostructured oil carriers (NLC)—using a mixture of olive and black seed essential oils. The synthesized ZnO NLC were extensively characterized. In addition, the constituent compounds in ZnO NLC were investigated as a potential inhibitor for the SARS-CoV-2 main protease (3CLpro or Mpro) where 27 bioactive constituents, along with ZnO in the nanostructure, were subjected to molecular docking studies. The resultant high-score compounds were further validated by molecular dynamics simulation. The study optimized the compounds dithymoquinone, δ-hederin, oleuropein, and zinc oxide with high docking energy scores (ranging from −7.9 to −9.9 kcal/mol). The RMSD and RMSF data that ensued also mirrored these results for the stability of proteins and ligands. RMSD and RMSF data showed no conformational change in the protein during the MD simulation. Histograms of every simulation trajectory explained the ligand properties and ligand–protein contacts. Nevertheless, further experimental investigations and validation of the selected candidates are imperative to take forward the applicability of the nanostructure as a potent inhibitor of COVID-19 (Coronavirus Disease 2019) for clinical trials.

    Abstract The present study assessed the ameliorative potential of a commercial probiotic, Protexin® and its nanoformulation against cadmium (Cd) toxicity in Wistar rats. The ‘green’ synthesized nanoparticles had a size of 170 nm (PDI 0.527) with spherical and tubular bacillary shapes. Rats were exposed to 70 ppm of cadmium chloride hydrate for 35 days. Group I and V were negative and positive control respectively. Groups II, III, and IV were exposed to Cd. The Cd exposed groups, III and IV were treated with the probiotic and nanoprobiotic (1 ml containing 2 × 108cfu) respectively. Cd intoxication led to significant bioaccumulation of cadmium in liver and kidney. A significant increase in the serum malondialdehyde (MDA) and 8 -hydroxydeoxyguanosine (8-OHdG) levels was observed. The metallothionien (MT) and reduced glutathione (GSH) levels in the liver were significantly increased. The advance oxidation protein products (AOPP) and methylglyoxal (MG) levels were also elevated. Treatment with probiotic and its nanoparticles markedly reversed the Cd induced alterations in the parameters. The nanoformulation was more profound in reducing the (GSH) and MT levels in the liver and 8-OHdG levels in the serum. The key findings suggest that a nutritional intervention in the form of probiotic supplementation is a safe and efficacious remediation against heavy metal toxicity/oxidative stress.

    Abstract Breast cancer constitutes a major health problem for women worldwide. However, its incidence varies between populations and geographical locations. These variations could be diet-related, since there are several carcinogenic compounds in the modern diet, while natural products contain various anti-cancer elements. Several lines of evidence indicate that, in addition to their clear preventive effect, these compounds could also be used as therapeutic agents. In the present report we have shown that oleuropein, a pharmacologically safe natural product of olive leaf, has potent anti-breast cancer properties. Indeed, oleuropein exhibits specific cytotoxicity against breast cancer cells, with higher effect on the basal-like MDA-MB-231 cells than on the luminal MCF-7 cells. This effect is mediated through the induction of apoptosis via the mitochondrial pathway. Moreover, oleuropein inhibits cell proliferation by delaying the cell cycle at S phase and up-regulated the cyclin-dependent inhibitor p21. Furthermore, oleuropein inhibited the anti-apoptosis and pro-proliferation protein NF-κB and its main oncogenic target cyclin D1. This inhibition could explain the great effect of oleuropein on cell proliferation and cell death of breast cancer cells. Therefore, oleuropein warrants further investigations to prove its utility in preventing/treating breast cancer, especially the less-responsive basal-like type.

    Abstract The present study was undertaken to determine the bioaccumulation of cadmium in livers of tilapia fish ( Oreochromis niloticus ) and to assess the histological alteration of intestine, liver and kidney tissues due to cadmium toxicity. Fish ( Oreochromis niloticus ) were exposed to cadmium at different concentrations (5 and 10 mg L -1 ) for 7 days. Cadmium assayed by using AA220FS atomic absorption spectrophotometry. The concentration of cadmium in the liver tissues in the fish exposed to 10 mg L -1 was found to be (9.09±0.51 μg g -1 dry wt.) while it was 5.17±0.25 μg g -1 dry wt. in the fish exposed to 5 mg L -1 . There was a significant increase of cadmium chloride concentration in the liver tissues of the fish exposed to 10 mg L -1 compared to the those exposed to 5 mg L -1 . Histological alterations on liver tissues were in the form severe fatty vacuolations, generalised necrosis of hepatocytes, fatty change, congestion of liver sinusoids and central veins. Intestines showed severe congestion of submucosal blood vessels and sloughing of mucosal epithelium. Kidneys showed severe glumerular shrinkage and necrosis, lymphocytic infiltration in the distal renal convoluted tubules. Histopathological changes were more pronounced in fish exposed to 10 mg L -1 cadmium chloride.
