Abstract We conducted a comprehensive review of existing prediction models pertaining to the efficacy of immune-checkpoint inhibitor (ICI) and the occurrence of immune-related adverse events (irAEs). The predictive potential of neutrophil-to-lymphocyte ratio (NLR) and platelet-to-lymphocyte ratio (PLR) in determining ICI effectiveness has been extensively investigated, while limited research has been conducted on predicting irAEs. Furthermore, the combined model incorporating NLR and PLR, either with each other or in conjunction with additional markers such as carcinoembryonic antigen, exhibits superior predictive capabilities compared to individual markers alone. NLR and PLR are promising markers for clinical applications. Forthcoming models ought to incorporate established efficacious models and newly identified ones, thereby constituting a multifactor composite model. Furthermore, efforts should be made to explore effective clinical application approaches that enhance the predictive accuracy and efficiency.

Abstract Objective: To investigate the correlation of the pathological features of benign and malignant thyroid nodules with ultrasonic elastography parameters. Methods: A total of 176 patients with thyroid tumor who underwent thyroid tumor resection in this hospital between September 2016 and February 2018 were chosen as the research subjects, the 119 patients with thyroid cancer were selected as thyroid cancer group and the 57 patients with thyroid adenoma were selected as thyroid adenoma group. The thyroid cancer-related proliferation gene and thyroid cancer-related invasion gene expression as well as ultrasonic strain ratio in tumor tissues were compared between the two groups, and Pearson test was used to evaluate the correlation between above malignant molecule expression and ultrasonic strain ratio. Results: Thyroid cancer-related proliferation genes FOXA1, YAP and TPX2 mRNA expression in lesion tissues of thyroid cancer group were higher than those of thyroid adenoma group whereas EGCG and PDCD4 mRNA expression were lower than those of thyroid adenoma group; thyroid cancer-related invasion genes BRD4, Twist1, FoxM1 and SIRT1 expression were higher than those of thyroid adenoma group whereas Wortmannin and HIC1 mRNA expression were lower than those of thyroid adenoma group; ultrasonic strain ratio was higher than that of thyroid adenoma group. The Pearson test showed that the proliferation gene and invasion gene expression in thyroid cancer tissues were directly correlated with the ultrasonic strain ratio level. Conclusion: Thyroid cancer-related proliferation gene and invasion gene expression are directly correlated with ultrasonic strain ratio level, and ultrasonic elastography can be the reliable means to judge benign and malignant thyroid nodules.

Abstract Objective: To explore the correlation of TI-RADS grading with oncogene and invasion molecule expression in thyroid cancer lesions. Methods: Patients who were diagnosed with thyroid cancer in this hospital between January 2017 and January 2018 were divided into TIRADS grade 4 group (n=54) and TI-RADS grade 5 group (n=56) according to TI-RADS grading. 50 patients who underwent surgery and were diagnosed with thyroid adenoma in this hospital during the same period were chosen as benign thyroid group. The differences in the expression of oncogenes and invasion molecules in lesion tissues were compared among the three groups. Results: Proto-oncogenes c-myc, c-erbB-2, RET and TRK mRNA expression in lesions of TI-RADS grade 4 group and TI-RADS grade 5 group were higher than those of benign thyroid group whereas tumor suppressor genes DPC4, p53, PTEN and HBB mRNA expression were lower than those of benign thyroid group; invasion molecules BCORL1, BRD4, STAT3, CD151, SATB1 and CXCR4 mRNA expression were higher than those of benign thyroid group whereas ST7L mRNA expression were lower than that of thyroid benign group. With the increase of TI-RADS grading, the changes in the expression of oncogenes and invasion molecules were aggravated. Conclusion: The changes in oncogene and invasion molecule expression were aggravated with the increase of TI-RADS grading of thyroid cancer, and the specific TI-RADS grading was positively correlated with tumor malignancy.

Abstract Previous studies have recognized glaucoma as a neurodegenerative disease that causes extensive brain damage and is closely associated with cognitive function. In this study, we employed functional MRI to examine the intrinsic functional connectivity patterns of the default mode network (DMN) in patients diagnosed with primary angle-closure glaucoma (PACG), exploring its association with cognitive dysfunction. A total of 34 patients diagnosed with PACG and 34 healthy controls (HC), who were matched in terms of sex, age, and education, were included in the control group. The posterior cingulate cortex (PCC) was selected as the region of interest to examine functional connectivity alterations. Compared with the HC group, functional connectivity was attenuated in left anterior cingulum cortex and left paracentral lobule between with PCC in the PACG group, the results are statistically significant. Our study revealed that patients with PACG exhibit weakened functional connectivity within the DMN. This finding suggests the presence of a neurological mechanism that is associated with both visual dysfunction and cognitive impairments in PACG patients. Furthermore, our study provides neuroimaging evidence that can aid in the exploration of spontaneous neurological alterations and facilitate a deeper investigation of alterations in the visual conduction pathways of PACG patients.

Abstract Objectives: We sought to investigate the value of combining intrauterine cerebral blood flow changes with brain electrical activity examination in evaluating the prognosis of brain injury.
Methods: A total of 90 preterm infants were enrolled and divided into two groups: the brain damaged preterm infants group (n=55) and the non-brain damaged preterm infants group (n=35).The diagnostic efficacy of combining intrauterine cerebral blood flow changes with EEG activity examination in predicting the prognosis of preterm infants with brain injury was evaluated usingT-test. Pearson linear correlation was applied to analyze the relationship between fetal intrauterine cerebral blood flow changes combined with electrical activity examination and the prognosis of brain injury.
Results: Significant differences were seen in pulse index, S/D ratio, and other indexes between the two groups (P<0.05). The combined approach of intrauterine cerebral blood flow changes with EEG activity examination demonstrated significantly higher values for area under the curve (AUC), sensitivityand negative predictive value compared to using intrauterine cerebral blood flow changes or EEG activity examination alone (P<0.05). A positive correlation was found between fetal intrauterine cerebral blood flow and electrical activity examination (P<0.05).
Summary: Combining the assessment of intrauterine cerebral blood flow changes with cerebral electrical activity examination proved beneficial in diagnosing the prognosis of brain injury and provided an important reference for early clinical intervention.
